Unveiling the gut-vascular connection: Gut microbiota regulates cardiovascular toxicity induced by combined exposure to PM2.5 and high-fat diet via butyric acid/NLRP3 pathway.

Environmental particulate matter and dietary factors have been found to induce gut microbiota dysbiosis and exert an impact on cardiovascular toxicity. However, the underlying biological connections among them remain unclear. Our results indicated co-exposure to PM2.5 and high-fat diet (HFD) notably elevated blood pressure and blood lipid in mice and serum nitric oxide and endothelin-1 were abnormal. Meanwhile, vascular diameter and end-diastolic velocity (EDV) decreased, while resistance index (RI) increased. There was a synergistic effect of co-exposure on EDV and RI. Furthermore, significan…

Air pollution modulates brown adipose tissue function through epigenetic regulation by HDAC9 and KDM2B.

Recent experimental and epidemiologic data have strongly associated air pollution in the pathogenesis of insulin resistance and type 2 diabetes mellitus. We explored the effect of inhalational exposure to concentrated ambient particulate matter smaller than 2.5 μm (PM2.5), or filtered air, using a whole-body inhalation system (6 hours/day, 5 days/week) for 24 weeks on metabolism and brown adipose tissue (BAT) function. Mechanistic evaluation of insulin resistance, glucose uptake with 18F-fluorodeoxyglucose positron emission tomography, alongside evaluation for differentially methylated regions…

Low-dose tire wear chemical 6PPD-Q exposure elicit fatty liver via promoting fatty acid biosynthesis in ICR mice.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPD-Q) as a major metabolite of tire wear chemical 6PPD has been demonstrated to be an emerging burden of exposure in human populations, via contamination from drinking water, air particulate matter and food sources. Whilst increasing attention has been moved toward its adverse effect, the potential hepatotoxicity of 6PPD-Q in mammals at realistic dose remains unknown. Here, the toxic effects of 6PPD-Q at environmentally relevant dose on the liver of adult mice and its underlying mechanism were investigated through an integrative app…

Identification and Assessment of lncRNAs and mRNAs in PM2.5-Induced Hepatic Steatosis.

Research indicates that fine particulate matter (PM2.5) exposure is associated with the onset of non-alcoholic fatty liver disease (NAFLD), the most prevalent chronic liver disorder. However, the underlying pathogenesis mechanisms remain to be fully understood. Our study investigated the hub long non-coding RNAs (lncRNAs) and messenger RNAs (mRNAs) associated with hepatic steatosis caused by PM2.5 exposure and their pathological mechanisms. The analysis of gene profiles in the GSE186900 dataset from the Gene Expression Omnibus (GEO) enabled the identification of 38 differentially expressed lnc…

The Rodent Electronic Nicotine Delivery System: Apparatus for Voluntary Nose-Only E-Cigarette Aerosol Inhalation.

Tobacco use is the leading cause of death globally and in the U.S. After decades of decline, driven by decreases in combusted tobacco use, n … inistration. We confirmed that RENDS sequesters nicotine aerosol in the nose port by measuring fine particulate matter (PM < 2.5 microns) generated by e-cigarettes. We also showed that rats robustly self-administer flavored nicotine aerosol, resulting in high blood levels of cotinine (the major nicotine metabolite) and spontaneous somatic withdrawal symptoms. Thus, …

Social Inequalities in Exposure to Air Pollution in the EPIC Cohorts of Turin and Varese.

In Europe, evidence on the relationship between socioeconomic position (SEP) and air pollution exposure is mixed. We assessed the associatio … ssion models. Nitrogen dioxide (NO2) and nitrogen oxide (NOx) data were available for both cohorts; particulate matter (PM2.5, PM10) only for Turin. Linear regression models (adjusted for sex, age, and marital status) estimated associations between SEP and annual mean pollutant concentrations (µg/m3), stratified by cohort. In Varese, lower education was associated w …

Impact of lifestyle and environmental factors on fertility.

PURPOSE OF REVIEW: Infertility affects approximately 15% of couples, with male factors implicated in more than 50% of cases. Concerns over d … nnabis, and opioids - may also suppress fertility. Environmental exposures such as heat waves, fine particulate matter, and endocrine-disrupting chemicals reduce semen quality and can contribute to testicular dysgenesis. Conversely, adherence to certain diets, antioxidant supplementation modestly improves sperm quality and reduce SDF. SUMMARY: Lifestyle and environ …

PM2.5 exposure deteriorates Th1/Th2 balance in pediatric asthma by downregulating ALKBH5 and enhancing SRSF1 m6A methylation.

