A rare cause of primary amenorrhoea, the XY female with gonadal dysgenesis.

Disorders of sexual development are conditions where sexual phenotype and genotype are discordant. Genetic sex is determined at conception as the ovum is fertilised by a spermatozoon that contains either an X or Y chromosome. A complex pathway determined by genes and hormones leads to gonadal differentiation into testis or ovary and promotes the development of internal and external genitalia. We present a case of an 18-year-old woman who presented with primary amenorrhoea. She was a virgin, and apart from hirsutism and overweight, had no complaints. Her family history was insignificant. The pa

FOXL2 versus SOX9: a lifelong "battle of the sexes".

Testis determination in most mammals is regulated by a genetic hierarchy initiated by the SRY gene. Early ovarian development has long been thought of as a default pathway switched on passively by the absence of SRY. Recent studies challenge this view and show that the ovary constantly represses male-specific genes, from embryonic stages to adulthood. Notably, the absence of the crucial ovarian transcription factor FOXL2 (alone or in combination with other factors) induces a derepression of male-specific genes during development, postnatally and, even more interestingly, during adulthood. Predicting phenotype from genotype: normal pigmentation.

Genetic information in forensic studies is largely limited to CODIS data and the ability to match samples and assign them to an individual. However, there are circumstances, in which a given DNA sample does not match anyone in the CODIS database, and no other information about the donor is available. In this study, we determined 75 SNPs in 24 genes (previously implicated in human or animal pigmentation studies) for the analysis of single- and multi-locus associations with hair, skin, and eye color in 789 individuals of various ethnic backgrounds. Using multiple linear regression modeling, five

Iron delocalisation in the pathogenesis of malarial anaemia.

There is consensus that the pathophysiology of malaria-associated anaemia is multifactorial, but the precise mechanisms behind many of the haematological changes during malaria remain unclear. In this review, we attempt to build a composite picture of the pathophysiology of malarial anaemia using evidence from experimental, human and animal studies. We propose that cytokine- and hepcidin-mediated iron delocalisation, a principal mechanism in the anaemia of inflammation, plays an important role in the aetiology of malarial anaemia, and can explain some of the clinical and laboratory findings. Homocysteine levels in aqueous humor and plasma of patients with primary open-angle glaucoma.

We determined homocysteine (Hcy) levels in aqueous humor (AH) and plasma and their association with B-vitamin levels in patients with primary open-angle glaucoma (POAG) and controls. Both AH Hcy and plasma Hcy levels were significantly increased in POAG, and elevation of AH Hcy and plasma Hcy was a significant risk factor for POAG. In contrast to controls, neither plasma nor AH Hcy of POAG patients demonstrated a significant association with important non-genetic determinants of elevated Hcy such as low B-vitamin levels, increasing age and caffeine consumption. Considering that Hcy is a neurot

Human red blood cell polymorphisms and malaria.

Genetic factors are a major determinant of child survival in malaria endemic countries. Identifying which genes are involved and how they affect the malaria disease risk potentially offers a powerful mechanism through which to learn more about the host-parasite relationship. The past few years have seen significant progress towards achieving this goal for some of the best-known malaria resistance genes that determine the structure or function of red blood cells: Gerbich blood group antigen negativity; polymorphisms of the complement receptor genes (most notably CR1); Southeast Asian ovalocytos

Cytogenetic findings at Down syndrome and their correlation with clinical findings.

Down syndrome is a genetic state characterized by trisomy of chromosome 21. In the retrospective study for 12 years period (1991-2002) we have conducted correlation between cytogenetics analyses and clinical findings in our centre at 96 male and 83 female patients. Down syndrome was confirmed by cytogenetics analyses in 84 (87.5%) male patients and excluded in 12 (12.5%) male patients. Down syndrome was confirmed by cytogenetics analyses in 71 (85.5%) female patients and excluded in 12 (14.5%) female patients. Most common karyotype is free trisomy found in 139 (89.7%) examinees, than follows t

SRY and karyotypic status of one abnormal and two intersexual marsupials.

An intersexual agile wallaby (Macropus agilis) with a penis, a pouch and four teats had a sex-chromosome constitution of XXY in lymphocytes and cultured fibroblasts; the sex-determining region Y (SRY) gene was present, consistent with the presence of a testis. An intersexual eastern grey kangaroo (Macropus giganteus) with a small empty scrotum and no penis, and an abnormal red kangaroo (Macropus rufus) with no penis, pouch or teats, both had XX sex-chromosome complements; the SRY gene was not present, consistent with testis absence. The agile wallaby and grey kangaroo described here provide fu

Natural history and determinants of clinical severity of sickle cell disease.

Some of the factors determining the extremely variable clinical course of homozygous sickle cell disease are being identified. Genetic factors include alpha-thalassemia, beta-globin gene haplotypes, heterocellular hereditary persistence of fetal hemoglobin, and high total hemoglobin. Other factors include a variety of environmental variables and socioeconomic status. These risk factors, when occurring in conjunction with apparently random precipitating events, produce features of the disease. Many complications--which are age specific, with highest morbidity and mortality in the first 5 years-

Primary causes of fetal deaths in Athens a clinicopathological study.

The authors studied 239 cases of fetal death, occurring before the 24th week of gestation, during a five-year period, from 1989 to 1993. Based on clinical information and autopsy findings, their cases were classified as follows: 1) amniotic infection syndrome; 2) congenital anomalies; 3) disorders of the placenta and adnexae, and 4) others. The causes of fetal death were determined and evaluated statistically. In 83 cases (39 %), only autopsy revealed the cause of fetal death. The correlation of clinical and pathological findings showed that autopsy constitutes an important source of informati

[Normal and pathologic development of the secondary palate--a new concept].

