[Atypical achondrogenesis. Review of the literature apropos of 2 cases].

The authors describe two cases of a minor form of achondrogenesis. This form has not been published previously. A discussion of the different possibilities of differential diagnosis follows.

The oto-palato-digital syndrome.

A boy aged ten years with oto-palato-digital syndrome is discussed. Because of severe conductive hearing loss tympanotomy was performed and abnormal poorly mobile ossicles were found. Stapedectomy was performed without improvement of hearing.

Unilateral dysplasia of the superior articular facet of the axis.

A case of unilateral dysplasia of the superior articular facet of the axis due to incomplete or delayed ossification is described in a child. This may result in asymmetry of the articular surfaces of the axis in adult life and be a cause of torticollis. To our knowledge, no similar case has been reported.

Simultaneous correction of pelvic obliquity, frontal plane, and sagittal plane deformities in neuromuscular scoliosis using a unit rod with segmental sublaminar wires: a preliminary report.

Ten patients with neuromuscular scoliosis and pelvic obliquity had segmental spinal instrumentation using a unit Luque rod with sublaminar wires and fixation into the pelvis. Nine of the 10 patients also had anterior spinal fusion without instrumentation before the posterior procedure. Average preoperative pelvic obliquity was 42 degrees which was corrected to 6 degrees (82% correction). Average preoperative scoliosis was 92 degrees, which was corrected to 16 degrees (81% correction). Complications included a wound hematoma in one patient and a superficial wound dehiscence in another. There ha

Dynamic myeloid suppressor states in cancer and inflammation and their therapeutic potential.

PURPOSE OF REVIEW: Myeloid-derived suppressor cells (MDSCs) are central mediators of immune dysregulation in cancer, infection, and inflammatory diseases. This review highlights recent advances redefining MDSCs' biology, with a focus on their developmental origin, functional plasticity, and the signaling and metabolic networks that regulate their expansion and activity within the bone marrow and peripheral tissues. RECENT FINDINGS: Single-cell and lineage-tracing studies demonstrate that MDSCs do not represent a discrete lineage but instead arise as dynamic functional states across the myeloi

Exploring the validity and consistency of antero-posterior skeletal dysplasia indicators: a comprehensive cephalometric study.

BACKGROUND: The aim of this study was to check the reliability of different antero-posterior skeletal discrepancy indicators and to compare and correlate various craniofacial measurements taken from cephalometric radiographs, to check the reliability of the linear and angular geometric measurements in clinical diagnosis. METHODS: A sample of 60 pretreatment lateral cephalographs were taken. Eighteen cephalometric landmarks were marked and considered for the cephalometric measurements; 5 reference planes were considered; 9 linear measurements and 11 angular measurements were taken into conside

Clinical and Oral Manifestations in a Patient with Lenz-Majewski Syndrome: A Rare Case Report.

BACKGROUND: Lenz-Majewski Syndrome (LMS) is a rare multisystem genetic disorder characterized by intellectual disability, cutis laxa, craniofacial abnormalities, and distinct dental features. It is caused by gain-of-function mutations in the Phosphatidylserine Synthase 1 (PTDSS1) gene, which plays a key role in phospholipid biosynthesis. Among the craniofacial and dental manifestations, a progeroid facial appearance with prognathism, midface hypoplasia, large auricles, enamel dysplasia, delayed tooth eruption, and malocclusion are commonly observed. Due to the rarity and phenotypic variability

Disease-related PSS1 mutant impedes the formation and function of osteoclasts.

Phosphatidylserine (PS) is an acidic phospholipid that is involved in various cellular events. Heterologous dominant mutations have been identified in the gene encoding PS synthase 1 (PSS1) in patients with a congenital disease called Lenz-Majewski syndrome (LMS). Patients with LMS show various symptoms, including craniofacial/distal-limb bone dysplasia and progressive hyperostosis. The LMS-causing gain-of-function mutants of PSS1 (PSS1LMS) have been shown to synthesize PS without control, but why the uncontrolled synthesis would lead to LMS is unknown. Here we investigated the effect of PSS1L

Bone dysplasia in Hutchinson-Gilford progeria syndrome is associated with dysregulated differentiation and function of bone cell populations.

Hutchinson-Gilford progeria syndrome (HGPS) is a premature aging disorder affecting tissues of mesenchymal origin. Most individuals with HGPS harbor a de novo c.1824C > T (p.G608G) mutation in the gene encoding lamin A (LMNA), which activates a cryptic splice donor site resulting in production of the toxic "progerin" protein. Clinical manifestations include growth deficiency, lipodystrophy, sclerotic dermis, cardiovascular defects, and bone dysplasia. Here we utilized the LmnaG609G knock-in (KI) mouse model of HGPS to further define mechanisms of bone loss associated with normal and premature

Clinical and Radiographic Assessment of Adult Function After Blount Disease in Childhood: An Exercise in Futility.

BACKGROUND: Few studies evaluate long-term function of patients with Blount disease. We sought to document patient radiographic and functional status 20 to 30 years after sugical treatment for infantile or adolescent Blount disease. METHODS: We reviewed the medical records and radiographs of patients operated at our institution for Blount disease between 1985 and 1995. Over a 4-year period, we recruited subjects for an IRB-approved call-back study. RESULTS: One hundred five patients were eligible for the call-back study. Thirty-one (30%) had a criminal record, 18 of sufficient gravity to pre

Recurrence in infantile tibia vara (Blount disease) after high tibia and fibula osteotomy.

Infantile Tibia Vara (ITV or Blount disease) often recurs after surgical correction of the deformity. Vastly different recurrence rates have been reported in the literature. The main objective of this study was to identify any risk factors for the recurrence of ITV following high tibia and fibula valgus osteotomy. This was a retrospective cohort comparative study of 44 patients with ITV (16 with bilateral involvement) who underwent a total of 60 high tibia and fibula osteotomies. Demographics, family history, BMI, walking age and age at surgery were recorded. Pre-, post- and follow-up weight-b
