Combination targeted therapy with two biologic/targeted synthetic DMARDs in 1200 patients with immune mediated inflammatory diseases. A systematic literature review for current landscape in safety and efficacy.

BACKGROUND: Immune-mediated inflammatory diseases (IMID) include rheumatoid arthritis (RA), psoriatic arthritis (PsA), axial spondyloarthritis (axSpA), Crohn's disease (CD), ulcerative colitis (UC), and psoriasis (PsO). While biologic (b) and targeted synthetic (ts) DMARDs are effective, nearly 60 % of patients fail to achieve low disease activity status. Combination targeted therapy (CTT) using concomitantly two different b- or ts-DMARDs has been explored, but results on safety and efficacy are unclear. OBJECTIVE: To systematically review the literature on CTT in IMID. METHODS: Following th

IBD Matchmaking: Rational Combination Therapy.

A growing number of patients with Crohn's disease and ulcerative colitis have disease that is refractory to multiple advanced therapies, have undergone multiple surgeries, and require further treatment options. For this reason, there has been increasing use of multiple simultaneous advanced targeted therapies. Although the knowledge on combined advanced targeted therapy (CATT) in inflammatory bowel disease (IBD) has been largely limited to observational data and early-phase randomized controlled trials, combination of therapies is commonplace in many other diseases. This review discusses conce

Genetics and therapy for pediatric eye diseases.

Ocular morphogenesis in vertebrates is a highly organized process, orchestrated largely by intrinsic genetic programs that exhibit stringent spatiotemporal control. Alternations in these genetic instructions can lead to hereditary or nonhereditary congenital disorders, a major cause of childhood visual impairment, and contribute to common late-onset blinding diseases. Currently, limited treatment options exist for clinical phenotypes involving eye development. This review summarizes recent advances in our understanding of early-onset ocular disorders and highlights genetic complexities in deve

[New therapeutic strategies in non-Hodgkin lymphomas and Hodgkin lymphoma].

In this review, we report the main advances of the last years in the four most common lymphomas in France, namely Hodgkin lymphoma, large cell diffuse B lymphoma, follicular lymphoma and mantle cell lymphoma. We have identified consensual practices in first line in France and then distinguished the targeting by new molecules. Thus, we wanted to highlight the problems for each of these four lymphomas and understand the tools used to find solutions. Finally, this review makes it possible to understand to what extent the new molecules (targeted therapies, immunotherapy) make it possible to contin

Targeted therapies for renal cell carcinoma.

The management of patients with metastatic renal cell carcinoma (RCC) has changed dramatically over the past few years. Nephrectomy remains an important intervention for localized RCC but systemic therapy is the mainstay of treatment for patients who relapse after surgery or who have metastatic RCC. Before 2005, medical therapies for RCC were limited to cytokine therapies, which are very toxic and benefit only a small percentage of patients. In 2017, therapeutic agents now include kinase and immune checkpoint inhibitors. Contemporary research with these agents is now focusing on combinatorial

2017 Clinical trials update in new treatments of β-thalassemia.

The underlying basis of β-thalassemia pathology is the diminished β-globin synthesis leading to α-globin accumulation and premature apoptotic destruction of erythroblasts, causing oxidative stress-induced ineffective erythropoiesis, bone marrow hyperplasia, splenomegaly, and increased intestinal iron absorption with progressive iron overload. Better understanding of the molecular mechanisms underlying this disease led to the recognition of new targets with potential therapeutic utility. Agents such as JAK2 inhibitors and TGF-β ligand traps that reduce the ineffective erythropoiesis process are

Androgen receptor as a therapeutic target in endometrial cancer: a narrative review.

Endometrial cancer continues to be the most common gynecologic malignancy in the United States. Endometrial tumors frequently express hormonal receptors, making this pathway targeting an attractive anti-cancer treatment. Recent advances have broadened our understanding of the androgen receptor's role in solid tumors beyond prostate cancer. Understanding the significance of androgen receptor signaling and its effects on tumor behavior and therapeutic outcomes in endometrial cancer is important for further clinical development. This review presents inter-connected pathways involving sex hormone-

CLDN18.2 in Gastric Cancer: Current Therapeutic Landscape and Future Perspectives.

Claudin 18.2 (CLDN18.2), a tight junction protein selectively expressed in normal gastric epithelium and widely retained during carcinogenesis, has emerged as a promising therapeutic target for advanced gastric cancer (AGC). SPOTLIGHT and GLOW trials evaluated the anti-CLDN18.2 monoclonal antibody (mAb) zolbetuximab in combination with first-line chemotherapy, and established CLDN18.2 as a therapeutic target, initiating a paradigm shift toward a biomarker-driven treatment approach in AGC. In addition, from zolbetuximab to a diverse pipeline of promising high-affinity mAbs, bispecific antibodie

Targeting claudins in cancer.

Epithelial-derived malignancies account for the majority of human tumours and present considerable treatment challenges owing to their heterogeneity, metastatic potential and resistance to therapy. The claudin family of tetra-transmembrane proteins was identified approximately 28 years ago as containing key regulators of epithelial function. These proteins are integral components of tight junctions, which control barrier integrity, selective channel permeability and cellular organization in epithelial tissues. Subsequent murine studies revealed that claudins also have tissue-specific physiolog

The NF-κB Signalling Pathway: Mechanisms, Consequences and Therapeutic Targets.

Nuclear Factor-kappa B (NF-κB) is a master transcriptional regulator orchestrating critical cellular processes, predominantly immunity and inflammation. However, its aberrant constitutive activation has emerged as a unifying pathogenetic hallmark across diverse malignancies, autoimmune disorders, and chronic inflammatory diseases. While the fundamental biology of canonical and non-canonical NF-κB signalling is well-established, translating this extensive knowledge into clinically viable therapeutics remains severely hindered by dose-limiting systemic toxicities and complex pharmacokinetic liab

Investigating metabotropic glutamate receptors as molecular targets for next-generation pharmacotherapies for alcohol use disorder.

INTRODUCTION: Alcohol use disorder (AUD) is a major public health concern with few available pharmacotherapy options. There remains a critical need to identify novel molecular targets for more effective treatments. Evidence suggests that AUD is associated with dysregulated glutamatergic neurotransmission, including a hyperglutamatergic state that contributes to the development and maintenance of alcohol seeking behaviors. AREAS COVERED: Recent advances in characterizing neuronal, glial, and synaptic changes underlying AUD pathophysiology have identified the metabotropic glutamate receptor 2 (

Matching hallmarks of cancer complexity with N-of-1 precision oncology.

Recent hallmarks of cancer iterations1 posit that durable control requires mechanism-guided co-targeting of hallmark capabilities rather than single-pathway inhibition. Translating this systems-level precision oncology framework at scale remains challenging. Sicklick et al.2 show that N-of-1, molecularly matched multi-drug regimens are feasible without traditional phase 1 trials; survival scales linearly with matching score, not number of drugs or dose.

[Recent Advances in the Treatment of HER2-Altered Solid Tumors].

Between 2024 and early 2026, HER2-directed oncology has rapidly evolved from a strategy confined to HER2-overexpressing malignancies to a histology-agnostic framework that accommodates diverse HER2 alterations, including amplification/overexpression, low-ultralow protein expression, and activating ERBB2 mutations. Antibody-drug conjugates (ADCs), led by trastuzumab deruxtecan, have established clinically meaningful activity across multiple solid tumors in HER2 IHC 3+ populations and have further expanded into HER2-low and ultralow expression settings, supporting the concept of HER2 as a biolog
