Modulation of tetracycline-phospholipid interactions by tuning of pH at the water-air interface.

This paper is part of a systematic study of the interactions of tetracycline antibiotics with phospholipid monolayers at the water-air interface. Tetracyclines are widespread antibiotics that undergo a series of protonation equilibria in solution, depending on the pH. The surface activity of tetracyclines was determined by means of surface tension measurements for three different systems, i.e. water, TRIS and McIlvaine-EDTA buffer. Surface pressure-molecular area and surface potential-molecular area isotherms were acquired for dipalmitoylphosphatidic acid monolayers on TRIS buffer (pH=7.0) and

Antifouling blood purification membrane composed of cellulose acetate and phospholipid polymer.

The ideal surface of an artificial blood purification membrane needs hemocompatibility and durability of high performance; it should not adsorb any proteins or cells but should still have high permeability in the desired range of solute size. To improve the anti-fouling property of cellulose acetate (CA) membranes, a CA membrane blended with poly(2-methacryloyloxyethyl phosphorylcholine (MPC)-co-n-butyl methacrylate (BMA)) (PMB30) was designed as a blood purification membrane. The polymer solutions for preparing the membrane were prepared using a solvent mixture composed of N, N-dimethylformam

The interaction of trichosanthin with supported phospholipid membranes studied by surface plasmon resonance.

Trichosanthin (TCS) is a toxic protein isolated from a Chinese herbal medicine, the root tuber of Trichosanthes kirilowii Maximowicz of the Curcurbitaceae family. It is now used in China to terminate early and mid-trimester pregnancies. The ribosome inactivating property is thought to be account for its toxicity; it can inactivate the eukaryotic ribosome through its RNA N-glycosidase activity. The interactions of TCS with biological membrane is thought to be essential for its physiological effect, for it must get across the membrane before it can enter the cytoplasm and exert its RIP function.

Hereditary spherocytosis and elliptocytosis erythrocytes show a normal transbilayer phospholipid distribution.

Phosphatidylserine (PS) asymmetry was determined in red blood cells from patients with hereditary spherocytosis and elliptocytosis. No PS-exposing subpopulations were detected using the very sensitive method with fluorescently labeled annexin V. Treatment with N-ethylmaleimide or adenosine triphosphate (ATP) depletion to inactivate the flipase did not lead to formation of PS-exposing subpopulations in these cells, but elevated intracellular calcium levels did lead to extensive scrambling of the PS asymmetry. Although interactions of the membrane skeleton with the phospholipid bilayer have been

Order in phospholipid Langmuir-Blodgett layers and the effect of the electrical potential of the substrate.

The ordering in dipalmitoylphosphatidylcholine (DPPC) Langmuir-Blodgett monolayers and bilayers on a semiconducting indium tin oxide (ITO) s … nset of hydrogen evolution at the substrate surface. For monolayers on hydrophobized ITO, where the phospholipids are initially with their tails directed toward the surface, the changes are more significant. At the highest positive potential applied, the derived order parameters indicate that nearly all probes are flat on the surface. This can be understood as …

Effects of molecular structures on the olfactory responses of phospholipid membranes to four alcohols.

In order to understand the relationship between phospholipid molecular structures and their olfactory responses to odorants, we designed and synthesized four phosphatidylcholine analogues with different long hydrocarbon (CH) chains and selected three natural phospholipids with different head-groups. By using interdigital electrodes (IEs) as olfactory sensors (OSs), we measured the responses of the IEs coated with these seven different lipid membranes to four alcohol vapors in a gas flow system. The IEs voltage changes were recorded and the voltage-relative saturate vapor pressure (V-P/P degree…

Is the increase in renal papillary phospholipid biosynthesis a protective mechanism against injury?

Mercuric chloride (HgCl2) is a well-known renal toxic that causes acute renal failure. The effect of HgCl2 treatment and the protection by thyroxine were studied in rat renal papilla (P), outer medullary inner stripe (OMIS), outer medullary outer stripe (OMOS) and cortical phospholipids (PhLs). HgCl2 brought about an increase in the total phospholipid content in P and OMIS but a drop in OMOS and cortex. Only phosphatidylcholine (PtdCho) and phosphatidylethanolamine (PtdEtn) accounted for such changes. Thyroxine, injected on HgCl2-treated rats, partially reversed the effect of the toxic metal i…

The putative role of cytokines in the induction of primary anti-phospholipid syndrome in mice.

Antiphospholipid syndrome (APLS) is characterized by thrombocytopenia, thromboembolic phenomena and recurrent fetal loss, associated with anti-cardiolipin antibodies (ACA) and/or lupus anticoagulant. The syndrome may be primary or may be associated with other conditions such as systemic lupus erythematosus (SLE). In this study we induced primary APLS following immunization of BALB/c mice with a human monoclonal ACA (H-3). Analysis of the cytokine profile of the mice with experimental APLS indicated low production of IL-2, IL-3 and granulocyte-macrophage colony-stimulating factor (GM-CSF) by co

Alterations of phospholipid metabolites in postmortem brain from patients with Alzheimer's disease.

