The incorporation of the concept of minimum RBE (RbEmin) into the linear-quadratic model and the potential for improved radiobiological analysis of high-LET treatments.

PURPOSE: The formulation of relative biological effectiveness (RBE) for high linear energy transfer (high-LET) radiation treatments is revisited. The effects of changed production of sub-lethal damage with varying LET is now considered via the RBEmin concept, where RBEmin represents the lower limit to which RBE tends at high doses per fraction. MATERIALS AND METHODS: An existing linear-quadratic formulation for calculating RBE variations with fractional dose for high-LET radiations is modified to incorporate the twin concepts of RBEmax (which represents the value of RBE at an effective dose-p

The radiosensitivity of human fibroblast cell lines correlates with residual levels of DNA double-strand breaks.

PURPOSE: To study the correlation of residual DNA double-strand breakage after irradiation and cellular radiosensitivity in cells showing marked differences in radiosensitivity. MATERIALS AND METHODS: The levels of DNA double-strand breaks remaining at 4 h after irradiation were measured by graded-voltage gel electrophoresis in fibroblast cell strains derived from seven individuals either with normal radiosensitivity (n = 2), or with genetic abnormalities known to show increased (two ataxia telangiectasia, one scid) or possibly decreased (two Li-Fraumeni family members) sensitivity. RESULTS:

Postmastectomy hypofractionated and accelerated radiation therapy with (and without) subcutaneous amifostine cytoprotection.

PURPOSE: Postmastectomy radiation therapy (PMRT) provides major local control and survival benefits. More aggressive radiation therapy schemes may, however, be necessary in specific subgroups, provided they are safely administered. We report the tolerance and efficacy of a highly accelerated and hypofractionated regimen (HypoARC). METHODS AND MATERIALS: One hundred twelve high-risk patients who had undergone mastectomy received 10 consecutive fractions of 3.5 Gy in 12 days (thoracic wall and axillary/supraclavicular areas). Two consecutive additional fractions of 4 Gy were given to the surgic

An unexpected role for leucyl aminopeptidase in UV tolerance revealed by a genome-wide fitness assessment in a model cyanobacterium.

UV radiation (UVR) has significant physiological effects on organisms living at or near the Earth's surface, yet the full suite of genes required for fitness of a photosynthetic organism in a UVR-rich environment remains unknown. This study reports a genome-wide fitness assessment of the genes that affect UVR tolerance under environmentally relevant UVR dosages in the model cyanobacterium Synechococcus elongatus PCC 7942. Our results highlight the importance of specific genes that encode proteins involved in DNA repair, glutathione synthesis, and the assembly and maintenance of photosystem II,

Delayed effects of prenatal low-dose irradiation in the white matter of the rat brain.

PURPOSE: This study is part of a general search for the anatomical bases of the severe mental retardation syndrome caused by prenatal irradiation. More specifically, it seeks reasons for the high radiosensitivity of a white matter area, the cingulum of the corpus callosum. MATERIALS AND METHODS: Pregnant primiparous Wistar rats were exposed to X-rays at 12, 13, 14 or 15 days of gestation (E12, E13, E14 or E15) with single low doses of 180 mGy. A high dose of 500 mGy was given at E15 for a complementary study. The brains of the female offspring were collected at 1 and 3 months of age. The affi

Clinical data for radiation embryology. Investigation programme 1967, report 1984.

In 1967 a prospective clinical study was started with the aim of providing optimum counselling for married couples who desire continuation of a pregnancy despite radiation exposure at an early stage as a result of X-ray diagnostics. Recommendations were devised as to whether an interruption of pregnancy should be applied for or not. These results were discussed repeatedly. Within the framework of the prospective study, embryological examinations were made in cases of interruption of pregnancy, and clinical and genetic examinations in cases of children who had been subject to radiation stress i

Diverting loop colostomy for the treatment of refractory gastrointestinal bleeding secondary to radiation proctitis.

Chronic radiation proctitis is an uncommon and serious complication of radiotherapy for pelvic malignancies. It has been suggested that fecal stream diversion decreases bowel irritation, resulting in decreased rectal bleeding. We report on the outcome of patients after a diverting loop colostomy for the treatment of severe radiation proctitis. All consecutive patients with chronic proctitis treated with diverting loop colostomy for bleeding at one institution between October 1991 and June 1999 were studied. Four patients presented with rectal bleeding and diarrhea, and two patients had only bl

Ultraviolet-B protection of ascorbate and tocopherol in plants related with their function on the stability on carotenoid and phenylpropanoid compounds.

Ascorbate and tocopherol are important hydrophilic or lipophilic antioxidants in plants, while their crucial roles in the antioxidant defense system under ultraviolet B radiation were not well understood. The mutants of Arabidopsis thaliana deficient in ascorbate (vtc1 and vtc2) or tocopherol (vte1) were used to analyze their physiological, biochemical and metabolic change in responses to Ultraviolet B radiation. Results showed that loss of either ascorbate or tocopherol caused reduction in phenylpropanoid and flavonol glycosides compounds, as well as reduction in superoxide dismutase activity

Hypofractionated gamma knife radiosurgery for large cavernous sinus hemangiomas: Outcomes of a case series with a review of literature.

Large cavernous sinus hemangiomas (CSHs) present a significant therapeutic challenge. While microsurgery carries risks of massive bleeding and morbidity, standard single-fraction stereotactic radiosurgery is often strictly limited by the radiation tolerance of the adjacent optic apparatus. Data regarding hypofractionated Gamma Knife radiosurgery (HfGKRS) for these lesions remain scarce. This study evaluates the safety and efficacy of a uniform HfGKRS protocol for large CSHs. This retrospective, single-center study analyzed four consecutive patients with large CSHs (median target volume: 25.85 …

Validating nasogastric tube placement with pH testing: A randomized controlled trial protocol.

