Role of long non-coding RNAs in therapeutic resistance and clinical applications in cancer.

Cancer is one of the leading causes of mortality worldwide and is recognized as a complex, multifactorial disease with no clearly defined etiology for its onset and progression. Long non-coding RNAs (lncRNAs) are widely distributed across the human body and play varied roles in regulating cellular processes. In recent years, they have gained the attention of the scientific community as key regulators of cancer due to their diverse functional roles and complex regulatory mechanisms. Aberrant expression of lncRNAs contributes to tumor progression, functioning as oncogenes that modulate various p

Wastewater surveillance reveals patterns of antibiotic resistance across the United States.

Antibiotic resistance is a growing public health threat, with over 2.8 million antibiotic-resistant infections and 35,000 attributable deaths annually in the U.S. Here, we sought to use wastewater monitoring to assess community-level burden of antibiotic resistance. This study quantifies concentrations of antibiotic resistance genes (ARGs) by digital droplet PCR in wastewater solids obtained from 163 wastewater treatment plants in 40 states in the United States. We measure 11 ARGs that confer resistance to beta-lactams (blaCMY, blaCTX-M, blaKPC, blaNDM, blaOXA-48, blaTEM, blaVIM), colistin (mc

A Case Series of Hypermucoid Klebsiella pneumoniae Liver Abscess in Caucasian Americans Without Travel History: An Epidemiologic Shift?

Hypervirulent Klebsiella pneumoniae (hvKp) is an emerging cause of community-acquired pyogenic liver abscess (PLA), classically described in East and Southeast Asia. We report two cases of hvKp-associated PLA in Caucasian women of American origin with no history of international travel, animal exposure, or Asian food consumption. Both patients presented with fever and right upper quadrant abdominal pain and were found to have solitary hepatic abscesses on imaging. Blood and abscess cultures confirmed K. pneumoniae with a positive string test (> 5 mm), K2 capsular serotype, and rmpA gene, estab

[Molecular mechanisms and control strategies for the potential transmission of bacterial antibiotic resistance along the food production chain].

The transmission potential of antimicrobial resistance (AMR) across the food chain constitutes a severe threat to global public health. Antibiotic overuse and misuse in intensive livestock farming drives the enrichment of antimicrobial resistance genes (ARGs) in animal-derived microbial communities, thereby enabling the formation of ARG dissemination networks that cover the entire food system from production to consumption. Molecular epidemiological evidence has validated the food chain as a pivotal bridge for the cross-species transmission of ARGs. Resistant bacteria and mobile genetic elemen

Exosome-like nanoparticles derived from Salvia miltiorrhiza promote diabetic wound healing by reducing oxidative stress and inflammation.

Chronic non-healing wounds remain a major cause of amputation and mortality for diabetic patients. Therefore, finding effective treatment options has become a significant issue that biomedical research needs to address. Nanovesicles that resemble exosomes participate in intercellular communication in plants and may transfer bioactive substances between animals and plants with implications for regenerative medicine. Salvia miltiorrhiza, a traditional medicinal plant, has been used for centuries to treat cardiovascular diseases, infections, inflammation, and wounds. However, little is known of e

Developmental, regenerative, and behavioral dynamics in acoel reproduction.

Acoel worms are an enigmatic and understudied animal lineage. Sparse descriptions suggest a diversity of reproductive anatomies across acoels, and likely a corresponding behavioral diversity. Here, we study the reproductive life history of the acoel Hofstenia miamia, an emerging lab-tractable model system. We describe H. miamia's reproductive organs, identifying structures previously unknown in acoels. Following worms from zygotes to adulthood, we find that their reproductive organs emerge in a stereotyped sequence as a function of increasing body size. These organs regenerate in a similar seq

Early observations on how children with and without autism take photographs: Preliminary implications for Augmentative and Alternative Communication design.

PURPOSE: Visual scene displays are contextual images designed to represent messages in augmentative and alternative communication systems. Currently, little is known about how children naturally compose photographs and select content. This study takes an exploratory approach to establish a preliminary baseline of how children take photographs to inform child-centred visual scene display design. The study aimed to examine the compositional features and thematic content of child-generated photographs, begin evaluating whether children with autism may differ from neurotypical peers in photographi

Evidence of predation events by marine mammals at offshore wind farms.

