Decision-Making Signatures of Methamphetamine and Alcohol Use Disorders.

BACKGROUND: Aberrant decision making is a hallmark of substance use disorders (SUDs), often impeding recovery. While uncertainty, which comprises risk and ambiguity, is central to real-world choices, its distinct effects in SUDs remain underexplored. In this study, we disentangle risk and ambiguity to identify context-specific impairments in methamphetamine use disorder (MUD) and alcohol use disorder (AUD). METHODS: We used a Choice under Risk and Ambiguity task to examine uncertainty decision making (UDM) in 101 individuals with MUD, 56 individuals with AUD, and their respective healthy cont

Expression of GDAP1 Gene Correlates With Alcohol Deprivation Effect.

Alcohol addiction is a widespread disease, and the exact causes and consequences are still not fully determined. Neurotransmitters and neuronal circuits are not only the target structure of alcohol and responsible for its direct effects but also play a central role in the development of addiction. A gene that has been linked to alcohol use disorder in recent studies is the ganglioside-induced differentiation-associated protein 1 (GDAP1) gene. The present study focuses on the hippocampus, a brain region particularly vulnerable to alcohol and rich in Gdap1 gene expression. Using an established d

A neural substrate of comorbid depressive symptoms in alcohol use.

Alcohol use disorder (AUD) is commonly associated with depression, which may exacerbate the clinical outcome and increase treatment difficulty. The neural substrates underlying such comorbid symptoms are unclear. Here, we identify the regulatory role of amygdala Menin (multiple endocrine neoplasia type 1) signaling in orchestrating local GABAergic inhibition, which modulates the emotional state following alcohol use. Alcohol intake reduces Menin expression in the amygdala, decreasing GAT1 transcription. Restoring Menin signaling or overexpressing GAT1 in the amygdala could suppress alcohol pre

Alcoholization as an Alternative Treatment for Prostatic Cyst and Abscess in Dogs.

Current therapeutic approaches for prostatic cysts and abscesses involve surgical methods like reduction and omentalization. This retrospective study evaluates the effectiveness and safety of ultrasound-guided percutaneous drainage combined with alcoholization as a minimally invasive treatment for prostatic cysts and abscesses in dogs. Forty-three male dogs with a single prostatic cavity (1-6 cm) were enrolled and categorized into two groups: group A (twenty-two dogs with prostatic abscesses treated via alcoholization) and group B (twenty-one dogs with BPH-associated cysts). Group B was furthe

An enzyme-free alcohol-based organoid harvesting solution.

Three-dimensional (3D) cell culture is a more physiologically relevant model for drug development than two-dimensional (2D) cell culture. A common method to culture cells in 3D consists in embedding cells in synthetic or animal-based matrices that provide structural support for cell growth. They partially mimic in vivo conditions and enable scalable culture. Here, we introduce an alcohol-based Solution for Harvesting Organoids Efficiently, denoted SHOE. We tested its harvesting potential on 2 cell lines grown as spheroids and 2 patient-derived organoids. It enables rapid, high-yield cell recov

Sex differences in the impact of trauma on alcohol self-administration.

Challenging encounters can lead to escalated alcohol consumption, as evidenced by individuals grappling with posttraumatic stress disorder (PTSD). However, the precise influence of prior stress experiences, as opposed to acute stressors, on alcohol intake remains incompletely understood. This study sought to simulate the enduring repercussions of trauma using a stress paradigm involving foot shocks presented in an odor-enriched environment. The presence of a scent throughout contextual fear conditioning is more likely to cause stress effects to generalize across various environments, and this

Female Cardioprotection in a Mouse Model of Alcohol-Associated Cardiomyopathy.

Chronic alcohol misuse is the leading cause of non-ischemic dilated cardiomyopathy, and the molecular mechanisms underlying the development of alcohol-associated cardiomyopathy (ACM), particularly regarding sex-specific susceptibility and mitochondrial contributions, are not fully known. In this study, we utilized a preclinical model of chronic + binge ethanol consumption to investigate sex differences in disease severity and mitochondrial function. Male and female C57BL/6J mice were fed ethanol or control liquid diets for 30 days, with 2 binge episodes on days 10 and 30. Cardiac morphology wa

Argument for the pharmacokinetically-informed preclinical researcher: A commentary for Alcohol.

