The effects of temperature on ferritin transport by endothelial cell vesicles in the capillaries of the frog mesentery.

Using electron microscopy we have determined the appearance of ferritin in the endothelial cell vesicles of frog mesenteric capillaries after varying periods of exposure of the luminal surface of the cell to a ferritin solution, at 17-20 degrees C and at 4-7 degrees C. At 17-20 degrees C, the pattern of labelling observed in the absence of flow through the capillary is very similar to that previously described by us in continuously perfused capillaries [1]. Some vesicles are labelled after the shortest exposures (less than 1 sec) and labelling increases with exposure to reach steady values at …

[Characteristics of the ultrastructural organization of endothelial cells in cardiac and omental blood capillaries based on cryofractographic data].

Using cryofractography, endotheliocytes of heart and omentum blood capillaries were studied, different patterns of distribution of membranous particles being observed of the "P" and "E" faces of the fracture plans of endothelial cell cytomembranes. Three types of distribution of micropinocytotic vesicles can be distinguished: the linear, hexagonal, and the clusteral type. The number of micropinocytotic vesicles was sharply reduced near the region of intercellular contacts. On the fracture plan, attached pinocytotic vesicles look like "craters" on the "E" face, and like circumvallatue papillae …

Improved Detection of Capillaries in High-Resolution Handheld Vital Microscopy by Use of the MicroTools Advanced Computer Vision Algorithm.

OBJECTIVE: Handheld vital microscopy (HVM) enables bedside visualization of the microcirculation, with major improvements from sidestream darkfield (SDF) to incident dark field (IDF) imaging. Although IDF offers high-resolution images, standard analysis methods require down-sampling to match SDF's lower field of view (FOV) and pixel density. MicroTools, an automated stabilization and analysis algorithm, has previously been validated for use with SDF image sequences. This study aimed to assess the accuracy and precision of microcirculatory analysis using MicroTools on full-frame, high-resolutio

Distinct changes in nailfold capillaries are associated with the clinical and serological features of patients with systemic lupus erythematosus.

BACKGROUND: The role of nailfold videocapillaroscopy (NVC) in systemic lupus erythematosus (SLE) remains undefined. Prognostic tools are needed for SLE as the phenotypes differ in prognosis and treatment. Recent studies have reported a high prevalence of capillary abnormalities in SLE. This study investigated whether distinct NVC patterns are associated with distinct SLE phenotypes. METHODS: We did NVC and comprehensive clinical characterisation in 56 patients with SLE. Correlations within NVC findings were tested using Spearman's rho. Subsets of patients with similar NVC changes were identif

Asymmetric dimethylarginine and vascular injury in rheumatoid arthritis, from capillaries to large vessels: results from a cross-sectional study.

INTRODUCTION: Rheumatoid arthritis (RA) is associated with elevated cardiovascular risk. Asymmetric dimethylarginine (ADMA), an endogenous nitric oxide synthase inhibitor and marker of endothelial dysfunction, has been implicated in inflammation-driven vascular injury. However, the association between ADMA and micro- and macrovascular function in RA remains unclear. METHODS: Patients with RA underwent nailfold videocapillaroscopy (NVC) to assess microvascular alterations in the dermal capillary network. Macrovascular function was evaluated with well-known markers of arterial stiffness [caroti

UVA causes dysfunction of ETBR and BMPR2 in vascular endothelial cells, resulting in structural abnormalities of the skin capillaries.

BACKGROUND: Capillary structural abnormalities cause skin disorders. Mottled redness, i.e., skin redness unevenness, may appear on the sun-e … 2, involved in the morphogenesis and maintenance of the blood vessels, and reduced migration of the capillaries. CONCLUSION: UV rays alter ETBR and BMPR2 expression in the skin capillaries, and cause partial dilation and decreased migration, resulting in capillary structural abnormalities and causing skin redness unevenness.

Three-dimensional optoacoustic imaging of nailfold capillaries in systemic sclerosis and its potential for disease differentiation using deep learning.

The autoimmune disease systemic sclerosis (SSc) causes microvascular changes that can be easily observed cutaneously at the finger nailfold. Optoacoustic imaging (OAI), a combination of optical and ultrasound imaging, specifically raster-scanning optoacoustic mesoscopy (RSOM), offers a non-invasive high-resolution 3D visualization of capillaries allowing for a better view of microvascular changes and an extraction of volumetric measures. In this study, nailfold capillaries of patients with SSc and healthy controls are imaged and compared with each other for the first time using OAI. The nailfo…

Skeletal muscle satellite cells are located at a closer proximity to capillaries in healthy young compared with older men.

BACKGROUND: Skeletal muscle satellite cells (SC) are instrumental in maintenance of muscle fibres, the adaptive responses to exercise, and there is an age-related decline in SC. A spatial relationship exists between SC and muscle fibre capillaries. In the present study, we aimed to investigate whether chronologic age has an impact on the spatial relationship between SC and muscle fibre capillaries. Secondly, we determined whether this spatial relationship changes in response to a single session of resistance exercise. METHODS: Muscle biopsies were obtained from the vastus lateralis of previou…

Coelectroosmotic capillary electrophoresis of phenolic acids and derivatized amino acids using N,N-dimethylacrylamide-ethylpyrrolidine methacrylate physically coated capillaries.

Two different families of compounds, i.e., phenolic and amino acids have been separated by capillary electrophoresis using a physically adsorbed polymer as capillary coating. The polymer used was N,N-dimethylacrylamide-ethylpyrrolidine methacrylate (DMA-EpyM) and it provided an stable coating by only flushing the capillary with a DMA-EpyM aqueous solution for 2min between runs. The usefulness of this procedure has been demonstrated through the fast analysis of different families of solutes. Two different detection systems, diode-array detector and laser-induced fluorescence, have been used to

[Relation between changes in the microcirculation in the capillaries supplying the toenails and the degree of chronic venous insufficiency].

Chronic venous insufficiency (CVI) is characterised by stasis which may lead to an imbalance in the cutaneous microcirculation resulting in a wide spectrum of complications. The use of capillaroscopy on the toenail folds of patients with CVI allows the morphological and microhemorrheological aspects of microcirculatory disorders to be studied. The aim of this study was to clarify the correlation between the extent of abnormalities of the nutritional capillary and the degree of severity of CVI according to Widmer's classification. Capillaroscopy of the toenail fold was used to study 100 patient
