Progress toward poliomyelitis eradication--south east Asia region, 1988-1994.

Since the adoption of the poliomyelitis eradication initiative by the World Health Organization (WHO) in 1988, substantial progress toward the eradication of polio has been achieved in the Region of the Americas and in the Western Pacific Region of WHO (1-3). A major step toward global eradication was made in 1994, when polio eradication activities--specifically the implementation of biannual National Immunization Days (NIDs)--were accelerated in the member countries of the South East Asia Region (SEAR) of WHO. In August 1994, Thailand became the first SEAR country to conduct NIDs; by February…

[The impact of a new strict obstetrical strategy on maternal mortality and obstetrical results].

In an attempt to reduce the caesarean section rate without an adverse effect on the obstetrical outcome, new guidelines for the management o … ring the latter period use of oxytocin increased from 3.4 to 17.4%. These findings suggest that the adoption of strict guidelines can lead to a decrease of the caesarean section rate and an improvement of the obstetrical outcome in the absence of new technology.

Urological problems in the management of quadriplegic women.

During the past 25 years enormous progress has been made in the management of the neuropathic bladder, largely as a result of the adoption of intermittent catheterisation, together with improvements in catheter-related techniques, and the judicious use of antibacterial drugs and sphincter surgery. A few quadriplegic women can be trained to do self-intermittent catheterisation, using a special technique. For the majority of these women, however, there is no practical alternative at present to indwelling catheters. Bladder spasms resulting from the mechanical stimulation of the catheter and/or r…

Immunophenotyping of Cellular Therapy Products Against Oncogenic and Viral Targets.

Flow cytometric analysis can provide valuable information on the viability, phenotype, and therapeutic potential of cell therapy products using a minimal amount of cell numbers. In this chapter, we describe methods for the analysis of clinical products that incorporate extra considerations within the protocols to help maintain consistency and confidence of staining. This can include methods for evaluating the transduction efficiency for T cells expressing chimeric antigen receptors (CAR-T), characterizing the frequency of diverse lymphocyte populations within polyclonal cell cultures and posti

Resistance mechanisms and translational bottlenecks in CAR-T cell therapy for pediatric acute lymphoblastic leukemia: a perspective.

Chimeric antigen receptor (CAR)-T cell therapy has achieved remarkable remission rates exceeding 80% in relapsed/refractory pediatric B-cell acute lymphoblastic leukemia (B-ALL), yet 40-50% of patients eventually relapse within the first year. This therapeutic ceiling is not a single failure mode but the convergence of at least four interrelated layers: loss of CAR-T persistence, antigen escape driven by CD19 splicing variants and lineage switch, CAR-T cell-intrinsic exhaustion from chronic tonic signaling, and an immunosuppressive bone marrow microenvironment that actively extinguishes effect

Cost-Effectiveness of AI-Powered Decentralized Production for Advanced Therapies in the Hospital-Generated Chimeric Antigen Receptor T-Cell Therapy for Multiple Myeloma From a German Hospital Perspective: Early Health Technology Assessment Study.

BACKGROUND: Although effective, current chimeric antigen receptor T-cell (CAR-T) production methods-centralized, manual, and complex-are cost-intensive, time-consuming, and prone to variability. AIDPATH (AI-Powered Decentralized Production for Advanced Therapies in the Hospital) proposes a decentralized, automated alternative that integrates patient-specific data; optimizes resource use; and potentially improves cell viability, manufacturing efficiency, and patient outcomes. OBJECTIVE: The aim of this study was to compare CAR-T therapy produced by the AIDPATH platform to both ciltacabtagene a

Microbiome and metabolite biomarkers of CAR T-cell therapy outcomes in relapsed/refractory diffuse large B-cell lymphoma.

CD19 chimeric antigen receptor (CAR) T-cell therapy has revolutionized treatment for relapsed/refractory diffuse large B-cell lymphoma (R/R-DLBCL), but challenges such as posttreatment failure and immune-related adverse events (AEs) persist. This study explores the gut microbiome as a predictive biomarker for CAR T-cell therapy outcomes and toxicity. Stool and serum samples from patients with R/R-DLBCL were analyzed at apheresis (47 samples) and 1 month after infusion (32 samples) using whole-genome sequencing metagenomics. When compared with healthy controls and newly-diagnosed DLBCL, R/R-DLB

Cognitive Recovery Following Immune Effector Cell-Associated Neurotoxicity Syndrome in the First 6 Months after Chimeric Antigen Receptor T-Cell Therapy.

Chimeric antigen receptor T-cell (CAR-T) therapy is associated with immune effector cell-associated neurotoxicity syndrome (ICANS), which can manifest as acute and sub-acute cognitive dysfunction. Few studies have investigated cognitive recovery post-CAR-T therapy. In this prospective longitudinal study, patients treated with CAR-T therapy at The Alfred Hospital, Melbourne, underwent cognitive testing at preinfusion and 3- and 6-months postinfusion. Psychometric tests measured attention, processing speed, visuospatial function, language, memory, and executive function. 197 cognitive evaluation

Emerging immune checkpoint targets and combination strategies in blood cancer immunotherapy.

