[Approaches to the combined therapy of helminthiases with leukinferon and etiotropic preparations].

The results of therapy with anthelmintics in combination with leukinferon show either higher efficiency of anthelmintics (medamin) or their same efficiency (albendazole). The outcomes of combined treatment of infected animals with anthelmintics and leukinferon are different and associated with the time of leukinferon administration as to the tissue stages of parasitic development, with the host's sensitivity and susceptibility to helminth.

Severe angiostrongyliasis with neuropsychiatric symptoms in vulnerable adults: Early diagnosis via next-generation sequencing and successful treatment.

Angiostrongylus cantonensis (AC) is a parasite that occasionally infects humans, causing eosinophilic meningitis (EM). In this study, we identified two adult males with severe angiostrongyliasis being treated at the ICU. Both presented with mental and behavioral abnormalities and were initially treated for psychiatric disorders before showing symptoms like cough, fever, and altered consciousness. Patient 1, a homeless individual, presented with elevated eosinophil levels and typical meningitis symptoms, prompting an early diagnosis of EM that was later confirmed via next-generation sequencing

Receptor for advanced glycation end products regulates the pulmonary and intestinal host responses to the infection with the parasitic nematode Nippostrongylus brasiliensis.

Parasitic nematode infection elicits goblet and tuft cell hyperplasia in host intestinal mucosa, resulting in the expulsion of intestinal nematodes. The receptor for advanced glycation end products (RAGE, alias Ager) is associated with pro-inflammatory responses following invasion by intestinal nematodes. We found here that the survival rate of RAGE-deficient Ager-null mice infected with Nippostrongylus brasiliensis (N. brasiliensis) larvae was higher than that of wild-type (WT) mice. In the lungs of infected mice, the destruction of the pulmonary alveoli was less pronounced in Ager-null mice

Experimental evolution of Plasmodium yoelii in single and helminth-coinfected mice.

BACKGROUND: Coinfection has the potential to affect key traits describing the infection dynamics, the severity of the disease and in fine parasite fitness. However, despite its pervasiveness, experimental work investigating how parasites adapt to the conditions provided by a coinfected host is mostly missing. METHODS: We adopted an experimental evolution approach to investigate if coinfection with the nematode Heligmosomoides polygyrus (Hp) affected the infection dynamics and virulence of the murine malaria parasite Plasmodium yoelii (Py). To this purpose, lines of Py were passaged either in

Inflammatory and immunopathological differences in brains of permissive and non-permissive hosts with Angiostrongylus cantonensis infection can be identified using 18F/FDG/PET-imaging.

BACKGROUND: Angiostrongylus cantonensis is a parasite that mainly infects the heart and pulmonary arteries of rats and causes human eosinophilic meningitis or meningoencephalitis in certain geographical areas. Current diagnostic methods include detection of the parasite in cerebrospinal fluid (CSF) and eosinophilic immune examination after lumbar puncture, which may be risky and produce false-positive results. 18F- Fluorodeoxyglucose (FDG), a Positron emission tomography (PET) tracer, has been used to assess different pathological or inflammatory changes in the brains of patients. In this stud

Identifying the function of novel cross-species microRNAs from the excretory-secretory products of Angiostrongylus cantonensis fifth-stage larvae.

BACKGROUND: Angiostrongylus cantonensis is a significant foodborne zoonotic parasite that causes severe neuropathological damage and symptoms in humans. Excretory-secretory products (ESPs) play a pivotal role in elucidating host-parasite interactions and can aid in penetrating host defensive barriers in helminths. Recently, secreted microRNAs have become important research targets for parasite-host communication. In this study, we determined the expression and function of novel microRNAs from A. cantonensis L5 ESPs and evaluated the effect of target microRNAs on the molecular mechanisms of mou

Natural and vaccine-mediated immunity to Salmonella Typhimurium is impaired by the helminth Nippostrongylus brasiliensis.

BACKGROUND: The impact of exposure to multiple pathogens concurrently or consecutively on immune function is unclear. Here, immune responses induced by combinations of the bacterium Salmonella Typhimurium (STm) and the helminth Nippostrongylus brasiliensis (Nb), which causes a murine hookworm infection and an experimental porin protein vaccine against STm, were examined. METHODOLOGY/PRINCIPAL FINDINGS: Mice infected with both STm and Nb induced similar numbers of Th1 and Th2 lymphocytes compared with singly infected mice, as determined by flow cytometry, although lower levels of secreted Th2,

Acute neurological signs as the predominant clinical manifestation in four dogs with Angiostrongylus vasorum infections in Denmark.

Four dogs with acute neurological signs caused by haemorrhages in the central nervous system were diagnosed with Angiostrongylus vasorum infection as the underlying aetiology. Two dogs presented with brain lesions, one dog with spinal cord lesions and one with lesions in both the brain and spinal cord. Only one dog presented with concurrent signs of classical pulmonary angiostrongylosis (respiratory distress, cough), and only two dogs displayed overt clinical signs of haemorrhages. Results of coagulation assays were inconsistent. Neurological signs reflected the site of pathology and included

Anaphylactic release of mucosal mast cell granule proteases: role of serpins in the differential clearance of mouse mast cell proteases-1 and -2.

The granule-derived mouse mast cell proteases-1 and -2 (mMCP-1 and -2) colocalize in similar quantities in mucosal mast cells but micrograms of mMCP-1 compared with nanograms of mMCP-2 are detected in peripheral blood during intestinal nematode infection. This differential systemic response was investigated both in vitro and in vivo. Bone marrow-derived mucosal mast cell homologs released similar quantities of mMCP-1 and-2 concomitantly with beta-hexosaminidase in response to calcium ionophore ( approximately 60% release) or IgE/DNP (25% release). In contrast, serum from mice sensitized by inf

Effects of intestinal nematodes during lactation: consequences for host morphology, physiology and offspring mass.

