Late Complete Endograft Occlusion After Endovascular Aneurysm Repair Successfully Treated With Unilateral Endograft Relining and Femoro-femoral Bypass: A Case Report.

Complete endograft occlusion involving the main body and both limbs after endovascular aneurysm repair (EVAR) is rare, and the optimal treatment remains unclear. We report an 85-year-old male with late complete endograft occlusion after EVAR for an abdominal aortic aneurysm. Computed tomography (CT) showed complete thrombotic occlusion extending from just below the renal arteries to both iliac limbs and distally to the level of the external iliac arteries. A hybrid procedure was performed. After proximal recanalization under right renal artery protection, thrombectomy was performed through the

Mapping structural aging across human tissues reveals tissue-specific trajectories and coordinated deterioration.

Organ structure, including the organization of cells, vasculature and extracellular matrix, underpins its function, yet how structure changes with age remains mostly unknown. Here we developed PathStAR, a framework that quantifies tissue structural aging from routine histopathology images, without being trained to predict chronological age. Applying PathStAR to 25,306 post-mortem biopsies from 40 tissues in 970 donors aged 21-70 years revealed that organ structural aging progresses via distinct, nonlinear temporal trajectories: vascular tissue structural aging accelerates early, uterus and vag

Lactate accumulation drives hepatocellular carcinoma metastasis through facilitating tumor-derived exosome biogenesis by Rab7A lactylation.

Previous studies have demonstrated that lactate accumulation, a common hallmark for metabolic deprivation in solid tumors, could actively drive tumor invasion and metastasis. However, whether lactate influences the biogenesis of tumor-derived exosomes (TDEs), the prerequisite for distant metastasis formation, remains unknown. Here, we demonstrated that extracellular lactate, after taken up by tumor cells via lactate transporter MCT1, drove the release of TDE mainly through facilitating multivesicular body (MVB) trafficking towards plasma membrane instead of lysosome. Mechanistically, lactate p

Quantification of Sympathetic Nerve Traffic to the Heart: Electrophysiological Recording of Cardiac Sympathetic Nerve Activity in a Rodent Model.

The accurate measurement of sympathetic nerve activity is essential for advancing knowledge related to the mechanisms that underpin sympathetic activation in diseased states, such as acute heart failure. Considering sympathetic outflow throughout the body is differentially modulated, the most reliable method of measuring sympathetic traffic to individual organs is by way of direct electrophysiological recording of nerve activity. However, the surgical approach of accessing, exposing, and isolating the sympathetic nerve of interest is technically demanding, especially in the small size of a sta

Breast Reconstruction in a Female Patient With Poland Syndrome.

Poland syndrome (PS) is a congenital anomaly characterized by unilateral hypoplasia or absence of the pectoralis major muscle, often associated with breast asymmetry. Achieving symmetry while preserving the contralateral breast remains a challenge in reconstruction. We present a case of a 19-year-old woman with left-sided PS, a fully developed right breast, and limited autologous soft tissue (body mass index 19.8 kg/m2). A 2-stage approach was performed: initial tissue expansion followed by implant-based reconstruction with fat grafting. A 395-mL microtextured polyurethane implant was placed,

Involvement of substance P/NK1 receptor system in central sensitization in chronic pain.

Chronic pain has become a serious health issue, affecting more than 30% of people worldwide. One of the main mechanisms of chronic pain is central sensitization. It is well known that substance P (SP) and its receptor, Neurokinin 1 receptor (NK1R), play an important role in transmission of nociceptive signals. However, whether SP/NK1R system is involved in central sensitization in chronic pain remains controversial. In the present study, we adopted spared nerve injury (SNI) mouse model to induce neuropathic pain and assessed the role of SP/NK1R system in the development of hyperalgesia and cen

Serotonin regulates nutrient-dependent food intake in Drosophila.