Immunotherapy has revolutionized cancer treatment, offering durable responses across various malignancies. However, blood cancers present unique challenges due to significant heterogeneity, an immunosuppressive tumor microenvironment (TME), and potent mechanisms of immune evasion. This review synthesizes the current landscape of immune checkpoint blockade (ICB), evaluating both established agents and emerging targets (e.g., LAG-3, TIM-3, CD47) in hematologic malignancies. We critically examine the role of cancer stem cells (CSCs) in driving therapeutic resistance and recurrence, highlighting h

Supercharging the metabolic engine: mitochondrial engineering strategies for CAR-T persistence in solid tumors.

Chimeric antigen receptor (CAR)-T cell therapy has achieved remarkable success in hematological malignancies, yet its efficacy in solid tumors is severely limited by the metabolically hostile tumor microenvironment (TME). Within this landscape, CAR-T cells undergo rapid functional exhaustion driven by mitochondrial dysfunction and metabolic insufficiency. This mini-review synthesizes emerging mitochondrial engineering strategies designed to restore metabolic fitness and persistence. We first examine the newly identified metabolic-epigenetic axis, where the pathological mitochondrial translocat

A clinical-grade automated platform for the manufacturing of CAR-γδ T cells for immunotherapy.

Gamma delta (γδ) T cells are promising candidates for adoptive immunotherapy due to their unique biological and safety profiles. In particular, ex vivo expanded Vγ9Vδ2 T cells engineered with chimeric antigen receptors (CARs) have gained considerable interest, as they combine potent innate-like antitumor activity with a low risk of graft-versus-host disease and cytokine release syndrome, making them ideal for off-the-shelf cancer immunotherapies. However, as they represent only 1-5% of peripheral T cells, effective activation and expansion protocols are essential to generate sufficient numbers

Chimeric Antigen Receptor T-Cell Therapy for the Treatment of Melanoma: A Systematic Review of Phase One Clinical Trials.

IntroductionMelanoma is a growing health concern, with global incidence increasing annually. However, treatment for refractory disease is currently limited. Chimeric antigen receptor (CAR) T-cell therapy has shown promising results in haematological malignancies. Despite this, its utility in solid cancers, such as melanoma, is not yet established. We performed a systematic review of phase one trials to help determine and describe foundational understandings of the safety and efficacy of CAR T-cell therapy for melanoma.MethodsThis systematic review was conducted in accordance with the Cochrane

Nonarmored GCC-Targeting CAR T-cell Therapy Demonstrates Significant Efficacy in Patients with Advanced Colorectal Cancer.

PURPOSE: This phase I clinical trial aims to evaluate the safety, expansion kinetics, and preliminary efficacy of nonarmored, nanobody-derived guanylyl cyclase 2C (GCC)-targeted chimeric antigen receptor (CAR) T cells in patients with heavily pretreated metastatic colorectal cancer. PATIENTS AND METHODS: In this single-arm, open-label, phase I clinical trial, 24 patients who received at least 2 prior lines of treatment received GCC-targeted CAR T cells across 4 dose levels: 0.5 × 105, 1 × 106, 2 × 106, and 3 × 106 CAR T cells/kg. The primary endpoint was safety; secondary endpoints included a

CAR cellular therapies for acute myeloid leukemia: current landscape and future directions.

Chimeric antigen T cell (CAR-T) therapy has revolutionized the treatment of B-cell malignancies, but has not seen this success replicated in acute myeloid leukemia (AML). Many antigens present on AML cells are shared with healthy hematopoietic progenitors, posing a risk of prohibitive toxicity with CAR-T therapy. Furthermore, clonal evolution of AML, antigen escape and effects of tumor microenvironment (TME) may hinder the efficacy of treatment. A number of innovative approaches to overcome these barriers have been attempted in recent years, including combinatorial and neoantigen targeting, lo

Anti-tumour activity of canine activated T-cells and cytokine-induced killer cells against canine malignant melanoma in vitro.

Adoptive cell transfer (ACT) immunotherapy represents a promising therapeutic approach for cancer treatment, utilising ex vivo-expanded immune effector cells such as T-lymphocytes, natural killer (NK) cells, and cytokine-induced killer (CIK) cells. CIK cells are characterised by dual T-cell and NK cell-like properties, exhibiting high proliferative capacity, cytolytic activity, and non-MHC-restricted tumour recognition. Canine malignant melanoma (CMM) is an aggressive neoplasm with significant translational relevance to human melanoma, yet the cytotoxic potential of CIK cells against CMM remai

Salvage Therapies After Anti-BCMA CAR-T Failure in Patients With Multiple Myeloma: A Meta-Analysis of Response Rates.

Although chimeric antigen receptor (CAR)-T cells targeting B-cell maturation antigen (BCMA) have significantly advanced multiple myeloma (MM) therapy, many patients eventually progress, prompting the search for the most effective salvage regimens. To address this demand, we performed a random-effects meta-analysis evaluating response rates to salvage treatments after anti-BCMA CAR-T failure. We identified 36 eligible studies comprising 476 patients and seven distinct interventions through systematic database screening. Among them, bispecific antibodies (bsAbs) were the most common choice, foll
