Cloning and characterization of an Eimeria necatrix gene encoding a gametocyte protein and associated with oocyst wall formation.

BACKGROUND: Gametocyte proteins of Eimeria (E.) spp. are important components of the oocyst wall and some have been used to develop transmission-blocking vaccines against avian coccidiosis. METHODS: Total RNA isolated from E. necatrix gametocytes was utilized as templates for RT-PCR amplification and sequencing of cDNA encoding a gametocyte protein using gene-specific primers. The cDNA was cloned into the bacterial expression vector pET28a(+) and expressed in E. coli BL21 cells. The antigenicity of the recombinant gametocyte protein and its localization in different E. necatrix life-cycle sta

Nanodiscs and electrospray ionization mass spectrometry: a tool for screening glycolipids against proteins.

Electrospray ionization-mass spectrometry (ESI-MS) is extensively employed to detect and quantify protein-carbohydrate interactions in vitro and is increasingly used to screen carbohydrate libraries against target proteins. However, current ESI-MS methods are limited to carbohydrate ligands that are relatively soluble in water and are, therefore, not generally suitable for studying protein interactions with glycolipids, an important class of cellular receptors. Here, we describe a catch-and-release (CaR)-ESI-MS assay, which exploits nanodiscs (NDs) to solubilize glycolipids and mimic their nat

HPV16 E7 protein and hTERT proteins defective for telomere maintenance cooperate to immortalize human keratinocytes.

Previous studies have shown that wild-type human telomerase reverse transcriptase (hTERT) protein can functionally replace the human papillomavirus type 16 (HPV-16) E6 protein, which cooperates with the viral E7 protein in the immortalization of primary keratinocytes. In the current study, we made the surprising finding that catalytically inactive hTERT (hTERT-D868A), elongation-defective hTERT (hTERT-HA), and telomere recruitment-defective hTERT (hTERT N+T) also cooperate with E7 in mediating bypass of the senescence blockade and effecting cell immortalization. This suggests that hTERT has ac

Quantitative profiling and identification of plasma proteins of spinocerebellar ataxia type 2 patients.

BACKGROUND: Spinocerebellar ataxia type 2 (SCA2) is an autosomal-dominant hereditary ataxia characterized by progressive gait and limb ataxia, dysarthria, slow saccades, neuropathy and dementia. The expansion of trinucleotide CAG repeats in the coding region of the ATXN-2 gene leads to expanded polyglutamine stretch in the mutated protein which causes neuronal death. OBJECTIVE: In this study, we investigated the blood plasma of SCA2 patients to find protein biomarkers. METHODS: Thirty-two ataxia patients clinically suspected for SCA2 were evaluated by the International Co-operative Ataxia Ra

Antibodies generated by immunization with the NS1 protein of West Nile virus confer partial protection against lethal Japanese encephalitis virus challenge.

Japanese encephalitis virus (JEV) and West Nile virus (WNV) are two medically important flaviviruses that can cause severe hemorrhagic and encephalitic diseases in humans. Immune responses directed against the NS1 protein of flaviviruses can confer protection against lethal viral challenge. Previous studies have shown that the WNV NS1 protein harbors epitopes that elicit antibodies that cross react with JEV. Here we demonstrate that the WNV NS1 protein not only contains cross-reactive epitopes, but that the antibodies elicited by these cross-reactive epitopes provide partial protection against

Expression of extracellular markers as criteria for treatment of peripheral arterial occlusive disease.

UNLABELLED: During the last decade, mesenchymal stromal cells (MSC) have been used in medical therapeutics, especially for reperfusion in peripheral arterial disease, ischemic cardiomiopathy and myocardial infarction. Studies demonstrated that membrane proteins are important in MSC endothelial differentiation. AIM: To determine the expression of extracellular markers of MSC, in order to ascertain if some expressed proteins (MSC specific cluster of differentiation proteins, essential for MSC characterization and proteins which have a role in adhesion and migration) have capability to induce an

F-box protein FBXL2 targets cyclin D2 for ubiquitination and degradation to inhibit leukemic cell proliferation.

