A Metabolites' Interplay Can Modulate DNA Repair by Homologous Recombination.

Small molecules either derived from cell metabolic reactions or produced by gut bacterial flora have shown the potential of affecting gene expression, which suggests the possibility of interactions able to modulate cellular functions. In this context, the reported experiments were aimed at verifying a possible interplay between lactate and butyrate in modulating the efficacy of antineoplastic drugs. Butyrate is a product of gut bacterial flora, shown to be endowed with anticancer properties; conversely, increased lactate levels in cancer cells were found to be associated with higher proliferat

Biopsy RNA-seq captures TROP-2-linked migration and clonal resistance to forecast aggressiveness in metastatic melanoma.

BACKGROUND: Unlike many other solid tumors, melanoma cells possess a remarkable ability to dynamically transition between distinct transcriptional states in response to environmental cues or therapeutic pressure. Among the adaptive mechanisms underlying this plasticity, the epithelial-to-mesenchymal transition (EMT) has garnered increasing attention, as it facilitates the shift from a proliferative, epithelial-like phenotype to a more invasive, mesenchymal-like phenotype frequently associated with cellular dormancy, quiescence and resistance to therapy. Despite growing interest in this phenome

Prognostic and Clinicopathological Evaluation of NOTCH-1 Expression in Endometrial Carcinoma.

BACKGROUND AND OBJECTIVE: Endometrial carcinoma (EC) is the most common gynecologic malignancy. NOTCH-1, a transmembrane receptor involved in cell fate regulation, may act as an oncogene or tumor suppressor depending on context. Although NOTCH signaling is implicated in many cancers, its role in EC remains unclear. This study aimed to investigate the relationship between NOTCH-1 protein expression and a wide range of clinicopathological and immunohistochemical parameters, as well as its prognostic significance in EC. METHODS: A total of 259 patients with EC were retrospectively analyzed. Immu

The USP7-VIM axis mediates lipid metabolic remodelling and compromises CD8+ T-cell mitochondrial fitness in glioblastoma.

Glioblastoma (GBM) remains a highly lethal malignancy for which current treatments offer only a limited therapeutic benefit. Although vimentin (VIM) is classically regarded as a cytoskeletal protein, its role in tumour immune regulation is poorly understood. Here, we showed that extracellular tumour-derived VIM was associated with impaired CD8+ T-cell antitumour immunity and altered metabolic fitness. Single-cell RNA sequencing revealed that CD8+ T cells within VIM-high tumour microenvironments exhibited dysfunctional transcription in GBM. Functional assays revealed that VIM knockdown (VIM-KD)

Tumor-derived progranulin promotes macrophage cholesterol efflux and associated immunosuppressive features in oral squamous cell carcinoma.

BACKGROUND: Cholesterol efflux is increasingly recognized as an important regulator of macrophage polarization and immunosuppressive features; however, the tumor-derived signals that govern this process in oral squamous cell carcinoma (OSCC) remain poorly defined. METHODS: We performed integrated single-cell RNA sequencing (scRNA-seq) on primary OSCC tumors (n = 3) and validated the findings in 77 head and neck squamous cell carcinoma (HNSCC)-related samples across five public datasets. The role of tumor-derived progranulin (PGRN) in macrophage cholesterol efflux and associated phenotypic cha

Humanized anti-Claudin-1 antibodies to treat colorectal cancer.

Claudin-1 (CLDN1), a tight junction protein overexpressed and mis-localized in colorectal cancer (CRC), plays a critical role in tumor progression, stemness, and therapy resistance. Integrative analyses of bulk and single-cell transcriptomic datasets revealed that CLDN1 is enriched in stem-like CRC cells, increases during metastatic progression, and is associated with microsatellite stable disease. High CLDN1 expression correlates with epithelial-to-mesenchymal transition and activation of oncogenic pathways including AKT/mTOR, Myc, and NF-κB. Here, we evaluate the therapeutic efficacy of an i

ID2 Suppresses Multiple Myeloma Cell Proliferation by Repressing the Activity of the Transcription Factor TCF3.