The purpose of this commentary is to advocate for the importance of carefully considering ethanol pharmacokinetics when conducting preclinical studies involving alcohol consumption. Researchers working in this field commonly use alcohol drinking as a principle or ancillary measurement. However, the amount of alcohol consumed cannot substitute for an understanding of dose, which is an essential referent in any study involving pharmacology, especially in studies using 24-h, two-bottle choice access to alcohol. Dose is the critical variable for understanding potency and efficacy, for translating

Alcohol-Related Liver Disease: Novel Insights into Mechanism.

Alcohol-related liver injury is complex and remains poorly understood. Research to date has pinpointed the role of oxidative stress, hepatic inflammation, and derangements in lipid metabolism as drivers of disease pathogenesis. More recent research has identified alterations in the gut microbiome, epigenetics, G protein-coupled receptors, and microRNA's as possible drivers of disease pathogenesis and may serve as potential new treatment targets.

Metabolic dysfunction and alcohol-associated liver disease (MetALD).

Metabolic dysfunction and alcohol-associated liver disease (MetALD) is a recently implemented nomenclature and disease terminology for patients with metabolic dysfunction-associated steatotic liver disease, who consume greater amounts of alcohol. MetALD is diagnosed in individuals who have at least one metabolic risk factor (such as obesity, type 2 diabetes mellitus, hypertension, etc) and consume 140-350 g/week of alcohol for women or 210-420 g/week for men. Conversely, alcohol-associated liver disease is diagnosed in individuals who consume >350 g/week of alcohol for women and >420 g/week fo

Mechanisms of alcohol influence on fear conditioning: A computational model.

BACKGROUND: A connection between stress-related illnesses and alcohol use disorders is extensively documented. Fear conditioning is a standard procedure used to study stress learning and links it to the activation of amygdala circuitry. However, the connection between the changes in amygdala circuitry and function induced by alcohol and fear conditioning is not well established. METHODS: We introduce a computational model to test the mechanistic relationship between amygdala functional and circuit adaptations during fear conditioning and the impact of acute vs. repeated alcohol exposure. Evaluation of ethylglucuronide and cocaethylene as hair markers of alcohol consumption.

BACKGROUND AND AIMS: The concurrent use of alcohol and cocaine is known to be a harmful condition in terms of behaviour and organ damage. The identification of this condition requires the use of objective biomarkers, such as hair ethylglucuronide (hEtG) and hair cocaethylene (hCE). The aim of the present work was to investigate their presence and their possible mutual relationship in a group of cocaine users to evaluate the different potentials offered by the two analytes in the retrospective investigation of alcohol intake. MATERIALS AND METHODS: 209 hair specimens, previously collected from

Defects of Cortical Microcircuits Following Early Exposure to Alcohol.

The interplay between excitatory pyramidal neurons and GABAergic interneurons is the basic building block of neocortical microcircuits and plays a critical role in carrying out higher cognitive functions. Cortical circuits are deeply and permanently disrupted by exposure to alcohol during brain development, the main non-genetic cause of intellectual disability. Here, I review experimental studies of fetal alcohol spectrum disorders, dealing with permanent cellular and molecular alterations of neocortical neurons and their connections.

The role of DNA methylation in alcohol-mediated neurodevelopmental toxicity.

Alcohol induces neurodevelopmental toxicity through multiple biological processes, including DNA methylation and histone modifications in epigenetic regulation. Epigenetic mechanisms involving DNA chemical modifications represent crucial molecular pathways that regulate gene expression during neurodevelopment, exhibiting high sensitivity to adverse lifestyle factors such as alcohol consumption, smoking, and stress. Prenatal alcohol consumption is a primary cause of fetal neurodevelopmental disorders. Alcohol alters DNA methylation and histone modification levels in the brain, which in turn dis
