Glucosylated cholesterol accumulates in atherosclerotic lesions and impacts macrophage immune response.

Atherosclerosis can be described as a local acquired lysosomal storage disorder (LSD), resulting from the build-up of undegraded material in lysosomes. Atherosclerotic foam cells accumulate cholesterol (Chol) and glycosphingolipids (GSLs) within lysosomes. This constitutes the ideal milieu for the formation of a side product of lysosomal storage: glucosylated cholesterol (GlcChol), previously found in several LSDs. Using LC-MS/MS, we demonstrated that GlcChol is abundant in atherosclerotic lesions. Patients suffering from cardiovascular diseases presented unaltered plasma GlcChol levels but sl…

Microbiota-innate immune crosstalk drives atherosclerosis: mechanisms, disease progression, and emerging therapeutic strategies.

Atherosclerosis (AS) is a complex cardiovascular disease driven by the interplay of dysregulated lipid metabolism, chronic inflammation, and immune dysfunction. Increasing evidence has revealed that the gut microbiota not only regulates host metabolic homeostasis but also actively contributes to the initiation and progression of AS through intricate interactions with the innate immune system. Microbial-derived signaling molecules, including lipopolysaccharides, outer membrane vesicles, extracellular nucleic acids, and TMAO, can activate Toll-like receptors, the NLRP3 inflammasome, and nucleic

Novel Foamed Magnesium Phosphate Antimicrobial Bone Cement for Bone Augmentation.

In dental implant surgery, infection is identified as the primary factor contributing to the failure of bone grafts. There is an urgent need to develop bone graft materials possessing antibacterial characteristics to facilitate bone regeneration. Magnesium phosphate bone cement (MPC) is highly desirable for bone regeneration due to its favorable biocompatibility, plasticity, and osteogenic capabilities. However, the limited porosity of conventional MPC hinders the nutrient supply, gas diffusion, and cell infiltration, thereby compromising its osteogenic efficacy. This research focused on the f

miR-146a contributes to atherosclerotic plaque stability by regulating the expression of TRAF6 and IRAK-1.

BACKGROUND: Atherosclerosis is a chronic inflammatory disease. The vulnerable plaque of atherosclerotic can lead to the development of many … AF6 expression. CONCLUSIONS: We display for the first time that miR-146a inhibits the formation of foam cells, RAW264.7 macrophage apoptosis and pro-inflammatory reaction through negative regulation of IRAK1 and TRAF6 expression, thereby enhancing the stability of atherosclerotic plaques.

Probing Inflammasome Activation in Atherosclerosis.

Atherosclerosis is driven by chronic inflammation in all stages of the disease. Inflammation is fueled by elevated levels of pro-inflammatory cytokines. Interleukins (IL) are cytokines of particular importance in atherosclerosis, due to their key involvement in various pro-atherogenic processes, including infiltration of immune cells to the lesion, stimulation of the production of other pro-inflammatory mediators by other sources, and generation of lipid laden foam cells, all of which contribute to plaque development and progression. Various stimuli that are abundant in atherosclerotic plaques…

Reaction-diffusion model of atherosclerosis development.

Atherosclerosis begins as an inflammation in blood vessel walls (intima). The inflammatory response of the organism leads to the recruitment of monocytes. Trapped in the intima, they differentiate into macrophages and foam cells leading to the production of inflammatory cytokines and further recruitment of white blood cells. This self-accelerating process, strongly influenced by low-density lipoproteins (cholesterol), results in a dramatic increase of the width of blood vessel walls, formation of an atherosclerotic plaque and, possibly, of its rupture. We suggest a 2D mathematical model of the…

[Gastric xanthelasma (author's transl)].

A report is given about 42 cases with xanthelasma of the gastric mucosa diagnosed with 5 1/2 years. The condition was found (considering the total number of gastroscopies) in men 1.38 times as often as in women; the average age of all patients at the time of diagnosis was 62.5 years. Xanthelasma lesions are yellow or yellowish-white spots in the mucosa, consisting of foam-cells microscopically. The cytoplasma stains pale pink in a HE preparation; there is almost no staining with PAS, and sudan black staining is definitely positive in most cases. PAS and sudan black staining do allow differenti

Case Report: Pulmonary artery biopsy findings in a patient with a BMPR2 variant-associated pulmonary arterial hypertension.

