Non-Hodgkin's lymphoma (NHL) is a prevalent hematological malignancy that includes a variety of B-cell and T-cell proliferations. The S1P (sphingosine-1-phosphate) pathway, involved in cell survival, proliferation, and migration, plays a critical role in NHL pathogenesis. Targeting S1PR1, the receptor for S1P, may provide a therapeutic strategy for NHL. The primary objective of this study was to identify and evaluate the efficacy of S1P analogues against the S1PR1 receptor through computational methods and experimental validation. The crystal structure of S1PR1 was obtained from the Protein Da

Myeloid-derived suppressor cells (MDSCs) are a heterogeneous group of immature myeloid cells playing a critical role in immune suppression. In vitro-generated MDSCs are a convenient tool to study the properties of tumour-associated MDSCs. Here, we compared six protocols for in vitro generation of functional mouse MDSCs from bone marrow progenitors. The protocols included granulocyte-macrophage colony-stimulating factor (GM-CSF) alone or in combination with interleukin-6 (IL-6) or granulocyte colony-stimulating factor (G-CSF), with or without a tumour-conditioned medium (TCM) derived from B16-F

The retinal pigment epithelium (RPE) cells are a single layer of cells with specific functions in vision. The Mertk gene, encoding a receptor tyrosine kinase, is critical for the phagocytic function of retinal RPE cells. Mutations in Mertk disrupt RPE function and contribute to retinal degeneration. This study aims to characterize the Mertk mutation in Royal College of Surgeons (RCS) rats and develop a polymerase chain reaction (PCR)-based method for genotyping these mutations to improve colony management. DNA was extracted from mutant Mertk-/- and wild-type rats, followed by PCR amplification

Ancestral sea gardens in British Columbia (BC) were managed to provide high long-term yields for millennia before European contact. While the ubiquity and productivity of sea gardens are well known, the ecological and cultural impact of the transition from ancestral to settler-colonial fisheries has not been investigated in a historical ecology context. Here, we examine that transition in the BC clam fishery, documenting the social and legal mechanisms used to dispossess Indigenous clam harvesters and identifying the ecological impacts of settler-colonial fisheries. Our results document a rapi

BackgroundPatients with hematological malignancies (HM) are at risk for complications, including neutropenia and admission to the intensive care unit (ICU). Granulocyte colony-stimulating factors (GCSF) can accelerate progenitor cells' proliferation and differentiation, and thus compensate for neutropenia. In patients with HM admitted to ICUs in Saudi Arabia, the outcome and impact of GCSF use on neutropenia duration and severity is understudied.ObjectiveTo evaluate the outcome and impact of GCSF on neutropenia in patients with HM admitted to the ICU of a tertiary care center in Saudi Arabia.M

BACKGROUND: Prostate and pancreatic cancers pose significant global health challenges. This study explored the potential of compound 5b, a novel phthalimido-1,3-thiazole derivative, as an anticancer agent against these malignancies.
METHODS: In vitro, compound 5b exhibited potent cytotoxic activity against both prostate (DU-145 and PC-3) and pancreatic (Panc-1 and Mia Paca-2) cancer cell lines. Notably, it significantly reduced colony formation in PC-3 cells, potentially hindering tumor growth. Furthermore, treatment with compound 5b suppressed cell migration and induced cell cycle arrest in

OBJECTIVE: Hepatocellular carcinoma (HCC), a lethal cancer with high global mortality, may be targeted through ferroptosis, an iron-dependent form of cell death. Despite its potential, the prognostic value of ferroptosis in HCC is underexplored.
METHODS: Our study leveraged single-cell and bulk sequencing datasets to identify ferroptosis-related genes and developed a prognostic model via Cox and LASSO regression analyses. Survival and mutation analyses led to the creation of a nomogram for predicting patient prognosis. Furthermore, we investigated the role of GRINA, a ferroptosis-related gene

Stray cats potentially act as reservoir for zoonotic agents, posing a risk of exposure to humans and domestic cats. The most prevalent Chlamydiaceae species in cats is Chlamydia (C.) felis, which is frequently associated with conjunctivitis and/or upper respiratory disease. The zoonotic potential of C. felis is believed to be relatively low, although exposure is possible through handling infected cats, by contact with their aerosol, and via fomites. Infection is more frequent in conditions of overcrowding, stress, poor hygiene and impairment of the immune system. For this reason, stray cats ap

Antimicrobial compounds are essential for controlling bacterial infections. Stress-induced bacterial tolerance and persisters can undermine antimicrobial activities under laboratory conditions, but their quantitative effects under physiological conditions remain unclear1,2. Here we determined constraints on clearance of Salmonella by antimicrobials in infected mice and tissue-mimicking chemostats. The antibiotics enrofloxacin and ceftriaxone exhibited poor anti-Salmonella activity under both conditions, primarily owing to severe nutrient starvation, which restricted Salmonella replication3-5.

BACKGROUND/OBJECTIVES: The tongue harbors about two-thirds of the microorganisms present in the mouth; the stable bacterial population consists mainly of aerobic and facultative anaerobic streptococci. These bacterial colonies, found more frequently on the tongue than on the outside of the hard part of the dental enamel in children younger than 18 months, suggest that the tongue is a potential bacterial reservoir. The aim of this review is to examine the scientific literature to clarify whether the mechanical removal of bacterial biofilm on the tongue can have a positive effect on caries preve

Parasites are ubiquitous in wildlife populations and have a profound impact on population dynamics. Interest in parasites of wildlife has increased significantly in recent years, particularly in those with relevant conservation status. Patagonia is one of the wildest and remote areas of the world. The Wolffsohn's viscacha lives in a small mountainous area of Patagonia. Until now, little is known about the biology and ecology of this species. The aim of this research was to study the gastrointestinal parasite diversity in this rodent from a coprological survey. A total of 125 fecal samples from

We have previously reported the transcriptomic and lipidomic profile of the first-generation, hygromycin-resistant (HygR) version of the BCGΔBCG1419c vaccine candidate, under biofilm conditions. We recently constructed and characterized the efficacy, safety, whole genome sequence, and proteomic profile of a second-generation version of BCGΔBCG1419c, a strain lacking the BCG1419c gene and devoid of antibiotic markers. Here, we compared the antibiotic-less BCGΔBCG1419c with BCG. We assessed their colonial and ultrastructural morphology, biofilm, c-di-GMP production in vitro, as well as their tra
