Effects of alpha1-adrenoceptor antagonists on agonist and tilt-induced changes in blood pressure: relationships to uroselectivity.

We evaluated the uroselectivity of a series of alpha1-adrenoceptor antagonists by comparing their potency against phenylephrine-induced increases in urethral perfusion pressure and diastolic blood pressure in the anesthetized rabbit and pithed rat. In the rabbit, Rec 15/2739 (N-[3-[4-(2-methoxyphenyl)-1-piperazinyl]propyl]-3-methyl-4-oxo-2-phenyl -4H-1-benzopyran-8-carboxamide) as well as analogs with a chlorine substituent on the methoxyphenyl ring (Rec 15/2869) or this substituent combined with the replacement of the phenyl substituent on the pyran ring by cyclohexyl (Rec 15/3011) were 2-6-f

Potential therapeutic use of the selective dopamine D1 receptor agonist, A-86929: an acute study in parkinsonian levodopa-primed monkeys.

The clinical utility of dopamine (DA) D1 receptor agonists in the treatment of Parkinson's disease (PD) is still unclear. The therapeutic use of selective DA D1 receptor agonists such as SKF-82958 (6-chloro-7,8-dihydroxy-3-allyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzaze pine hydrobromide) and A-77636 ([1R, 3S] 3-[1'-admantyl]-1-aminomethyl-3,4-dihydro-5,6-dihydroxy-1H-2-benzo pyran hydrochloride) seems limited because of their duration of action, which is too short for SKF-82958 (< 1 hr) and too long for A-77636 (> 20 hr, leading to behavioral tolerance). We therefore conducted the present acut

An in situ-forming injectable silk hydrogel with a cocktail of chemotherapeutic drugs simultaneously targeting cancer cells and cancer stem cells.

Breast cancer recurrence and therapeutic resistance are primarily driven by breast cancer stem cells (bCSCs), which are poorly eliminated by conventional chemotherapy. To address this limitation, we developed an in situ-forming injectable silk hydrogel for sustained dual delivery of doxorubicin (DOX) and salinomycin (SAL), enabling concurrent targeting of bulk tumor cells and bCSCs. The hydrogel forms under physiological conditions without the need for UV irradiation or toxic initiators and exhibits tunable mechanical and degradation properties. An optimized 8% hydrogel exhibited controlled re

Co-delivery of TRAIL DNA and salinomycin for cancer therapy via enhanced apoptosis and cancer stemness inhibition.

Cancer treatment faces challenges including drug resistance to intrinsic apoptosis pathways and the persistence of cancer stem cells (CSCs). The tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) selectively induces extrinsic apoptosis in tumor cells by binding to death receptors while sparing normal cells. TRAIL-based gene therapy has emerged as a promising solution to address the short half-life limitation of recombinant TRAIL proteins. To further enhance the therapeutic efficacy, combination strategies with TRAIL sensitizer salinomycin (Sali) appear to be particularly effective

Cyclocurcumin potently inhibits human aromatase as a potential therapeutic agent.

Curcuminoids, including curcumin and its derivatives, show potent inhibition of aromatase (CYP19A1), crucial for estradiol synthesis and breast cancer metastasis. Our study evaluated the efficacy and mechanism of 10 curcuminoids and their metabolites against human and rat CYP19A1 using placental microsomes, revealing species-specific IC50 values. Cyclocurcumin (IC50, 4.43 μM) and curcumin (IC50, 3.49 μM) were the most effective inhibitors for human and rat CYP19A1, respectively. These compounds acted as mixed or competitive inhibitors, reducing estradiol production in human BeWo cells. Docking

Overcoming Resistance to Platinum-Based Drugs in Ovarian Cancer by Salinomycin and Its Derivatives-An In Vitro Study.

Polyether ionophore salinomycin (SAL) and its semi-synthetic derivatives are recognized as very promising anticancer drug candidates due to their activity against various types of cancer cells, including multidrug-resistant populations. Ovarian cancer is the deadliest among gynecologic malignancies, which is connected with the development of chemoresistant forms of the disease in over 70% of patients after initial treatment regimen. Thus, we decided to examine the anticancer properties of SAL and selected SAL derivatives against a series of drug-sensitive (A2780, SK-OV-3) and derived drug-resi

High anti-inflammatory activity of harpagoside-enriched extracts obtained from solvent-modified super- and subcritical carbon dioxide extractions of the roots of Harpagophytum procumbens.

Solvent-modified carbon dioxide extractions of the roots of Harpagophytum procumbens have been investigated with respect to extraction efficiency and content of harpagoside, and compared with a conventional extract. The effects of pressure, temperature, type and concentration of the modifier have been examined. Two extraction steps were necessary in order to achievehigh anti-inflammatory harpagoside-enriched extracts. The first extraction step was carried out in the supercritical state using carbon dioxide modified with n-propanol to remove undesired lipophilic substances. The main extraction

Neuroprotective effect of KR-31378 via KATP channel opening against ischemic insult.

