Non-SMC condensin I complex subunit G (NCAPG) is implicated in tumor progression and TGF-β pathway activation, yet its roles in oncogenesis and immune modulation within the lung squamous cell carcinoma (LUSC) microenvironment remain largely unexplored. Here, we used bioinformatics analysis to identify key upregulated genes in LUSC and assessed the effects of NCAPG and its upstream regulator E2F transcription factor 8 (E2F8) on malignant phenotypes via gain- and loss-of-function assays in NCI-H226 and NCI-H1703 LUSC cell lines, with in vivo efficacy validated using KLN205 cell isografts and NTC

The contribution of putrescine (PUT) metabolism to impaired tumor immunosurveillance in colorectal cancer (CRC) requires a thorough examination of its biosynthesis, catabolism, and transport during tumor development. This study found, unexpectedly, that a better prognosis was associated with higher expression of the PUT biosynthesis genes ODC1 and AGMAT, while elevated expression of biosynthesis inhibitors and transport genes predicted worse outcomes. Two independent cohorts were deconvoluted by integrating data from 517,191 cells across 221 samples from 131 subjects. Higher percentages of epi

Emerging preclinical evidence challenges the long-standing assumption that Interleukin-1β (IL-1β) uniformly promotes non-small cell lung cancer (NSCLC). We show that, in the context of chemo-immunotherapy, IL-1β enhances anti-tumor immunity by inducing tumor-cell CXCL10 expression and recruiting CD8+ T cells, thereby sensitizing "cold" tumors to treatment. These findings contrast sharply with the failure of multiple CANOPY trials targeting IL-1β, suggesting that blockade may be effective only in prevention or early carcinogenesis. Instead, controlled IL-1β activation, guided by biomarkers and

Neoadjuvant chemotherapy (NAC) for colorectal cancer liver metastasis (CRLM) remains a major clinical challenge, with outcomes varying widely among patients, yet the mechanisms that drive therapeutic resistance remain incompletely understood. In this study, we integrated single‑cell RNA‑sequencing and spatial transcriptomics of patient tumor specimens to dissect the interplay between the hepatic tumor microenvironment and chemotherapy resistance. We identified a specific macrophage subpopulation marked by elevated expression of Secreted Phosphoprotein 1 (SPP1) that is significantly enriched in

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline accompanied by chronic neuroinflammation. Emerging evidence implicates T-cell infiltration and microglial activation as key immune events that accelerate AD pathology, yet therapeutic approaches targeting this neuroimmune interface remain scarce. Cobrotoxin (CTX), a short-chain neurotoxin derived from Naja atra venom, exhibits potent anti-inflammatory and immunomodulatory properties and is clinically approved in China for the treatment of chronic pain syndromes. Here, we investigated whether

BACKGROUND: Small cell lung cancer (SCLC), an aggressive neuroendocrine malignancy, exhibits high intertumoral heterogeneity and limited treatment options. Immune checkpoint inhibitors (ICIs) provide only modest benefits for SCLC, underscoring the need for clinically actionable phenotypes.
METHODS: Consensus clustering of bulk transcriptomic data identified SCLC molecular phenotypes. Bulk and single-cell RNA sequencing (scRNA-seq) revealed their molecular and immune characteristics, as well as tumor microenvironment interactions. Survival benefits of ICIs were assessed in 41 newly collected e

Natural systems use metal ions to form ordered structures that regulate biological processes, inspiring the rational design of nanotherapeutics. The cyclic guanosine monophosphate-adenosine monophosphate synthase-stimulator of interferon genes (cGAS-STING) pathway drives antitumor immunity but has been difficult to activate systemically owing to poor pharmacology and toxicity. Here, we report CRYSTAL, a structurally ordered intermetallic nanoparticle for potent systemic STING activation. CRYSTAL self-assembles from manganese ions intercalated with cyclic dinucleotides, enabling precise structu

BACKGROUND: Glioblastoma (GB) is the most aggressive primary brain tumor, characterized by poor prognosis, profound immunosuppression, and limited response to current therapies. To overcome these barriers, we developed CTX-CNF1, a brain-penetrant recombinant protein that integrates direct glioma targeting with myeloid cell-mediated immune reprogramming.
METHODS: CTX-CNF1 was generated by fusing Chlorotoxin (CTX), which enables selective delivery across the blood-brain barrier, with Cytotoxic Necrotizing Factor 1 (CNF1), a bacterial protein with antitumor and immunostimulatory properties. Ther

