Antigen and molecular testing using one swab: Lessons learned from a COVID-19 project.

Rapid SARS-CoV-2 detection is essential in limiting transmission. We evaluated the performance of RT-PCR on the same nasopharyngeal swab used for Panbio antigen testing. High concordance with standard PCR and improved case detection-including asymptomatic infections-demonstrates this one-swab-two-test strategy enhances diagnostic yield while reducing patient discomfort and resource burden.

Long-term sero-epidemiological trends of Nine respiratory pathogens in a predominantly pediatric population in Southwest China: a multicenter retrospective study (2019-2023).

BACKGROUND: Acute respiratory tract infections (ARTIs) remain a major global health burden. Southwest China, with its complex geography and climate, represents a critical region for respiratory pathogen ecology. This study aimed to characterize the epidemiological dynamics of nine respiratory pathogens among ARTI patients in Southwest China across the pre-COVID-19 period (January 2019 to December 2019), the COVID-19 non-pharmaceutical interventions (NPIs) period (January 2020 to December 2022), and the post-NPIs period (January 2023 to December 2023). METHODS: A multicenter, retrospective stu

Discrepant toxoplasma latex agglutination test results.

The analysis of 4450 toxoplasma serology results showed that 59 (1.3%) latex agglutination reactions were not confirmed in the dye test. These discrepant results were associated with an unspecified IgM antibody but not associated with kit batch variation, inactivation of sera, concurrent cytomegalovirus infection, or the presence of hepatitis B virus "e" antigen. The latex agglutination test is useful as a screen for toxoplasma infection but false positive reactions do occur. Patients at risk of severe toxoplasmosis should be investigated by additional tests.

Polyvalent heat-killed antigen for the diagnosis of infection with Legionella pneumophila.

A polyvalent antigen composed of heat-killed agar-grown Legionella pneumophila serogroups 1-4 suspended in a suspension of yolk-sac from embryonated hens' eggs has been examined for use in the indirect fluorescent antibody test for Legionella infection. The serological response detected by monovalent antigen correlated well with that detected by polyvalent antigen. The use of polyvalent antigen forms a useful screening test for the detection of antibody to L pneumophila, but positive results must be confirmed by test using monovalent antigen.

The complement fixation test and African trypanosomiasis. II. The complement fixation test as an aid for assessing therapy.

Complement-fixing antibody titres decline after successful treatment of trypanosoma infected cattle during a fixed period. For investigating the practicability of the resulting double test procedure in the field, cattle under controlled drug regimes were tested at treatment and again 3 weeks later. Alternatively, the results from groups of treated and untreated animals from the same area were compared. Both serological and parasitological test results indicated an insuffieicnt effectiveness of therapy in the area under investibation.

A rapid micro-agglutination technique for the detection of antibody to Legionella pneumophila serogroup 5.

A rapid micro-agglutination test (RMAT) for the detection of antibody to Legionella pneumophila serogroup 5 is described. It was found to be both sensitive and specific when compared with the indirect immunofluorescence test. Evaluation of 89 paired sera from patients with respiratory symptoms showed that the incidence of L. pneumophila serogroup 5 respiratory infection in East Anglia is low: only one case was found in this study. The RMAT would be easy to perform as a screening test in a routine serological laboratory.

Understanding the patient's experience of coeliac disease diagnosis: a qualitative interview study.

BACKGROUND: Coeliac disease (CD) presents with non-specific symptoms, and delays to diagnosis are common. The traditional diagnostic pathway involves serological testing followed by endoscopic biopsy; however, the evidence is increasing about the effectiveness of a diagnosis without the need for a biopsy. AIM: To understand the patient's experience of being diagnosed with CD. DESIGN AND SETTING: A qualitative study was conducted, which involved semi-structured interviews with adults diagnosed with CD living in the UK. METHOD: Participants (n = 20) were purposefully sampled from 200 adults w

Production and evaluation of a new set of recombinant antigens for the serological diagnosis of human cysticercosis.

Human cysticercosis caused by Taenia soliun (T. soliun) is endemic in certain areas of Latin America, Asia and Sub-Saharan Africa. Neurocysticercosis (NCC) is mainly diagnosed by neuroimaging, which, in most cases, is unavailable in endemic areas. Due to their high sensitivity and specificity, serological tests such as enzyme-linked immunosorbent assay (ELISA) and Western blot (WB) based on the glycosylated fraction of the cyst CS50 are widely used for the detection of the anti-cysticercus IgG antibodies despite their significant cost and the need of cysticercus material. Given their cost-effe

Antigenic diversity of type 1 polioviruses and its implications for the efficacy of polio vaccines.

Inactivated Polio Vaccines (IPV) and live Oral Polio Vaccine (OPV) were introduced in the mid-20th century, and their coordinated worldwide use led to almost complete elimination of the disease, with only one serotype of poliovirus remaining endemic in just two countries. Polio eradication will lead to discontinuation of OPV use and its replacement with IPV or other vaccines that are currently under development that will need to be tested in clinical trials. Despite decades of research, questions remain about the serological correlates of polio vaccine efficacy, specifically whether the vaccin

The Tip of Brucella O-Polysaccharide Is a Potent Epitope in Response to Brucellosis Infection and Enables Short Synthetic Antigens to Be Superior Diagnostic Reagents.

Brucellosis is a global disease and the world's most prevalent zoonosis. All cases in livestock and most cases in humans are caused by members of the genus Brucella that possess a surface O-polysaccharide (OPS) comprised of a rare monosaccharide 4-deoxy-4-formamido-D-mannopyranose assembled with α1,2 and α1,3 linkages. The OPS of the bacterium is the basis for serodiagnostic tests for brucellosis. Bacteria that also contain the same rare monosaccharide can induce antibodies that cross-react in serological tests. In previous work we established that synthetic oligosaccharides, representing elem

External quality assurance of serological diagnosis of dengue, chikungunya and Japanese encephalitis virus infection.

INTRODUCTION: Dengue, chikungunya and Japanese encephalitis are the most common arthropod-borne viral diseases in India. Due to overlapping clinical symptoms, accurate, high-quality and timely laboratory-based differential diagnosis is essential for control and containment of outbreaks. This is most commonly done by detection of IgM antibodies in serum using enzyme-linked immunosorbent assays. The Resource Centre for Virus Research and Diagnostic Laboratories (VRDLs) in Pune, India organized an external quality assurance (EQA) study to check the accuracy of serological diagnostics in the VRDL

Evaluating peer-supported screening as a hepatitis C case-finding model in prisoners.

BACKGROUND: Hepatitis C Virus (HCV) infection is endemic in prison populations, and HCV management in prisons is suboptimal. Incarceration is a public health opportunity to target this cohort. Community peer support increases HCV screening and treatment uptake. Prison peer workers have the potential to support the engagement of prisoners with health services and reduce stigma. This study's primary aim is to evaluate peer-supported screening as a model of active HCV case finding with a secondary aim to describe the HCV cascade among those infected including linkage to care and treatment outcome