The opening of the adenosine triphosphate (ATP)-sensitive potassium (KATP) channel has been proposed as a therapeutic approach for ischemia. Here we examined the opening effect of KR-31378 on the KATP channel using patch clamp recording in neuroblastoma 2a (N2a) cells and investigated the neuroprotective effect of KR-31378 in organotypic hippocampal slice cultures exposed to oxygen/glucose deprivation. The treatment with KR-31378 (10 microM) to N2a cells seemed to induce KATP channel opening in a dose dependent manner. The opening effect of KR-31378 was more significant than that of other know

Biological activity of Juglans regia leaf extract and in silico evaluation of its phytochemicals against Plasmodium berghei target proteins.

Parasitic infections remain a major global health challenge, with helminthiasis causing significant morbidity and economic losses. Limitations of synthetic anthelmintics, such as resistance and toxicity, have encouraged the search for natural plant-based alternatives. Juglans regia (walnut) leaves are rich in bioactive phytochemicals with potential antiparasitic effects. This study aimed to evaluate the anthelmintic efficacy of the methanolic leaf extract of J. regia (JRLE) against Eisenia fetida and to assess, through molecular docking, the potential interactions of its phytochemicals with se

Diterpene-phenylpropanoid hybrids and structurally diverse diterpenoid monomers from the endangered conifer Podocarpus annamiensis and their bioactivities.

In this phytochemical investigation, a total of 16 previously undescribed and 27 known diterpene-related compounds were isolated from the endangered Chinese endemic conifer Podocarpus annamiensis. Podoannamiacids A (1) and B (2) represent the first examples of a new chemical class of diterpene-phenylpropanoid hybrids characterized by a unique 7'-phenyl-7',8'-dihydrobenzofuran unit. This structure is hypothesized to be biogenetically derived through the formation of newly generated C-12-C-8' and C-13-O-C-7' bonds via a free radical coupling reaction between a totarane unit and a phenylpropanoid

Newborn screening for Pompe disease by measuring acid alpha-glucosidase activity using tandem mass spectrometry.

BACKGROUND: Pompe disease, caused by the deficiency of acid alpha-glucosidase (GAA), is a lysosomal storage disorder that manifests itself in its most severe form within the first months of life. Early detection by newborn screening is warranted, since prompt initiation of enzyme replacement therapy may improve morbidity and mortality. We evaluated a tandem mass spectrometry (MS/MS) method to measure GAA activity for newborn screening for Pompe disease. METHODS: We incubated 3.2-mm punches from dried blood spots (DBS) for 22 h with the substrate [7-benzoylamino-heptyl)-{2-[4-(3,4,5-trihydroxy

A novel cannabinoid peripheral cannabinoid receptor-selective inverse agonist blocks leukocyte recruitment in vivo.

The expression of the cannabinoid peripheral cannabinoid receptor (CB(2)) receptor on peripheral immune cells suggests that compounds specific for CB(2) might be effective anti-inflammatory agents. In this report, we present the initial biological profiling of a novel triaryl bis-sulfone, Sch.336 (N-[1(S)-[4-[[4-methoxy-2-[(4-methoxyphenyl)sulfonyl]phenyl]-sulfonyl]phenyl]ethyl]methanesulfonamide), which is selective for the human cannabinoid CB(2) receptor (hCB(2)). Sch.336 is an inverse agonist at hCB(2), as shown by its ability to decrease guanosine 5'-3-O-(thio)triphosphate (GTPgammaS) bin

Pharmacological inhibition of O-GlcNAcase reduces pS129-α-synuclein positive aggregates in the substantia nigra of mThy1-hSNCA mice.

BackgroundThe aggregation and spread of α-synuclein within brain are associated with the loss of dopaminergic neurons and the formation of Lewy bodies as seen in Parkinson's disease. Blocking the initiation of α-synuclein aggregation, or the spread of such aggregates, may offer disease-modifying approaches to slow disease progression. Previous studies have demonstrated that modification of aggregation prone proteins, including α-synuclein, with O-linked β-N-acetylglucosamine (O-GlcNAc) reduces their aggregation. Small molecule inhibitors of the enzyme O-GlcNAcase (OGA), which removes O-GlcNAc

Salinomycin inhibits porcine epidemic diarrhea virus infection by targeting Wnt/β-catenin pathway.

Porcine epidemic diarrhea virus (PEDV) is a re-emerging pathogen that causes severe economic losses in the pig industry. Commercial PEDV vaccines provide limited protection against PEDV virulent strains. Therefore, the development of novel vaccines and antiviral drugs is urgently required. In this study, we investigated the inhibitory effects of Salinomycin (SLM) against PEDV infection in vitro. First, the half-maximal cytotoxic concentration (CC50) and half-maximal inhibitory concentration (IC50) of SLM were measured by a cell counting kit 8 (CCK-8) and cytopathic effect (CPE).