Bluetongue virus (BTV) is a vector-transmitted orbivirus that infects ruminant livestock and wildlife, causing substantial economic losses owing to its impact on animal health and trade. Despite extensive study, the mechanisms underlying infection and immune dynamics remain unclear. Here, we develop and calibrate a mechanistic within-host model of BTV infection in experimentally infected sheep to quantify the contributions of innate and adaptive immune responses to viral control. The model integrates longitudinal data on viral RNA dynamics reverse transcriptase quantitative polymerase chain re

BACKGROUND: Vaccination is critical for controlling infectious diseases; however, assessing efficacy solely through neutralizing antibody titers oversimplifies immune protection. The cellular and molecular mechanisms driving variable vaccine-induced immune responses remain underexplored, limiting comprehensive vaccine evaluation.
METHODS: Using single-cell sequencing, we profiled and compared cellular dynamic, transcriptomic profiles, the immune repertoire, and cellular communication in vaccine recipients with high and low antibody titers after Ad5-nCoV booster vaccination, using peripheral b

Prostate cancer (PCa) is a leading malignancy in men, particularly challenging in its advanced stages due to treatment resistance and immune evasion. MicroRNA-1290 (miR-1290) was identified to be implicated in tumor progression, but its role in PCa remains insufficiently characterized. Recent advances in exosome-based delivery systems provide opportunities to target oncogenic miRNAs in vivo. This study aimed to evaluate the therapeutic efficacy and immunomodulatory mechanism of exosome-delivered miR-1290 inhibitor in a PCa mouse model. Exosomes derived from RM-1 cells were loaded with miR-1290

Metabolic alterations are increasingly recognized during influenza virus infection, but how local lactate accumulation shapes antiviral immunity remains poorly characterized. By integrating time-series targeted energy metabolomics, single-cell RNA sequencing, flow cytometry, and functional perturbation, we show that influenza virus infection preferentially increases lactate within the lung microenvironment, where it restrains pulmonary CD8+ T cell response. Mechanistically, extracellular lactate enters dendritic cells through monocarboxylate transporter (MCT)-dependent transport and induces a

CD8 + T cells are indispensable effectors of adaptive immunity. While cytokine-mediated regulation of CD8 + T cell responses has been extensively characterized in mammals, the underlying mechanisms in fish remain largely unexplored. Here, we identified and characterized an interleukin-21 (IL-21) gene from Nile tilapia ( Oreochromis niloticus). In vitro stimulation with IL-21 significantly upregulated the transcription and expression of key CD8 + T cell effector molecules, including Granzyme B and interferon-gamma (IFN-γ), in splenic leukocytes, indicating a crucial role of IL-21 in modulating

Mycobacterium tuberculosis (Mtb) remains the leading cause of death from a single infectious agent, despite the availability of the Bacillus Calmette-Guérin (BCG) vaccine and antibiotic therapies. New tuberculosis (TB) vaccines are urgently needed to effectively protect adults against pulmonary Mtb infections, whether used alone, as a BCG booster, or as a therapeutic strategy. We developed TBpV1, a multivalent TB vaccine, using recombinant Pichinde virus (rPICV) vector rP18tri. TBpV1 comprises three rP18tri-vectored components that collectively deliver 13 known and novel antigens targeting bot

BACKGROUND: Oncolytic virus M1 (OVM), a naturally occurring alphavirus, has demonstrated potent antitumor activity in various solid tumor models by inducing immunogenic cell death and activating CD8+ T cells. However, its in vivo efficacy varies widely, and resistance mechanisms remain poorly understood. Tumor-associated macrophages (TAMs), key immunosuppressive cells within the tumor microenvironment, may limit OVM therapeutic potential.
METHODS: We investigated the role of TAMs in OVM resistance using multiple syngeneic mouse tumor models (MC38 colorectal cancer, KPC1199 pancreatic cancer,

In chronic inflammatory disorders of the central nervous system (CNS), tissue-resident self-reactive T cells perpetuate disease. The specific tissue factors governing the persistence and continuous differentiation of these cells remain undefined but could represent attractive therapeutic targets. In a model of chronic CNS autoimmunity, we find that oligodendrocyte-derived IL-33, an alarmin, is key for locally regulating the pathogenicity of self-reactive CD8+ T cells. The selective ablation of IL-33 from neo-self-antigen-expressing oligodendrocytes mitigates CNS disease. In this context, fewer