Wether the secondary palate develops in the normal or pathological manner is determined by the following processes: Within the contact-epithelium of each palate process there is a still invisible differentiation boundary between the potential oral and nasal epithelium. It extends from rostral to pharyngeal. A genetically determined and timed regular separation of nasal and oral epithelium and an initial epithelial shift along these differentiation boundaries allow the well-timed "primary" fusion of mesenchyme. A great oral part of the contact-epithelium in the thick primordia of the human hard

Studies on the familial character of endemic Balkan nephropathy. Possible role of the toxic hydric factor in the determination of "familial agglomerations" in endemic Balkan nephropathy.

The study was carried out in an attempt to explain the familial character of endemic Balkan nephropathy and of agglomeration of cases in certain households and in the ascendency of certain families. The hypotheses of a genetic disease or that of an infectious etiology have been taken into consideration in previous studies - by chromosomal analyses and statistical-mathematic tests of contagiousness - without being able to find arguments in support of one or the other hypotheses. This study brings important elements in favour of the possible role of ecologic factors and mainly of the hydrictoxic

Hyperimmunoglobulinemia E in the Waorani, an isolated Amerindian population.

The Waorani Indians of Eastern Ecuador have the highest blood levels of immunoglobulin E that have been recorded in a human population. Using a radial immunodiffusion technique for IgE determination, we found the mean plasma IgE concentration for the entire sample (n = 227) to be 11,975 International Units per milliliter (normal: 5--500 IU/ml). The reason for the elevated IgE concentrations is unclear, although genetic factors and a high prevalence of parasitic infection may be involved. Atopic disease is rare among the Waorani as determined by medical history, physical examination, and immedi

Distribution of Virulence Factors and Resistance Determinants in Three Genotypes of Staphylococcus argenteus Clinical Isolates in Japan.

Staphylococcus argenteus, a novel staphylococcal species independent of S. aureus, causes a wide spectrum of infectious diseases. As detection of this species from humans and animals has been increasingly reported worldwide, its growing virulence and drug resistance via external genetic determinants has become concerning. In this study, the prevalence and genetic characteristics of virulence factors and drug resistance determinants were investigated for 82 S. argenteus clinical isolates in Hokkaido, Japan, for a one-year period starting in August 2019. These S. argenteus isolates corresponded

Integrating survey and molecular approaches to better understand wildlife disease ecology.

Infectious wildlife diseases have enormous global impacts, leading to human pandemics, global biodiversity declines and socio-economic hardship. Understanding how infection persists and is transmitted in wildlife is critical for managing diseases, but our understanding is limited. Our study aim was to better understand how infectious disease persists in wildlife populations by integrating genetics, ecology and epidemiology approaches. Specifically, we aimed to determine whether environmental or host factors were stronger drivers of Salmonella persistence or transmission within a remote and iso

Leukocytes and age-related macular degeneration.

Age-related macular degeneration (AMD) is a leading cause of vision loss. Leukocyte subtypes have been implicated in the pathogenesis, but causality remains uncertain. We investigated whether leukocyte counts are causally associated with clinically diagnosed AMD using observational and genetic analyses in three prospective cohorts. We included 117,714 individuals from the Copenhagen General Population Study (CGPS), 388,758 from the UK Biobank (UKB), and 285,612 from the Million Veteran Program (MVP), all of European ancestry and without clinically diagnosed AMD at baseline. In total, 27,130 in

Schizophrenia Genetic Liability Drives Chronic Disease Risk in Unaffected Individuals through Immune and Metabolic Pathways.

INTRODUCTION: Comorbidity between schizophrenia (SCZ) and chronic diseases is well documented. However, it remains unknown whether unaffected individuals with elevated SCZ genetic liability also face increased risk, and what biological pathways may underlie these associations. METHODS: We analyzed 426,237 UK Biobank participants without SCZ to test associations between SCZ polygenic risk score (SCZ-PRS) and 24 chronic diseases using logistic regression. Multi-omics mediation analysis incorporated seven inflammatory markers, 1,463 plasma proteins, and 250 circulating metabolites to delineate i

Evidence for genetic correlation between appendix and inflammatory bowel disease: A bidirectional Mendelian randomization study.

BACKGROUND: Observational studies highlighted a link between the appendix and inflammatory bowel disease (IBD), but proving causality has been difficult due to the lack of a clear temporal sequence. METHODS: This research used a two-sample bidirectional univariable Mendelian randomization (MR), multivariable MR and linkage disequilibrium score regression (LDSC) analyses to explore the relationship between acute appendicitis, appendectomy, and IBD. Eligible instrumental variables were screened from previous genome-wide association studies (GWAS) of European ancestry for analysis. The inverse v

Molecular Surveillance of Malaria in Nigeria Reveals a Low Frequency of Antimalarial Resistance Markers and Clonal Expansion of Chloroquine-Sensitive Infections.

BACKGROUND: The success of malaria elimination in sub-Saharan Africa rests largely on the sustained efficacies of the chemointerventions used in malaria treatment and chemoprophylaxis in the highest-burden countries such as Nigeria. Data on the impact of these interventions can guide policy for sustained malaria control toward elimination. We report the complexity of infection (number of genetically distinct same-species parasite strains observed in an infection) and genetic diversity of Plasmodium falciparum in Nigeria and their antimalarial resistance profiles. METHODS: We used targeted amp
