Exome sequencing identifies a novel variant in ACTC1 associated with familial atrial septal defect.

BACKGROUND: The genetics of congenital heart disease (CHD) remain incompletely understood. Exome sequencing has been successfully used to id … novel rare variants causing CHD without the limitations of a candidate gene approach. When mutation prediction algorithms are not helpful, studies of familial disease can help distinguish rare pathologic mutations from benign variants. Consideration of the family history can lead to genetic insights into CHD.

Regulation of endoderm formation and left-right asymmetry by miR-92 during early zebrafish development.

microRNAs (miRNAs) are a family of 21-23 nucleotide endogenous non-coding RNAs that post-transcriptionally regulate gene expression in a seq … which led to abnormal Kupffer's vesicle development and left-right patterning defects. Using target prediction algorithms and reporter constructs, we show that gata5 is a target of miR-92. Alteration of gata5 levels reciprocally mirrored the effects of gain and loss of function of miR-92. Moreover, genetic epistasis experiments showed that miR-92-mediated defects could …

Methylation inhibitor therapy in the treatment of myelodysplastic syndrome.

The class of DNA methyltransferase inhibitors is represented by azacitidine and decitabine. Azacitidine is approved for the treatment of patients in both low- and high-risk subtypes of myelodysplastic syndrome (MDS), and decitabine is currently under review by the FDA. Azacitidine phase III trial data, based upon the Cancer and Leukemia Group B (CALGB) study 9221, showed durable clinical and symptomatic improvement in bone marrow function, a reduction in the risk of leukemic transformation, and significant improvements in the quality of life of patients treated with azacitidine compared with s

Patient-recognition data-mining model for BCG-plus interferon immunotherapy bladder cancer treatment.

Bladder cancer is the fifth most common malignant disease in the United States with an annual incidence of around 63,210 new cases and 13,180 deaths. The cost for providing care for patients with bladder cancer disease is high. Bladder cancer treatment options such as immunotherapy, chemotherapy, radiation therapy, transurethral resection, and cystectomy, are used with varying success rates. In this research, data from a nationwide bacillus Calmette-Gue rin (BCG) plus interferon-alpha (IFN-alpha) immunotherapy clinical trial was considered. Data mining algorithms were used to analyze the effec

Perceived pitch of synthesized voice with alternate cycles.

Both in normal speech voice and in some types of pathological voice, adjacent vocal cycles may alternate in amplitude or period, or both. When this occurs, the determination of voice fundamental frequency (defined as number of vocal cycles per second) becomes difficult. The present study attempts to address this issue by investigating how human listeners perceive the pitch of alternate cycles. As stimuli, vowels /a/ and /i/ were synthesized with fundamental frequencies at 140 Hz and 220 Hz, and the effect of alternate cycles was simulated with both amplitude- and frequency-modulation of the gl

Determination of fluorescent probe orientations on biomolecules by conformational searching: algorithm testing and applications to the atomic model of myosin.

The ability of a localized conformational searching method to predict probe orientation was tested on model nucleic acid and protein structures and applied to the prediction of skeletal myosin integrity upon chemical modification of its reactive thiols. Double-stranded oligonucleotides were chemically labeled with donor and acceptor resonance energy transfer probes at each end for distance determinations. These measurements were made independently using a terbium chelate as a donor to each of four chemically and spectroscopically distinct acceptor probes from the xanthene and cyanine dye group

Insertion mutagenesis as a tool to predict the secondary structure of a muscarinic receptor domain determining specificity of G-protein coupling.

The N-terminal segment of the third intracellular loop (i3) of muscarinic acetylcholine receptors and other G protein-coupled receptors has been shown to largely determine the G-protein coupling selectivity displayed by a given receptor subtype. Based on secondary-structure prediction algorithms, we have tested the hypothesis that this region adopts an alpha-helical secondary structure. Using the rat m3 muscarinic receptor as a model system, a series of five mutant receptors, m3(+1A) to m3(+5A) were created in which one to five additional alanine residues were inserted between the end of the f…

A novel method for biological age assessment utilizing hospitalization risk of cardiovascular disease.