Serotonin is a key neurotransmitter in feeding behavior. However, its role in regulating food intake based on nutrient composition remains unclear. In this study, we utilized the Drosophila model to investigate the influence of serotonin on feeding behavior. By employing transient thermogenetic tools, we transiently manipulated the activity of serotonergic neurons while minimizing effects on other physiological processes, including sleep. By measuring the intake of sucrose, sucralose, yeast, and tryptone, we evaluated the role of serotonin signaling in nutrient-dependent food intake. Our findi

Learning neural dynamics through instructive signals.

Rapid learning is essential for flexible behavior, but its basis in the brain remains unknown. Here we introduce the PRISM plasticity rule, a unifying mechanistic model of three well-established, fast-acting synaptic plasticity rules-in hippocampus, cerebellum and mushroom body-which relies exclusively on pre-synaptic activity and an "instructive signal" from another brain area. Using a multi-region network model we show that guiding PRISM plasticity with instructive signals enables the network to quickly learn extremely flexible nonlinear dynamics underlying behaviorally relevant computations

The role and mechanism of METTL3 in cancer: emerging insights into m6A methylation and therapeutic potential.

N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, plays a crucial role in regulating RNA metabolism and gene expression. Among m6A regulators, METTL3 functions as the catalytic core of the m6A methyltransferase complex and has emerged as a critical epitranscriptomic modulator in cancer. An expanding body of evidence demonstrates that METTL3 is aberrantly expressed in various malignancies, contributing to tumor progression, metastasis, stemness, immune evasion, and resistance to chemotherapy and targeted therapies. Nonetheless, its role remains highly context

A Synthetic Dataset for in Silico Studies of Parameters Influencing Changes in Mean Arterial Blood Pressure During Hemodialysis.

Intradialytic hypotension is a major complication of hemodialysis (HD) and is still incompletely understood. Similarly, intradialytic hypertension is also associated with adverse outcomes and increased mortality and also remains not fully elucidated. Here we present and discuss a synthetic dataset (HD-SIM-MAP) with various cases of dialysis-induced changes in mean arterial blood pressure (MAP) simulated using a lumped-parameter model of the cardiovascular system and the whole-body water and solute kinetics in 5,000 virtual patients with randomly adjusted values of 90 physiological parameters.

Berberine combined with stachyose protects against obesity by modulating gut microbiota and increasing leptin sensitivity.

Leptin resistance is a hallmark of obesity. Recent evidence shows that gut microbiota is associated with the development and progression of leptin resistance. However, whether intervention targeting gut microbiota can improve leptin sensitivity remains to be fully established. In the current investigation, we have demonstrated that co-administration of berberine, an isoquinoline alkaloid compound, and stachyose, a prebiotic, could enhance leptin sensitivity and reduce body weight in diet-induced obesity (DIO) mice, via an effect associated with gut microbiota modulation. Notably, the combinati

AI-driven multi-omics integration for multi-scale predictive modeling of genotype-environment-phenotype relationships.

Despite the wealth of single-cell multi-omics data, it remains challenging to predict the consequences of novel genetic and chemical perturbations in the human body. It requires knowledge of molecular interactions at all biological levels, encompassing disease models and humans. Current machine learning methods primarily establish statistical correlations between genotypes and phenotypes but struggle to identify physiologically significant causal factors, limiting their predictive power. Key challenges in predictive modeling include scarcity of labeled data, generalization across different dom

Wet-Adhesive Hydrogel with Antiswelling Capacity for Strain Sensing.

The development of functional hydrogels with wet adhesion strength and antiswelling properties is crucial for applications ranging from biomedicine to wearable electronics but remains difficult to realize. Herein, we developed a high-performance TA/PAAc/EMA/C-CNT (TPEC) hydrogel via a solvent displacement process. The TPEC hydrogel showed no observable swelling behavior even after 8 days of immersion in a water bath, demonstrating its excellent antiswelling property. The TPEC hydrogel possesses strong adhesive strength in the underwater environment, allowing it to adhere firmly to rigid substr