Accumulating evidence has shown that long-term exposure to particulate matter with aerodynamic diameter of less than 2.5 μm (PM2.5) causes Th1/Th2 imbalance and increases the risk of allergic asthma (AA) in children. However, the mechanism underlying such effect remains elusive. Here, an AA mouse model was developed by intranasal administration of ovalbumin (OVA) and uncovered that OVA-sensitized mice exhibited pathological damage of lung tissues, mucus production, augmented serum IgE levels, enhanced Th2 cells and associated cytokine levels, and diminished Th1 cells and associated cytokine le…

Ambient Air Pollution and Parkinson's Disease and Alzheimer's Disease: An Updated Meta-Analysis.

BACKGROUND: Previous epidemiological evidence regarding the associations between ambient air pollution and two major neurodegenerative diseases, Alzheimer's disease (AD) and Parkinson's disease (PD), remains inconclusive. OBJECTIVE: This study aimed to evaluate the associations between long-term and short-term exposure to PM2.5 and PM10 (i.e., particulate matter with an aerodynamic diameter of, or smaller than, 2.5 μm or 10 μm), nitrogen dioxide (NO2), ozone, sulfur dioxide, and carbon monoxide and the risks of AD and PD. METHODS: A random-effects model was used to summarize individual effec…

Short-Term Relationship Between Air Pollution and Mortality from Respiratory and Cardiovascular Diseases in China, 2008-2020.

Most existing epidemiological studies on the impact of air pollution on noncommunicable diseases have focused on urban areas, rather than nationwide studies that include rural areas. This study utilized a time-stratified case-crossover study that included deaths registered in the National Mortality Surveillance System from 2008 to 2020. Atmospheric particulate matter (PM10 and PM2.5), nitrogen dioxide (NO2), sulfur dioxide (SO2), and carbon monoxide (CO) were evaluated via the National Nested Air Quality Prediction Modeling System. Conditional logistic regression was used to assess the associa…

Impact of ozone on kidney transplant outcomes.

Earlier work has shown adverse effects of air pollution on kidney disease outcomes, yet the specific impact of ozone (O3) on kidney transplant (KT) recipients remains unclear. We investigated the effects of long-term O3 exposure on KT outcomes by employing a multipollutant model. This multicenter, longitudinal cohort study included 4796 adult KT recipients who survived ≥1 year after transplantation from 2002-2020 at 3 major hospitals in South Korea. O3 and fine particulate matter (PM2.5) exposures were estimated using residential zip code-based high-resolution machine learning models. Associat…

Xanthan gum and tamarind seed polysaccharide crosslinked metformin film: Potential against pollution-induced conjunctivitis and diabetes-induced elevated vitreous vascular endothelial growth factor.

Metformin (MET) is known to modulate pathways that are also activated by particulate matter 2.5 (PM2.5) for induction of conjunctivitis. It also reduces vascular endothelial growth factor (VEGF) expression, a significant reason for diabetic retinopathy. Despite the prophylactic effects of oral MET, topical use is necessary to achieve the higher levels needed for therapeutic efficacy. So, tamarind seed polysaccharide (TSP) was crosslinked with xanthan gum (XG) to provide a high polar surface area to load higher MET (5 mg/6 mm). Citric acid (CA) further crosslinked the matrix, improving the mech…

YTHDC2 mediated RNA m6A modification contributes to PM2.5-induced hepatic steatosis.

Exposure to fine particulate matter (PM2.5) is a significant risk factor for hepatic steatosis. The N6-methyladenosine (m6A) is implicated in metabolic disturbances triggered by exogenous environmental factors. However, the role of m6A in mediating PM2.5-induced hepatic steatosis remains unclear. Herein, male C57BL/6J mice were subjected to PM2.5 exposure throughout the entire heating season utilizing a real-ambient PM2.5 whole-body inhalation exposure system. Concurrently, HepG2 cell models exposed to PM2.5 were developed to delve the role of m6A methylation modification. Following PM2.5 expo…