BACKGROUND: Aging is a complex process involving physiological changes that gradually impair function in organisms. Chronological age (CA) does not fully account for variability in the aging process, whereas biological age (BA) better reflects disease risk. This study introduces Cardiovascular Disease Probability-based Biological Age (CDPBA), a novel BA algorithm. METHODS: We analyzed clinical data from 11.7 million individuals aged ≥ 40 years in the National Health Insurance Service database. Using a Cox proportional hazards model, clinical biomarkers, lifestyle factors, fine particulate mat

Application of Machine Learning to Osteoporosis and Osteopenia Screening Using Hand Radiographs.

PURPOSE: Fragility fractures associated with osteoporosis and osteopenia are a common cause of morbidity and mortality. Current methods of diagnosing low bone mineral density require specialized dual x-ray absorptiometry (DXA) scans. Plain hand radiographs may have utility as an alternative screening tool, although optimal diagnostic radiographic parameters are unknown, and measurement is prone to human error. The aim of the present study was to develop and validate an artificial intelligence algorithm to screen for osteoporosis and osteopenia using standard hand radiographs. METHODS: A cohor

Predicting the Risk of Driving Under the Influence of Alcohol Using EEG-Based Machine Learning.

Driving under the influence of alcohol (DUIA) is closely associated with alcohol use disorder (AUD). Our previous study on machine learning (ML) algorithms revealed a very high accuracy of decision trees with neuropsychological features in predicting the risk of DUIA despite limited data availability. Thus, this study aimed at comparing six well-known ML algorithms based on electroencephalographic (EEG) signals to differentiate adults with AUD and DUIA (AUD-DD) from those with AUD without DUIA (AUD-NDD) and controls. Fifteen AUD-DD and 10 AUD-NDD participants were recruited from a single terti

Implementation of a Cardiovascular Implantable Electronic Device Heart Failure Prediction Tool-Guided Management Pathway.

Cardiovascular implantable electronic devices (CIEDs) monitor physiologic variables that could identify subacute heart failure (HF) decompensation and impending HF hospitalization. One such algorithm uses measurements from the previous 30 days of CIED remote monitoring data to predict low-, medium-, or high-probability of HF hospitalization in the next 30 days. We sought to understand how to prospectively implement the use of such algorithms in routine HF care. From January 18, 2024 to April 19, 2024, HF risk categories were predicted from scheduled remote transmissions every 30 days and from

Using machine learning models to predict HBeAg seroconversion in CHB patients receiving pegylated interferon-α monotherapy.

BACKGROUND AND OBJECTIVE: Though there are many advantages of pegylated interferon-α (PegIFN-α) treatment to chronic hepatitis B (CHB) patients, the response rate of PegIFN-α is only 30 ~ 40%. Therefore, it is important to explore predictors at baseline and establish models to improve the response rate of PegIFN-α. METHODS: We randomly divided 260 HBeAg-positive CHB patients who were not previously treated and received PegIFN-α monotherapy (180 μg/week) into a training dataset (70%) and testing dataset (30%). The intersect features were extracted from 50 routine laboratory variables using the

Simulated four-dimensional CT for markerless tumor tracking using a deep learning network with multi-task learning.

INTRODUCTION: Our markerless tumor tracking algorithm requires 4DCT data to train models. 4DCT cannot be used for markerless tracking for respiratory-gated treatment due to inaccuracies and a high radiation dose. We developed a deep neural network (DNN) to generate 4DCT from 3DCT data. METHODS: We used 2420 thoracic 4DCT datasets from 436 patients to train a DNN, designed to export 9 deformation vector fields (each field representing one-ninth of the respiratory cycle) from each CT dataset based on a 3D convolutional autoencoder with shortcut connections using deformable image registration.