UNLABELLED: Transcription factors and their cofactors are major and selective nononcogene dependencies in multiple myeloma cells. By performing a gain-of-function perturbation screen in human multiple myeloma cell lines, we identified the inhibitor of DNA binding (ID) genes as putative suppressors of multiple myeloma cell fitness. Among them, ID2 was found to be downregulated in multiple myeloma patient cells and acted as a tumor suppressor by directly binding and repressing the basic helix-loop-helix factor TCF3, also known as E2A. Lower ID2 expression in multiple myeloma cells conferred a pr

Coordinated Multicellular Immune Programs and Drug Targets Revealed by Single-Cell Analysis in Driver-Mutated NSCLC.

Oncogenic driver mutations in non-small cell lung cancer (NSCLC) activate defined signaling pathways that sustain tumor growth and influence the immune landscape. Yet, how coordinated interactions among diverse cell populations within the tumor immune microenvironment (TIME) contribute to this process remains largely unresolved. To address this, we profiled approximately 200,000 single cells from 45 treatment-naïve NSCLC patients representing seven major driver mutations. This analysis uncovered five multicellular modules (CM1-5) with distinct functional properties, each linked to specific mal

The role of microRNAs in non-small cell lung cancer response and resistance to immune checkpoint inhibitors: From potential biomarkers to therapeutic tools.

Immunotherapy with immune checkpoint inhibitors (ICIs) has been a breakthrough in the treatment of Non-Small Cell Lung Cancer (NSCLC) patients. Nonetheless, only a subset of patients responds to this treatment, and those who respond will eventually develop resistance. Recent evidence supports the strong association of numerous microRNAs (miRNAs) with ICIs resistance mechanisms in NSCLC. This review presents the latest findings from studies on patients' samples, strongly suggesting a role for miRNAs in NSCLC resistance to ICIs. Collective data highlights the emerging potential of miRNAs as pred

Mucin expression in pancreatic ductal adenocarcinoma cell lines in 2D and 3D cultures: A proteomic and immunocytochemical analysis.

Pancreatic ductal adenocarcinoma (PDAC) exhibits diverse phenotypes, including epithelial and mesenchymal characteristics, yet these features have not been effectively translated into clinical applications. Mucins are implicated in tumor progression and therapeutic resistance and are considered potential diagnostic and therapeutic targets. In this study, five epithelial and three mesenchymal PDAC cell lines were cultured under two-dimensional (2D) and three-dimensional (3D) conditions to investigate mucin expression. Proteomic analysis identified five mucins (MUC1, MUC4, MUC5B, MUC19, and MUC2

Targeting NOP14 remodels the tumor immune microenvironment and enhances the antitumor efficacy of PD-1 blockade in DLBCL.

BACKGROUND: Nucleolar Protein 14 (NOP14), a highly conserved factor in eukaryotes, is essential for pre-18S rRNA processing and small ribosomal subunit assembly. Emerging evidence underscores its pivotal regulatory role in the progression of various malignancies. Nevertheless, its functional involvement in Diffuse large B-cell lymphoma (DLBCL), particularly its capacity to modulate the tumor immune microenvironment, remains to be fully elucidated. METHODS: Genomic and clinical data from DLBCL patients were retrieved from the GEO database (GSE12195, GSE32018, GSE181063 and GSE10846) for integr

Large-scale and high-resolution mass spectrometry-based proteomics defines molecular subtypes of nasopharyngeal carcinoma for therapeutic targeting.

Challenges in the precise diagnosis and treatment of nasopharyngeal carcinoma (NPC) remain, mainly due to the absence of a multi-omics-based molecular classification and effective targeted therapies. In this study, we performed proteomic and phosphoproteomic analyses of NPC and non-cancerous nasopharyngeal tissues to identify key dysregulated proteins and phosphorylation networks. Based on these profiles, we classified NPC into two distinct molecular subtypes: S1 and S2, which exhibit significant clinical heterogeneity. Notably, the S2 subtype displayed stronger immune suppressive characterist

Modulation of cell migration and invasion of thyroid cancer cells using miRNA-enriched extracellular vesicles.

BACKGROUND: Extracellular vesicles (EVs) have been investigated as promising vehicles for delivering drugs and microRNAs (miRNAs) in distinct types of cancer. However, their use as a delivery system in papillary thyroid carcinoma (PTC) remains underexplored. PTC is the most prevalent subtype of thyroid cancer, and despite its good prognosis, a fraction of the cases evolve into aggressive and fatal forms. Here, we investigated whether non-tumor thyroid cells can be engineered to produce EVs-enriched with tumor suppressor miRNAs that could be delivered to PTC cells. METHODS: EVs from NThy-ORI c