Mutations in BMPR2 are the most common genetic cause of heritable pulmonary arterial hypertension (PAH). We report a 47-years-old male with a BMPR2 variant (c.246A > G) who presented with dyspnea and recurrent hemoptysis. Right heart catheterization confirmed PAH with a mean PAP of 46 mmHg, PAWP of 3 mmHg, and PVR of 10.75 WU. A pulmonary artery biopsy was performed to exclude alternative diagnoses such as vasculitis and chronic thromboembolism. Histology showed foam cell accumulation in the intima, infiltration of CD3+ T cells, CD20+ B cells, and CD68+ histiocytes in the intima and media, and

S-Nitrosylation: Mechanistic Links between Nitric Oxide Signaling and Atherosclerosis.

PURPOSE OF REVIEW: This review explores current knowledge on the beneficial and detrimental roles of nitric oxide (NO) in vascular biology, with a particular focus on the emerging role of protein S-nitrosylation in the pathophysiology of atherosclerosis. RECENT FINDINGS: Major risk factors for atherosclerosis include hypercholesterolemia, low-density lipoprotein (LDL) oxidation, hyperglycemia, hyperhomocysteinemia, chronic inflammation, and obesity. Recent studies have shown that protein S-nitrosylation interacts with these risk factors, influencing atherogenesis either by promoting or inhibi

A case report of palmar xanthoma with xanthomatous neuropathy.

Xanthomas are plaques or nodules consisting of an accumulation of excess lipids, resulting in the formation of foam cells in the skin or tendons. Typically, xanthomas are not accompanied by other symptoms. Here, we report a patient with a presentation of painful palmar xanthomas and subsequent diagnosis of metabolic and cardiovascular morbidities. A 44-year-old man presented with multiple yellowish, firm, and painful nodules on his right palm and fingers. Lipid panel and medical examination revealed a diagnosis of type III hyperlipoproteinemia and diabetes mellitus type II. Histopathological e…

Fabrication and characterization of Ag- and Ga-doped mesoporous glass-coated scaffolds based on natural marine sponges with improved mechanical properties.

Natural marine sponges were used as sacrificial template for the fabrication of bioactive glass-based scaffolds. After sintering at 1050°C, the resulting samples were additionally coated with a silicate solution containing biologically active ions (Ag and Ga), well-known for their antibacterial properties. The produced scaffolds were characterized by superior mechanical properties (maximum compressive strength of 4 MPa) and total porosity of ~80% in comparison to standard scaffolds made by using PU foam templates. Direct cell culture tests performed on the uncoated and coated samples showed po

The surgical pathology of pulmonary infarcts: diagnostic confusion with granulomatous disease, vasculitis, and neoplasia.

Twenty-three cases of surgically resected pulmonary infarcts sent in consultation were reviewed to evaluate their morphology and to assess reasons for consultation. The morphology of these infarcts demonstrated that only a minority had the classical triangular shape at low magnification (26%) whereas the majority were either spherical (17%) or had a geographic pattern of necrosis (35%). The margin of the infarcted tissue often had a pseudogranulomatous appearance due to palisaded histiocytes, foam cells, or perpendicularly oriented proliferations of fibroblasts and myofibroblasts (74%) and occ…

Regulated expression of the human acetylated low density lipoprotein receptor gene and isolation of promoter sequences.

The acetylated low density lipoprotein (AcLDL) receptor is expressed on tissue macrophages after their differentiation from monocyte precursors and has been proposed to play a role in the generation of foam cells in atherosclerotic lesions. In the present studies, THP-1 human monocytic leukemia cells were used to investigate mechanisms responsible for expression of the AcLDL receptor gene after treatment with phorbol 12-myristate 13-acetate (TPA). TPA-dependent accumulation of AcLDL receptor mRNA was not detected until after a lag phase of 12 hr and was blocked by concurrent treatment with cyc…
