Lipidic Prodrug Co-Crystals: A Platform for Combination Therapy Targeting Fungal Biofilms.

Fungal infections are often complicated by biofilm formation and concurrent inflammation, limiting the efficacy of conventional antifungal therapies. To address these challenges, we developed a novel hybrid drug delivery platform-lipidic prodrug co-crystals (LPCCs)-that combines the benefits of lipidic self-assembly and pharmaceutical co-crystallization. In this study, a lipidic prodrug is synthesized by linking catechol-containing α-aminophosphonate with phenylboronic acid-modified bifonazole (Bfz), an antifungal agent, via boronate bonding. The resulting self-assembled structures exhibit hig

Regulation of hepatic Sirt1 expression and lipid metabolism through TNF receptor signaling.

The production of tumor necrosis factor -alpha (TNFα) has been associated with fatty liver disease (i.e, hepatosteatosis) for many years. In fact, cytokine production has been thought of as a consequence of hepatic lipid accumulation which then becomes a critical factor in the development of chronic liver pathologies as well as in the pathogenesis of insulin resistance. The purpose of this study was to test the hypothesis that TNFα directly regulated lipid metabolism in liver. Wild type mice and mice lacking the receptor for TNFα (TNFR1-/-) were fed control diet or a choline-deficient diet. In

Lipidated SNAPP-Stars Target and Kill Multidrug-Resistant Bacteria within Minutes.

The fast emergence of bacteria resistance has already threatened global health, and immediate action is required before the emergence of another global pandemic. Despite substantial progress in the chemical synthesis of novel antimicrobial compounds and advancements in understanding antimicrobial resistance, there has been only a handful of new antibiotics coming to the market. Structurally Nanoengineered Antimicrobial Peptide Polymers (SNAPP-stars) are a new class of antimicrobials. Here, we show that lipidation of lysine-valine 16-armed SNAPP-star, S16 (lipo-SNAPP-star) where the N-terminal

APOE Genotype Impacts Polyunsaturated Neutral Lipid Storage and Ferroptosis Vulnerability in Astrocytes.

Astrocytes play a critical role in regulating the metabolism of cholesterol and other lipids in the brain. Increasing evidence suggests that dysfunctional lipid metabolism and lipid peroxide-mediated cell death, or ferroptosis, are involved in the pathophysiology of late-onset Alzheimer’s disease. The Alzheimer’s disease risk allele of APOE, APOE4, is expressed in astrocytes and is associated with increased accumulation of cholesterol as well as highly polyunsaturated and peroxidation-prone neutral lipids in lipid droplets. However, a direct connection between APOE4 genotype-dependent lipid dy…

Resolution of innate immune cells with proresolving lipid mediators in idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive incurable lung disease characterized by chronic inflammation and fibrosis, with innate immune cells such as neutrophils and macrophages playing central roles in its pathogenesis. This review explores the involvement of these immune cells in the inflammatory process of IPF, focusing on their contribution to disrupted tissue repair and impaired resolution. The balance between host defense mechanisms, including leukocyte recruitment, and the release of proresolving mediators is crucial for maintaining healthy tissue function and returning to pr

An ANGPTL4 inhibitory antibody safely improves lipid profiles in non-human primates.

BACKGROUND: Angiopoietin-like protein 4 (ANGPTL4) inhibition is a promising approach to manage atherogenic dyslipidaemia and residual atherosclerotic cardiovascular disease (ASCVD) risk. Human ANGPTL4 loss-of-function (LoF) is associated with reduced plasma triglyceride (TG), remnant cholesterol (RC), and apolipoprotein B (ApoB) levels, and lower risk of type 2 diabetes and ASCVD, without observable safety concerns. However, development of ANGPTL4 inhibitors has been stalled by adverse findings in Angptl4 knockout mice fed a high-saturated-fat diet (HSFD), which show lipid accumulation in mese

In vitro biophysical and biological profiling of commercial lipid-based dry eye products.

There is an increasing number of products available for treatment of dry eye disease (DED), thereby creating a challenge in selecting a suitable product. Commercial products have rarely been studied in the same experimental setup, and in the case of lipid-based products, their baseline capabilities to target central defects of DED requires a more thorough investigation. This study aims to discern potential differences in their abilities to stabilize the tear film, reduce the evaporation of water, and impact on corneal epithelial cell viability and recovery utilizing various biophysical and bio

Commentary for: a lipid scramblase TMEM41B is involved in the processing and transport of GPI-anchored proteins.

Glycosylphosphatidylinositol (GPI) anchoring is a conserved post-translational modification in eukaryotes. This modification allows acceptor proteins to be expressed at the cell surface as GPI-anchored proteins (GPI-APs), which play critical roles in various biological processes. It has been proposed that remodelling of GPI after transferring acceptor proteins, including the PGAP1-dependent deacylation of GPI-inositol, functions as a checkpoint for transporting mature GPI-APs from the endoplasmic reticulum (ER) to the Golgi. A previous study identified several factors involved in regulating PG

A mechanistic model of pure and lipidic α-synuclein aggregation for advancing Parkinson's therapies.

Alpha-synuclein (aSyn) plays a crucial role in Parkinson's disease, with various aggregates proposed as pathogenic triggers and therapeutic targets. However, anti-aSyn aggregation compounds often fail due to limited knowledge of the underlying molecular basis. In particular, interactions with lipid membranes are central to both physiological and pathological roles of aSyn, yet their underlying mechanisms remain unclear. Disrupting this balance may drive Parkinson's onset and progression, underscoring the need for a mechanistic understanding of pure and lipid-mediated aggregation. Building on w

Lipid-free, thermostable mRNA vaccines prepared using atomic layer deposition.

Lipid nanoparticle (LNP) formulations of mRNA vaccines have played a pivotal role in combatting SARS-CoV-2 infections and are expected to be a useful vaccine modality against other pathogens. However, the instability of mRNA-LNP vaccines requires their storage at temperatures below 0 °C and presents manufacturing challenges. We describe thermally stable, lipid-free mRNA vaccines prepared by spray-drying to embed mRNA within glassy polysaccharide microparticles, followed by atomic layer deposition (ALD) to encapsulate the microparticles within protective alumina shells that provide temporally c

Effects of Thermal Sterilization Conditions on Flavor and Lipid Oxidation of Sauced Duck Necks.

This study aimed to investigate the effect of thermal sterilization on the volatile flavor of sauced duck necks. The study revealed that thermal sterilization significantly reduced the content of unsaturated fatty acids (e.g., oleic acid C18:1n9c) in sauced duck necks. This was accompanied by elevated thiobarbituric acid reactive substances (max 0.86 mg/100 g) and peroxide values (max 1.13 g/100 g), indicating intensified lipid oxidation. Through PLS-DA, six key differential free fatty acids distinguishing the sterilization treatment groups were identified: cis-9-tetradecadecarbonate, methyl t