BACKGROUND: Enteral nutrition (EN) is preferred when oral feeding is not possible. The use of the Nasogastric Tube (NGT) ensures rapid and low-risk nutrient administration. However, confirming the placement through chest radiography, besides delaying the initiation of nutritional therapy, exposes patients to radiation. The pH test of gastric aspirate provides a quicker check for NGT placement, but its reliability is compromised by challenges related to aspirating gastric secretions. STUDY OBJECTIVE: The main objective of this study is to assess the high-performance placement of NGTs for nutri

Magnetic resonance imaging: an accurate, radiation-free, alternative to computed tomography for the primary imaging and three-dimensional reconstruction of the bony orbit.

PURPOSE: To determine the extent to which the accuracy of magnetic resonance imaging (MRI) based virtual 3-dimensional (3D) models of the intact orbit can approach that of the gold standard, computed tomography (CT) based models. The goal was to determine whether MRI is a viable alternative to CT scans in patients with isolated orbital fractures and penetrating eye injuries, pediatric patients, and patients requiring multiple scans in whom radiation exposure is ideally limited. MATERIALS AND METHODS: Patients who presented with unilateral orbital fractures to the Royal Brisbane and Women's Ho

Radiation therapy-activated nanoparticle and immunotherapy: The next milestone in oncology?

Radiotherapy (RT) is a fundamental treatment at the locoregional or oligometastatic stages of cancer. In various tumors, RT effects may be optimized using synergistic combinations that enhance tumor response. Innovative strategies have been designed that explore the radiation mechanisms, at the physical, chemical and biological levels, to propose precision RT approaches. They consist in combining RT with immunotherapy to revert radiation immunosuppressive effects or to enhance radiation-induced immune defenses against the tumor to favor immunogenic cell death. Radiotherapy-activated nanopartic

Acclimation and Compensating Metabolite Responses to UV-B Radiation in Natural and Transgenic Populus spp. Defective in Lignin Biosynthesis.

Plants have evolved to protect leaf mesophyll tissue from damage caused by UV-B radiation by producing an array of UV-absorbing secondary metabolites. Flavonoids (phenolic glycosides) and sinapate esters (hydroxycinnamates) have been implicated as UV-B protective compounds because of the accumulation in the leaf epidermis and the strong absorption in the wavelengths corresponding to UV. Environmental adaptations by plants also generate a suite of responses for protection against damage caused by UV-B radiation, with plants from high elevations or low latitudes generally displaying greater adap

Vaginal dilator therapy for pelvic cancer patients: a review.

Vulvovaginal atrophy affects up to 84% of postmenopausal women and is even more prevalent among cancer survivors. Hypoestrogenism leads to thinning of vaginal epithelium, decreased elasticity, reduced lubrication, and increased tissue fragility, resulting in symptoms such as vaginal dryness, irritation, dyspareunia, and pain with pelvic examination. Cancer treatments-including chemotherapy, endocrine therapy, pelvic radiation, and gynecologic surgery-can induce abrupt estrogen deficiency and accelerate vulvovaginal atrophy beyond what seen in natural menopause. Pelvic radiation in particular i

Clinical applicability of biologically effective dose calculation for spinal cord in fractionated spine stereotactic body radiation therapy.

BACKGROUND: The aim of the study was to investigate whether biologically effective dose (BED) based on linear-quadratic model can be used to estimate spinal cord tolerance dose in spine stereotactic body radiation therapy (SBRT) delivered in 4 or more fractions. PATIENTS AND METHODS: Sixty-three metastatic spinal lesions in 47 patients were retrospectively evaluated. The most frequently prescribed dose was 36 Gy in 4 fractions. In planning, we tried to limit the maximum dose to the spinal cord or cauda equina less than 50% of prescription or 45 Gy2/2. BED was calculated using maximum point do

2-deoxy-D-glucose causes cytotoxicity, oxidative stress, and radiosensitization in pancreatic cancer.

Glucose metabolism as assessed by (18)FDG PET imaging provides prognostic information in patients with pancreatic cancer but the implications of manipulating glucose metabolism for therapeutic purposes are unknown. Based on previous results with other cancer cell types, we hypothesized that inhibition of glucose metabolism in pancreatic cancer cells would cause cell killing via oxidative stress resulting from disruptions in thiol metabolism. 2-Deoxy-D-glucose (2DG), a chemical inhibitor of glucose metabolism, and glucose deprivation induced cytotoxicity in human pancreatic cancer cells in a ti

Single-fraction high-dose palliative radiotherapy for facial cutaneous squamous cell carcinoma: a case report.

BACKGROUND: Cutaneous squamous cell carcinoma (cSCC) is a common malignancy affecting the skin, and its incidence increases with age; as such, it disproportionately affects the elderly. It is especially difficult to treat advanced skin cancers in elderly patients with dementia, who may not tolerate radiotherapy. CASE DESCRIPTION: A 100-year-old woman with advanced dementia was referred to dermatology for a large cSCC involving the nasal bridge and abutting the bilateral orbits. She was initially deemed a poor candidate for surgical resection, but over time the tumor grew and became increasing

Paget's disease of scrotum and penis case report of a re-irradiation and review of the literature.

Extramammary Paget's disease (EMPD) is a rare cutaneous adenocarcinoma generally arising in the anogenital region. Surgery is still considered the treatment of choice for patients with EMPD, while Radiotherapy is a common alternative for inoperable cases and it's necessary in case of lack of surgical radicality. In this article, we described our experience and a review of the literature, with a particular focus on radiation-induced toxicity and on the feasibility of re-irradiation. A 70-year-old patient with EPMD underwent adjuvant radiotherapy in 2015. After 28 months for recurrence another r