To test the hypothesis that brain cell membranes degenerate in Alzheimer's disease (AD), we measured the levels of phospholipids, their water-soluble metabolites, and glycerophosphocholine (GPC) cholinephosphodiesterase activity in postmortem brain tissue from patients with AD and age-matched controls. We found significantly higher levels of the phospholipid catabolite GPC in AD brain. In contrast, choline and ethanolamine levels were significantly lower in AD, and phospholipid levels were slightly decreased. Furthermore, in AD the activity of the GPC-degrading enzyme GPC cholinephosphodiester…

Calcium-activated, phospholipid-dependent protein kinase activity in calf platelets.

Protein kinase C (PKC) has been widely studied from different tissues of mammals. Human platelets display higher levels of PKC activity, if compared with other sources. The PKC activity from calf platelets crude extract was determined in the presence of various protease inhibitors such as PMSF, Leupeptin or Trypsin inhibitro, and the Ca(2+)-chelators EGTA and EDTA. The free calcium requirement was 0.25 mM, calculated with the help of the Solgas-water computer program, which represents 1 mM CaCl2, in these assay conditions. Optimum PKC activity was obtained at 4 min in the presence of PS plus D

X-ray and fluorescence studies on phospholipid bilayers. IX. Interactions with pentachlorophenol.

Pentachlorophenol (PCP) is a widely used and highly toxic fungicide. Its toxicity is mainly expressed at the cell membrane level. It is, therefore, of interest to test its ability to alter the lipid bilayer organization. The present study was performed by X-ray diffraction techniques on dimyristoylphosphatidylethanolamine (DMPE) and dimyristoylphosphatidylcholine (DMPC) bilayers and by fluorescence on DMPC liposomes. These two phospholipids are respectively found at the inner and outer monolayers of human erythrocyte membranes. Each type of phospholipid was made to interact with different conc…

Prevention of oxidative injury to cardiac phospholipid by membrane-active "stabilizing agents".

Three lipophilic, membrane-active "stabilizing agents," cepharanthine, chlorpromazine, and trifluoperazine, were found to protect myocardial membrane phospholipid from peroxidative injury. The compounds prevented, in a concentration-dependent manner, the cardiac phospholipid peroxidation which resulted from lipid exposure to superoxide-dependent, iron-promoted oxygen-radical chemistry of the type thought to be a causative factor in ischemic-reperfusion tissue damage. Chlorpromazine's antiperoxidant IC50 (i.e., concentration at which peroxidation was inhibited by 50%) was 180 microM; the antipe

Successful imaging of human cancer with indium-111-labeled phospholipid vesicles.

Twenty-four patients with proven primary and/or metastatic cancer received single intravenous injections of phospholipid vesicles containing 0.5 mCi of Indium-111. Gamma camera scintigraphy 1 to 72 hours later visualized tumors in 22 patients (92%), including carcinomas of breast, lung, colon, prostate, kidney, cervix, thyroid, and soft tissue sarcoma, lymphoma, and melanoma. Tumor sites that were identified included soft tissues, bone, lung, liver, lymph node, and spinal cord. There were only two false-positive images in metastatic sites and four false-negative images in metastatic sites. Ove

Phospholipid/Ca2+-dependent protein kinase activity in human parathyroid adenoma.

We have examined the activities of phospholipid/Ca2+-dependent and cyclic AMP-dependent protein kinases of the parathyroid adenomas and the atrophic glands which were resected from three patients with primary hyperparathyroidism. Phospholipid/Ca2+-dependent protein kinase activity of atrophic parathyroid gland was exclusively present in cytosol fraction (90.7 +/- 12.3%). On the other hand, phospholipid/Ca2+-dependent protein kinase activity of parathyroid adenomas was 66.9 +/- 6.4% in cytosol and 33.1 +/- 6.4% in membrane fraction, suggesting a translocation of the enzyme from the cytosol to t

[Energy profile of a channel formed by alpha-latrotoxin in a bilayer phospholipid membrane].

Current-voltage characteristics are obtained for a channel formed by alpha-latrotoxin when inserting into a bilayer lipid membrane separating solutions of different ionic composition. They are used for determining parameters of a two-barrier model of this channel energy profile. It is shown that selectivity of these channels is based on the same principles that in the calcium channels of biological membranes and is mainly determined by the ion binding inside the channel. Affinity of the channel for the penetrating ions of alkali-earth metals decreases in the sequence: Mg2+ greater than Ca2+ gr

Phospholipid composition and membrane function in phosphatidylserine decarboxylase mutants of Escherichia coli.

Temperature-sensitive conditional lethal mutants in phosphatidylserine decarboxylase (psd) accumulate large amounts of phosphatidylserine under nonpermissive conditions (42 degrees C) prior to cell death. In addition, the ratio of cardiolipin to phosphatidylglycerol is increased. At an intermediate temperature (37 degrees C), high levels of phosphatidylserine can be maintained with little effect on cell growth or viability. Under these conditions, both the rate of induction and the function of the lactose transport system are normal. At 42 degrees C addition of Mg2+ or Ca2+ to mutant cultures

Phospholipid metabolism during renal regeneration after acute tubular necrosis.

Renal function, structure, and membrane metabolism were studied during regeneration of proximal tubular cells in rats. A reversible syndrome … the synthesis of renal choline-containing phospholipid. The rate of [14C]choline incorporation into phospholipids in inner cortical slices was lowest 1 day after mercury administration, then increased constantly for the next 4 days to reach a maximal value 104% above control. The rate declined slowly for the next 11 days and returned to normal by 28 days. The i …
