Activity and Safety Optimization of Mesoricin: A Dual-Domain Antifungal Peptide from Mesorhizobium sp.

Cryptococcus neoformans infections pose a significant global health threat. This study introduces mesoricin, a novel dual-domain antimicrobial peptide (AMP) scaffold derived from Mesorhizobium sp. identified using an in silico quantitative antifungal activity index (AFI). The peptide structure comprises an α-helix domain, which disrupts microbial membranes but exhibits highly hemolytic activity, and a β-sheet domain, which targets intracellular energy metabolism and resilient pathways. Rational design through α-helix domain removal and AFI-guided mutations yielded a mesoricin variant with enha

Inhibitory effects of reumycin produced by Streptomyces sp. TPMA0082 on virulence factors of Pseudomonas aeruginosa.

Pseudomonas aeruginosa is an opportunistic human pathogen that causes a wide range of infections. The increasing multidrug-resistance of P. aeruginosa poses a critical challenge for medical care. P. aeruginosa employs virulence factors and biofilms to establish infections in humans and protect itself from environmental stress or antibiotics. These factors are regulated by a quorum sensing mechanism involving multiple regulatory systems that act interdependently through signaling molecules. Therefore, interference with quorum sensing systems can suppress the pathogenicity of P. aeruginosa. In t

Detection and Characterization of Colistin Heteroresistance in an Enterobacter cloacae ST48 Strain Isolated From Raw Milk in Algeria.

An Enterobacter cloacae ST48 isolate exhibiting a colistin-heteroresistant phenotype was obtained from raw cow milk in Algeria. Two subpopulations were detected, both carrying the blaTEM-1 gene. While one subpopulation (E. cloacae TS34-a) was highly susceptible to colistin (MIC 0.06 µg/mL), the other (E. cloacae TS34-b) was resistant (MIC 8 µg/mL). Population analysis profiling (PAP) confirmed heteroresistance. PCR amplification revealed the absence of mcr genes in both subpopulations. However, multiple non-synonymous mutations, including a stop-codon mutation in phoP gene, were found in the r

An assessment of the potential impact of eugenol on multidrug-resistant pathogens.

Multi-drug resistance is the ability of some microorganisms, such as bacteria and viruses, to tolerate the effects of numerous medications. This phenomenon decreases the efficacy of current therapies, which causes longer illnesses and higher mortality rates. As a result, it presents an enormous threat to the healthcare sector. Research and countermeasures are of vital importance to address this global health problem, as the development of multi-drug resistance is frequently associated with inappropriate use of antibiotics or prolonged use. Our objective is to find the medicine that effectively

Novel cationic peptide OB1111 is a dual anti-planktonic and anti-biofilm agent against P. aeruginosa strains PA14 and PAO1.

BACKGROUND: Pseudomonas aeruginosa is a Gram-negative pathogen frequently responsible for nosocomial infections and a significant problem in intensive care units. P. aeruginosa, as an opportunistic pathogen, increases mortality risks for severely wounded and immunocompromised individuals. The inherent drug-resistance of P. aeruginosa now requires novel therapeutics with multiple mechanisms that will offer lasting potency in the post-antibiotic era. Synthetic antimicrobial peptides (AMPs) are ideal, as their multiple modes of action slow resistance development. In this study, we investigated th

Nanoparticles as New Antifungals in the Prevention of Bovine Mycotic Mastitis Caused by Candida spp. and Diutina spp.-In Vitro Studies.

Bacterial infections are the primary cause of mastitis in dairy cattle. Fungal mastitis occurs in 1-12% of cases. Antibiotic therapy, the standard treatment for mastitis, has led to antibiotic-resistant bacteria, reducing treatment efficacy and increasing fungal mastitis occurrence. Antibiotics lack biocidal effects on fungi, which often exhibit resistance to antifungal agents. This study evaluated the antifungal properties of nanoparticles (NPs) against Candida albicans, Candida glabrata, Candida parapsilosis, Diutina rugosa var. rugosa, Diutina catenulata, and Diutina rugosa. Tested NPs incl