Sublethal parasites are often assumed to have no detrimental effects on their host. However, the sublethal intestinal nematode Heligmosomoides polygyrus affects both the morphology and the physiology of its laboratory mouse (Mus musculus) host and therefore has the potential to affect host life history. The objectives of the present study were to determine (1) whether lactating and non-lactating mice responded similarly to experimental infection with H. polygyrus and (2) whether the changes in morphology and physiology that occurred with parasite infection affected host reproductive performanc

Differential production of interferon-gamma and interleukin-4 in response to Th1- and Th2-stimulating pathogens by gamma delta T cells in vivo.

Exposure to various pathogens can stimulate at least two patterns of cytokine production by CD4-positive T cells. Responses that result in secretion of interferon-gamma (IFN-gamma), lymphotoxin and interleukin-2 (IL-2) are classified as T-helper-1 (Th1); CD4+ T-cell production of IL-4, IL-5, IL-9, IL-10 and IL-13 is called a T-helper-2 response (Th2). Differentiation of CD4+ T cells into either Th1 or Th2 cells is influenced by the cytokine milieu in which the initial antigen priming occurs. Here we use flow cytometry to identify the presence of intracellular cytokines (cytoflow) and analyse T

Activation of the COX-2/mPGES-1/PGE-2 cascade through the NLRP3 inflammasome contributes to Angiostrongylus cantonensis-induced eosinophilic meningoencephalitis.

Prostaglandin E2 (PGE-2) is synthesised by cyclooxygenase-2 (COX-2) and microsomal prostaglandin E synthase 1 (mPGES-1). PGE-2 exhibits pro-inflammatory properties in inflammatory conditions. However, there remains limited understanding of the COX-2/mPGES-1/PGE-2 pathway in Angiostrongylus cantonensis-induced meningoencephalitis. This study revealed several key findings regarding the activation of the COX-2/mPGES-1/PGE-2 pathway and its correlation with eosinophilic meningoencephalitis induced by A. cantonensis infection. Immunostaining revealed an increase in the expression of COX-2 and mPGES

Intestinal nematode infection exacerbates experimental visceral leishmaniasis.

Leishmania donovani exposure often results in subclinical infection in immunocompetent individuals, and the factors dictating development of visceral leishmaniasis (VL) are not known. Infection with intestinal worms skew immunity towards type 2 and regulatory responses, thereby theoretically increases susceptibility to intracellular infections controlled by type 1 responses. Here we have tested how chronic infection with the intestinal nematode Heligmosomoides polygyrus affected immunity to a secondary infection with L donovani. We found that mice infected with H polygyrus displayed higher Lei

Angiostrongylus (Parastrongylus) cantonensis on intermediate and definitive hosts in Ecuador, 2014-2017.

Introduction: Angiostrongylus cantonensis is a serious public health problem and is widely distributed in the country. When the parasite infects the snails, it becomes deeply embedded in their interior and accidentally, it can infect people and other mammals. Objective: To establish the geographical distribution of A. cantonensis intermediate hosts (Achatina fulica) and definitive hosts (Rattus spp.) in Ecuador from 2014 to 2017. Materials and methods: We collected 2,908 A. fulica specimens in 16 provinces using the capture method per unit of effort for 30 minutes. We captured 211 hosts of whi

Comparison of RELMα and RELMβ Single- and Double-Gene-Deficient Mice Reveals that RELMα Expression Dictates Inflammation and Worm Expulsion in Hookworm Infection.

Resistin-like molecules (RELMs) are highly expressed following helminth infection, where they impact both the host and helminth. While RELMα (Retnla) impairs helminth expulsion by inhibiting protective Th2 immunity, RELMβ (Retnlb) can promote its expulsion. We employed Retnla(-/-) and Retnlb(-/-) mice to delineate the function of both proteins following infection with Nippostrongylus brasiliensis, a hookworm that infects the lung and intestine. Whereas wild-type (WT) and Retnlb(-/-)mice exhibited equivalent infection-induced inflammation, Retnla(-/-) mice suffered a heightened inflammatory res

Macrophage-derived human resistin is induced in multiple helminth infections and promotes inflammatory monocytes and increased parasite burden.

Parasitic helminth infections can be associated with lifelong morbidity such as immune-mediated organ failure. A better understanding of the host immune response to helminths could provide new avenues to promote parasite clearance and/or alleviate infection-associated morbidity. Murine resistin-like molecules (RELM) exhibit pleiotropic functions following helminth infection including modulating the host immune response; however, the relevance of human RELM proteins in helminth infection is unknown. To examine the function of human resistin (hResistin), we utilized transgenic mice expressing th

Enhanced protection against Heligmosomoides polygyrus in IL-2 receptor β-chain overexpressed transgenic mice with intestinal mastocytosis.

IL-2 receptor β-chain overexpressed transgenic (Tg2Rβ) mice lack NK cells, but the development of other lymphocyte subsets and macrophages remained apparently intact. These mice also exhibit intestinal mastocytosis. Helminth infection induces various immune responses, such as mast cells, goblet cells, eosinophils and IgE, mediated by Th2 cytokines. IL-4 is also important in the regulation of resistance and susceptibility to Heligmosomoides polygyrus infection. However, there are contradictory results about the relation between resistance to H. polygyrus and intestinal mastocytosis. The present