Hematologic maligancies exhibit a growth advantage by up-regulation of components within the molecular apparatus involved in cell-cycle progression. The SCF (Skip-Cullin1-F-box protein) E3 ligase family provides homeostatic feedback control of cell division by mediating ubiquitination and degradation of cell-cycle proteins. By screening several previously undescribed E3 ligase components, we describe the behavior of a relatively new SCF subunit, termed FBXL2, that ubiquitinates and destabilizes cyclin D2 protein leading to G(0) phase arrest and apoptosis in leukemic and B-lymphoblastoid cell l

Complications of angioplasty and stenting of cervicocerebral arteries in Iran.

Learning and memory processes critically involve the orchestrated regulation of de novo protein synthesis. On the other hand it has become clear that regulated protein degradation also plays a major role in neuronal plasticity and learning behavior. One of the key pathways mediating protein degradation is proteosomal protein destruction. The anaphase-promoting complex/cyclosome (APC/C) is an E3 ubiquitin ligase that targets proteins for proteosomal degradation by the 26S proteasome. While the APC/C is essential for cell cycle progression it is also expressed in postmitotic neurons where it has

Proteomic analysis of rat skeletal muscle submitted to one bout of incremental exercise.

Exercise can alter gene transcriptional and protein translational rates leading to changes in protein abundance toward adaptation to exercise. We investigated the alterations in protein abundance in skeletal muscle after one bout of an exhaustive exercise through proteomic analysis. Gastrocnemius muscles were sampled from non-exercised control rats and from rats exercised on a treadmill with incremental increases in speed until exhaustion (approximately 30 min). Rats were sacrificed 3 and 24 h after exercise cessation. Two-dimensional gel electrophoresis was performed and spots with a signific

[Progress on lactococcus lactis expressing heterologous antigens as live mucosal vaccines].

Lactic acid bacteria (LAB) are natural commensals of the gastro-intestinal tracts of humans and most animals. They are considered to be safe bacteria with a GRAS (generally regarded as safe) status. The studies on LAB as live vehicles for expression of heterologous proteins or antigens have gained great progress in the past decades. Lactococcus lactis, an important species of LAB, possesses many properties that make it an ideal candidate for expressing and delivering heterologous proteins. With the significant advances in the genetic study of LAB, a series of constitutive or inducible gene exp

Abnormal expression of neurofilament proteins in dysmyelinating axons located in the central nervous system of jimpy mutant mice.

Myelination in the peripheral nervous system is considered to increase the phosphorylation level of neurofilament proteins in the axon, resulting in an increase in axonal calibre. To understand the relationship between myelination and neurofilament proteins in axons, we examined jimpy mutant mice with a point mutation in the proteolipid protein gene and dysmyelination in the central nervous system. The jimpy mice exhibited a characteristic similarity in neurofilament nature to the myelin-deficient mice in the peripheral nervous system reported previously. The following novel results were obtai

Two Gq class G proteins are expressed in human keratinocytes.

G proteins link many cell surface receptor generated signals to activation of multiple cellular processes in all tissues. There is specificity in the receptor interaction with the G protein and in the interaction of the specific G protein with different effector molecules. The purpose of this study was to determine some of the biologically relevant G proteins in keratinocytes. The G alpha subunit of the heterotrimeric G protein was investigated because much of the biologic activity and the receptor specificity resides there. A polymerase chain reaction strategy was used that amplified multiple

Isolation and partial characterization of a 110-kD dimer actin-binding protein.

Two Triton-insoluble fractions were isolated from Acanthamoeba castellanii. The major non-membrane proteins in both fractions were actin (30-40%), myosin II (4-9%), myosin I (1-5%), and a 55-kD polypeptide (10%). The 55-kD polypeptide did not react with antibodies against tubulins from turkey brain, paramecium, or yeast. All of these proteins were much more concentrated in the Triton-insoluble fractions than in the whole homogenate or soluble supernatant. The 55-kD polypeptide was extracted with 0.3 M NaCl, fractionated by ammonium sulfate, and purified to near homogeneity by DEAE-cellulose an

The alleviation of oxidative damage in celiac disease by black soybean peptides and their mechanism.

