Severe enduring anorexia nervosa with significant height increase in adulthood: A case report.

BACKGROUND: Anorexia nervosa (AN) is a severe psychiatric disorder characterized by restrictive eating, low body weight, and distorted body image, and is often complicated by endocrine dysfunction. While hypogonadism and delayed puberty are well documented in female patients, reports in males remain scarce because of the lower prevalence of AN in this group. Hypogonadism may result in impaired pubertal development, growth arrest, and reduced testicular volume in male patients. Although weight restoration can sometimes enable catch-up growth when AN begins before puberty, the long-term potentia

A novel mechanism in regulating drug sensitivity, growth, and apoptosis of bortezomib-resistant multiple myeloma cells: the USP4/KLF2/HMGA2 cascade.

BackgroundMultiple myeloma (MM) is a malignant disorder originating from plasma cells. Bortezomib (BTZ) resistance has become a huge obstacle to MM treatment. Herein, we elucidated the action of Kruppel-like factor 2 (KLF2), a crucial transcription factor (TF), on BTZ resistance of MM.MethodsTwo BTZ-resistant cell lines (MM1.S/BTZ and NCI-H929/BTZ) were generated and used. KLF2 mRNA was quantified by quantitative PCR, and protein expression was analyzed by immunoblotting. MTT cell cytotoxicity assay was used to test BTZ sensitivity. Cell growth was detected by MTT and EdU assays. Flow cytometr

Placental mesenchymal dysplasia: a rare case associated with second trimester fetal growth restriction.

BACKGROUND: Placental mesenchymal dysplasia (PMD) is a rare, benign, placental disorder characterised macroscopically by an enlarged multi-cystic placenta. It is a condition associated with a range of reported clinical outcomes and can be misdiagnosed as a molar or partial molar pregnancy given the similarities in clinical presentation. We present an unusual case of PMD complicated by fetal growth restriction and oligohydramnios in the second trimester. CASE PRESENTATION: A 33 year old female was referred to our fetal medicine unit with a multi-cystic placenta at dating scan suspected initial

A heterozygous microdeletion of 20p12.2-3 encompassing PROKR2 and BMP2 in a patient with congenital hypopituitarism and growth hormone deficiency.

Congenital growth hormone deficiency is a rare disorder with an incidence of approximately 1 in 4,000 live births. Pituitary development is under the control of a multitude of spatiotemporally regulated signaling molecules and transcription factors. Mutations in the genes encoding these molecules can result in hypopituitarism but for the majority of children with congenital hypopituitarism, the aetiology of their disease remains unknown. The proband is a 5-year-old girl who presented with neonatal hypoglycaemia and prolonged jaundice. No definitive endocrine cause of hypoglycaemia was identifi

Childhood glaucoma in association with congenital disorder of glycosylation caused by mutations in fucosyltransferase 8.

A rare form of congenital disorder of glycosylation (CDG) was recently discovered in individuals with biallelic mutations in fucosyltransferase 8 (FUT8). The clinical characteristics of patients with FUT8-CDG include intrauterine growth retardation, feeding difficulties, hypotonia, microcephaly, seizures, short stature, developmental delay, and respiratory abnormalities. We report the first case of glaucoma in an infant with FUT8-CGD and hypothesize a pathogenesis for glaucoma.

Epiphora and proptosis as a presenting complaint in acromegaly: Report of two cases with review of literature.

Acromegaly is an endocrine disorder characterized by the excess of growth hormone leading to bony and soft-tissue enlargement. Eye involvement due to extraocular muscle enlargement has been described rarely in these patients. Epiphora has been described only once earlier in a patient with acromegaly. Here, we describe two females with acromegaly who presented with the complaints of watering from the eye and proptosis.

[Utility of fetal ultrasonography in the prenatal diagnosis of Smith-Lemli-Opitz syndrome].

Smith-Lemli-Opitz syndrome is a rare autosomal recessive genetic disorder which diagnosis is usually made postnatally. We describe a case of a prenatal diagnosis based only on specific ultrasound findings: intra-uterine growth retardation with facial dysmorphia, polydactyly and genital anomalies. We suggest giving more consideration to the ultrasound scanning for the diagnosis of the syndrome in the prenatal period.

Endocrine manifestations of histiocytosis.

Diabetes insipidus and growth retardation are the prominent endocrine manifestations of histiocytosis. Galactorrhea, hypogonadism, and panhypopituitarism are rarely associated with this disorder. To date, the vast majority of morphologic and functional studies of the pituitary and hypothalamus in affected patients indicate that histiocytic infiltration results in a hypothalamic dysfunction with a secondary partial or complete hypopituitarism. This hypopituitarism is due to deficient trophic stimulation or inhibition by hypothalamic releasing or inhibitory factors.

Colorectal cancer in a man with silver-Russell syndrome: a case report.