Anthropogenic structures in the marine environment alter the availability and distribution of food for marine animals with potential consequences for individual fitness, population and community dynamics. Knowledge of how marine mammals' behaviour or foraging activities are influence by existing wind farms is essential to predict the effect of future large-scale renewable energy development on local or regional populations. During acoustic tracking of demersal gadoid fish at two offshore wind farms in Scotland, six Atlantic cod were predated by a marine mammal close to or in the vicinity of tu

Cardiotoxicity adverse outcome pathway network: towards mechanistic and quantitative modelling.

INTRODUCTION: Chemical-induced heart toxicity remains a major challenge in drug development and environmental safety, largely because current testing often relies on narrow, late-stage endpoints that miss the complex biological progression of the toxicities. To address this, we developed a comprehensive Adverse Outcome Pathway (AOP) network that maps how early (bio) chemical triggers evolve into organ-level dysfunction. METHODS: By integrating data from the OECD AOP-Wiki, we constructed a unified network of 64 biological events and 94 documented relationships that identifies the critical biol

Empathy and Love as Protective Factors in Opioid Use Disorder Recovery.

Opioid use disorder (OUD) is a debilitating condition that impacts multiple domains of life, including social attachment systems, which makes recovery and relapse prevention challenging. The Brain Opioid Theory of Social Attachment suggests opioids hijack mechanisms involved in love and bonding. Previous studies show that social interaction can help reduce opioid use in both animals and humans, but little is known about whether prosocial emotions like love and empathy can protect against opioid use among people treated with medications for OUD. In this cross-sectional study, data were analyzed

A Multiagent Quantitative Microbial Risk Assessment of Raw Milk Cheeses: From Farm-Level Practices to Consumer Risk.

Ensuring the microbial safety of raw milk cheeses demands a robust, quantitative understanding of contamination pathways and risk mitigation strategies. In this study, we developed an innovative agent-based Quantitative Microbial Risk Assessment (QMRA) model that explicitly represents farms, animals, cheese batches, and consumers as individual agents. This approach enables a realistic simulation of microbial transmission throughout the entire production chain and facilitates the assessment of the effectiveness of control measures. Grounded in extensive knowledge of the French raw milk cheese s

A screen for adherens junction proteins regulating collective cell migration and testis morphogenesis reveals important roles for the Rab GAP RN-tre and the kinase Par-1.

Animal tissues have diverse architectures and cell behaviors across the epithelial-mesenchymal spectrum. Cell adhesion mediated by classical cadherins is foundational. Cadherins nucleate complexes of dozens of proteins connecting junctions to the cytoskeleton and signaling downstream. Many junctional proteins are well-studied in epithelia, but less is known about roles during mesenchymal migration. The nascent myotubes of the pupal Drosophila testis provide an excellent model for N-cadherin mediated mesenchymal migration. We combined a proximity proteomics dataset of adherens junction proteins

Bluetongue virus: current perspectives on characterisation and the need to formalise nomenclature in an era of rapidly evolving technology.

Bluetongue virus (BTV) is a complex, transboundary arboviral pathogen that circulates globally in ruminant populations. Bluetongue disease is a World Organisation for Animal Health listed disease affecting economically important farmed ruminants. BTV is a dsRNA virus from the Sedoreoviridae family that has a segmented genome. Classification of BTV traditionally focussed on antigenic characteristics. More recently, however, Sanger sequencing methods have been adopted for typing based on specific genome segment analysis and targeted whole-genome sequencing (WGS) is being more widely applied. Des

Conjunctival Sac-Derived Lactobacillus mucosae MM-1 Metabolite Indole-3-Lactic Acid Attenuates Lens-Induced Myopia in Mice via AHR Activation and TGF-β/Smad Inhibition.

Emerging research has revealed a diverse microbiota in the high myopia (HM) conjunctival sac, highlighting the therapeutic potential of modulating this community to influence disease outcomes. However, current research on targeted modulation of this microbiota for myopia intervention lacks in-depth investigation and definitive conclusions regarding its efficacy and underlying mechanisms. In this study, high-throughput sequencing and culturomics were first used to screen the conjunctival sac microbiota of healthy people, identifying Lactobacillus mucosae MM-1 as a potential probiotic for myopia
