Research status of facial nerve repair.

The facial nerve, also known as the seventh cranial nerve, is critical in controlling the movement of the facial muscles. It is responsible … of facial nerve repair. Different approaches, including direct suture, autologous nerve grafts, and tissue engineering, have been utilized for the repair of facial nerve injury. This article mainly summarizes the clinical methods and basic research progress of facial nerve repair in the past ten years.

Pathogenesis of wound healing disorders in the elderly.

The elderly constitute the age group most susceptible to wound healing disorders and chronic wounds, the most prevalent being venous leg ulc … herapeutic strategies for chronic wounds. In this context, the use of growth factors and cytokines, tissue engineering, and cell therapy - including stem cells - have proven very promising. Nevertheless, prior to their introduction into routine clinical practice, large controlled clinical trials are required to assess the safety of these techniques.

Photodegradable iron(III) cross-linked alginate gels.

Biocompatible photoresponsive materials are of interest for targeted drug delivery, tissue engineering, 2D and 3D protein patterning, and other biomedical applications. We prepared light degradable hydrogels using a natural alginate polysaccharide cross-linked with iron(III) cations. The "hard" iron(III) cations used to cross-link the alginate hydrogel were found to undergo facile photoreduction to "soft" iron(II) cations in the presence of millimolar concentrations of sodium lactate. The "soft" iron(II) cations have a decreased ability to cross-link the alginate which results in dissolution o…

Preliminary results of the application of a silk fibroin scaffold to otology.

The surgical treatment to repair chronic tympanic membrane perforations is myringoplasty. Although multiple autologous grafts, allografts, and synthetic graft materials have been used over the years, no single graft material is superior for repairing all perforation types. Recently, the remarkable properties of silk fibroin protein have been studied, with biomedical and tissue engineering applications in mind, across a number of medical and surgical disciplines. The present study examines the use of silk fibroin for its potential suitability as an alternative graft in myringoplasty surgery by …

Tooth agenesis: in search of mutations behind failed dental development.

Tooth agenesis are the most common craniofacial malformations. Its prevalence in permanent dentition reaches 20% and its expressivity ranges from only one tooth, usually a third molar, to the whole dentition. Genetic linkage and molecular biology studies have allowed, in the last decade, the identification of mutations responsible for some patterns of syndromic and nonsyndromic tooth agenesis. The mutated genes are key genes for the development of dentition, like the ones that encode the transcription factors MSX1, PAX9 and PITX2, the signalling protein EDA and its receptor EDAR. Current resea

Methods for targeting biologicals to specific disease sites.

Cytokines are mediators of cell communication. Their therapeutic use requires frequent high doses to achieve effective local biological levels. However, the clinical use of some cytokines is limited because of their pleiotropism, which can result in unwanted side effects. Here, we review novel protein engineering technologies that overcome these limitations and enable the targeting of cytokines to specific sites. One such technology uses antibody-based recognition to direct the cytokine to a particular tissue, and another creates encapsulated latent cytokines that are released only at the site

Experimental and theoretical investigation of aortic wall tissue in tensile tests.

BACKGROUND: Understanding the mechanical properties of aortic tissue is essential for developing numerical computation tools and assessing the risk of aortic aneurysm fractures. Tensile tests using aortic wall specimens allow for the determination of stress and strain depending on the location and direction of the sample. OBJECTIVE: The aim of this study was to perform a mechanical tensile test using canine aorta samples and create a numerical model of aortic tissue tension from the processed data. METHODS: Dogbone-shaped samples were dissected from canine aortic segments. The initial measur

Hierarchically aligned fibrin nanofiber hydrogel accelerated axonal regrowth and locomotor function recovery in rat spinal cord injury.

BACKGROUND: Designing novel biomaterials that incorporate or mimic the functions of extracellular matrix to deliver precise regulatory signals for tissue regeneration is the focus of current intensive research efforts in tissue engineering and regenerative medicine. METHODS AND RESULTS: To mimic the natural environment of the spinal cord tissue, a three-dimensional hierarchically aligned fibrin hydrogel (AFG) with oriented topography and soft stiffness has been fabricated by electrospinning and a concurrent molecular self-assembling process. In this study, the AFG was implanted into a rat dor…

ADSC-derived mitochondrial nanovesicles transplantation alleviates capsular fibrosis and inflammation and improves joint mobility in a rat model of adhesive capsulitis.

BACKGROUND: Adhesive capsulitis (frozen shoulder) is a prevalent condition characterized by shoulder pain and progressive motion loss. Mitochondrial metabolic dysregulation is an underlying driver of chronic inflammation and fibrosis. This study aimed to characterize mitochondrial metabolic abnormalities in patient capsular tissue and evaluate a therapy using adipose-derived stem cell (ADSC) derived mitochondrial nanovesicles transplantation. METHODS: Single-cell RNA sequencing was utilized to analyze the expression of nuclear-encoded genes related to mitochondrial metabolism in fibroblast su

Evaluation of the effect of 3D-bioprinted gingival fibroblast-encapsulated ADM scaffolds on keratinized gingival augmentation.

BACKGROUND AND OBJECTIVES: The keratinized gingiva plays an important role in maintaining healthy periodontal and peri-implant tissue. Acellular dermal matrix (ADM), as a substitute biomaterial, has a porous structure and good biocompatibility. 3D-bioprinting has the potential for tissue engineering because it enables precise loading of cells layer-by-layer. Herein, we bioprinted ADM scaffold encapsulating gingival fibroblasts (GFs) and evaluated its efficacy in keratinized gingiva augmentation in vivo to assess its potential for clinical periodontal tissue regeneration. METHODS: GFs were ext…

A thin endometrium is a significant contributor to female infertility and adverse pregnancy outcomes and remains a persistent challenge in reproductive medicine. Stem cell therapy, when integrated with biomaterials and tissue engineering, has emerged as a promising approach for thin endometrial repair. This review systematically summarizes the critical molecular pathways involved in stem cell-based therapy for thin endometrium and explores the regulatory mechanisms within the immune microenvironment. This review focuses on the role of stem cells and their exosomes in promoting angiogenesis, ex…

A novel phosphorylated chitin-hydroxyapatite composite promotes angiogenesis and osteogenesis for bone defect repair.

Current bone repair materials face limitations including donor site morbidity, immune rejection, and insufficient osteogenic capacity. To overcome these challenges, tissue engineering has introduced bioactive composites that better mimic the natural bone microenvironment. Here, we developed a novel composite by integrating phosphorylated chitin, which is an excellent biocompatible and bioactive organic polymer, with biogenic hydroxyapatite, a mineral phase closely resembling native bone. The composite was systematically characterized for its physicochemical properties. In vitro studies demonst…