Silver-Russell syndrome (SRS), also known as Russell-Silver syndrome (RSS), is a congenital growth disorder characterized by intrauterine and postnatal growth retardation, craniofacial disproportion, asymmetry, and other distinctive features. It was first described by Alex Russell and Henry Silver in the 50s of the 20th century. Its broad range of manifestations makes its true incidence difficult to determine. While gastrointestinal anomalies such as gastroesophageal reflux disease and esophagitis have been reported in SRS patients, an association with colorectal cancer (CRC) has not been desc

Congenital Alopecia, Hypoplastic Kidneys, Growth Restriction, Growth Hormone Resistance, and Liver Fibrosis: Confirmation of a New Syndrome Caused by Biallelic Variants in ZPR1.

We report two female siblings, a 13-month-old and a newborn, with multiple anomalies including hypoplastic kidneys, severe growth restriction, facial dysmorphism, and alopecia, both found to be homozygous for the c.587 T>C variant in ZPR1. Their clinical features are strikingly similar to those previously reported in a patient who was homozygous for the same variant. Our report confirms that homozygosity for c.587 T>C in ZPR1 underlies a novel genetic syndrome with autosomal recessive inheritance and that c.587 T>C is a founder variant for ZPR1 disorder in the Middle Rio Grande Valley. Inhibition of KDM6B prevents osteoarthritis by blocking growth plate-like H3K27me3 loss in bivalent genes.

Osteoarthritis (OA) is the most prevalent joint disorder occurring with articular cartilage degradation. It includes a switch from an articular to a growth plate chondrocyte phenotype. Here, we investigated the histone modification profiles and found significant H3K27me3 loss in OA, which led to disease-associated gene expression. Surprisingly, these genes were occupied by both H3K27me3 and H3K4me3 in normal chondrocytes, showing a poised bivalent state. Furthermore, we observed the derepression of similar bivalent genes in growth plate chondrocytes. Finally, a KDM6B inhibitor GSK-J4 prevented

[Pharmacological therapies for height improvement in pubertal children with short stature].

Short stature in puberty significantly affects the physical and mental health of adolescents. The continuous acceleration of skeletal maturation, caused by sex hormones during puberty, limits the time available for growth and poses a considerable challenge for the treatment of short stature. To date, there is still no standardized treatment protocol for this disorder. However, puberty is the last period to improve the final adult height. Currently, commonly used pharmacological treatments in clinical settings include recombinant human growth hormone, gonadotropin-releasing hormone analogs, and

Hashimoto Thyroiditis in a Patient With Dubowitz Syndrome and Short Stature.

Dubowitz syndrome (DS) is a rare genetic disorder characterized by multiple morphological abnormalities, short stature, and different degrees of mental disability. Endocrinological evaluation should be done for these subjects, as they can suffer from multiple hormonal derangements. We present a case of a 12-year-old Lebanese girl, diagnosed with Dubowitz syndrome, who presented to our clinic for short stature. She had received growth hormones (GHs) and improved her height. More investigations showed the presence of Hashimoto thyroiditis with normal thyroid stimulating hormone, so hormonal foll

Femoral-facial syndrome: Report of 2 fetal cases.

Femoral-facial syndrome (FFS) is a congenital disorder, characterized by facial dysmorphism and femoral hypoplasia. We describe the prenatal ultrasound and autopsy findings of FFS in 2 female fetuses born to diabetic mothers. Prenatal ultrasound showed micrognathia, low-set dysplastic ears and very short femora. Autopsy also demonstrated cleft palate, hypoplastic genitalia and visceral malformations including interventricular communication and posthemorrhagic hydrocephalus. Histologic study showed hyperplasia of the islets of Langerhans and femoral cartilage abnormalities. The growth plate dis

Hypopituitarism and successful pregnancy.

Hypopituitarism is a disorder characterized by the deficiency of one or more of the hormones secreted by the pituitary gland. Hypopituitarism patients may present the symptoms of amenorrhea, poor pregnancy potential, infertility, and no production of milk after delivery. Successful pregnancy in hypopituitarism patient is rare because hypopituitarism is associated with an increased risk of pregnancy complications, such as abortion, anemia, pregnancy-induced hypertension, placental abruption, premature birth, and postpartum hemorrhage. Hypopituitarism during pregnancy and perinatal period should

Comparison of mineral metabolites as risk factors for adverse clinical outcomes in CKD.

Patients with chronic kidney disease are at increased risk for progressing to end-stage renal disease, developing cardiovascular disease, and dying prematurely. Recent evidence has suggested that disordered mineral metabolism, which includes hyperphosphatemia, secondary hyperparathyroidism, vitamin D deficiency, and fibroblast growth factor 23 excess, may contribute to the high rates of adverse outcomes in this population. However, marked within-subject variability for some of these biochemical parameters exists, potentially detracting from the utility of certain metabolites as prognostic tool