BACKGROUND: Celiac disease (CD) is a chronic autoimmune condition that occurs in genetically predisposed individuals when they consume gluten. The α-gliadin peptide exacerbates the CD pathological process by inducing oxidative stress, intestinal cell damage, and apoptosis. Addressing oxidative stress in CD may offer a valuable strategy for supplementary treatment. RESULTS: Black soybean peptides (BSPs) have been shown to inhibit oxidative stress and inflammation, although the specific mechanisms are not fully understood. This study investigated the effects of BSPs on Caco-2 cytotoxicity induc

The Emerging Role of CACNA1E in Cancer: Molecular Mechanisms and Therapeutic Implications.

Voltage-gated calcium channels (VGCCs) are crucial membrane proteins that mediate calcium influx. The CACNA gene family, which encodes the alpha subunits of VGCC complexes, is essential for forming functional calcium channels and plays significant roles in various cancers. This review focuses on the CACNA1E gene, which encodes the Cav2.3 channel α1E subunit, and systematically elaborates its role in cancer. Studies have identified CACNA1E as a key prognostic stemness-related gene in bladder cancer. Its somatic mutations are associated with the development of air pollution-related lung cancer a

MtvS1 and MtvS2 Interact with RNA polymerase to regulate the Francisella type V-A CRISPR-Cas system.

Bacteria and archaea often harbor multiple CRISPR-Cas loci to defend against mobile genetic elements. Little is known, however, about whether and how different CRISPR-Cas systems are differentially regulated, in many instances due to the impossibility of studying CRISPR immunity in native hosts. Here we investigated the regulation of the endogenous type II-B and type V-A CRISPR-Cas systems present in the opportunistic human pathogen Francisella novicida U112. We found that while the type II-B system is constitutively expressed, the type V-A system is differentially expressed at stationary phas

The SL-I structural element of the 3' UTR region of West Nile virus participates in the regulation of viral translation.

The genome of West Nile virus (WNV) is a positive, ssRNA molecule, which encodes a single ORF flanked by UTRs (5' and 3' UTRs). These UTRs are enriched with structural RNA elements that play critical roles in the viral cycle, including translation, replication and encapsidation. The 3' UTR is crucial for translation control by the combined enhancer and repressor activities of different structural elements. This manuscript provides new roles for the 3' UTR SL-I element as a translation inhibitor in WNV in cis. This inhibitory effect is primarily observed in a cap-dependent translation context,

Increased hydrogen and ethanol production in transformants of the filamentous cyanobacterium Phormidium lacuna.

Research on biofuel production by cyanobacteria has largely focused on unicellular species. To date, there are no reports on engineered filamentous cyanobacteria for bioethanol or biohydrogen production. However, biofilm formation in filamentous cyanobacteria could provide an advantage for biotechnological applications. We used natural transformation and homologous recombination to establish recombinant expression at the psaD locus of the filamentous cyanobacterium Phormidium lacuna. A similar approach has already been successful in the unicellular cyanobacterium Synechocystis sp. PCC 6803 (Ap

A natural depsipeptide antibiotic binds the E-site of the bacterial ribosome.

A key challenge in addressing the antibiotic resistance crisis is identifying new antimicrobial compounds1. Although natural products produced by fungi and bacteria, particularly actinomycetes, have been the source of most antibiotics discovered over the past 80 years, they have fallen out of favour owing to the frequent rediscovery of known drug scaffolds2. The current perception is that antibiotic-producing actinomycetes have been over-mined and possess little novelty left to yield. Here we demonstrate that by using improved fractionation approaches that enrich previously overlooked minor pr

The deubiquitinase CYLD inhibits thrombin-induced p38 MAPK-p65 NF-κB signaling and inflammatory cytokine production to suppress triple-negative breast cancer progression.

Triple-negative breast cancer (TNBC) is an aggressive subtype characterized by chronic inflammation with limited therapeutic options. Although thrombin activation of protease-activated receptor-1 (PAR1) promotes ubiquitin-dependent signaling and inflammatory responses in endothelial cells, its contribution to inflammatory effects in TNBC remain poorly understood. Here, we show that loss of the deubiquitinase cylindromatosis (CYLD) markedly increases basal phosphorylation of p38 mitogen-activated protein kinase and p65 nuclear factor kappa B subunit to levels comparable to thrombin stimulation,
