Sialic acid-functionalized nanomaterials for targeted cancer therapy, diagnosis, and theranostics.

Targeted cancer therapy, by leveraging the overexpression of specific molecules on tumors, such as sialic acid (SA), offers a promising strategy to enhance antitumor efficacy while minimizing off-target effects. SA modification of nanomaterials can target both tumor cells and tumor-associated immune cells, thereby improving drug delivery and therapeutic outcomes. Tumor cells with high SA expression bind to sialic acid-binding immunoglobulin-like lectins (Siglecs) and selectins expressed on tumor-associated immune cells, potentially contributing to immunosuppression. By binding to these recepto

Amphiphilic Janus nanoparticles for image-guided cancer treatment: cell internalization mechanism, molecular action, challenges, and outlook.

Cancer is recognized as one of the most fatal diseases across the globe. It accounts for approximately 10 million deaths in 2022, while these statistics are even more alarming for low-income countries due to restricted medical facilities for diagnosis and treatment. The application of conventional treatments, including chemotherapy, radiation therapy, and surgery, causes adverse side effects by damaging healthy cells and inducing drug resistance. Therefore, these treatments are increasingly approached with caution by medical practitioners and clinicians, who are actively seeking safer and more

Nanoparticle-based approaches for sepsis treatment: Current trends and perspectives.

Sepsis is a devastating syndrome resulting from organ dysfunction caused by a dysregulated host response to infection. The management of patients is currently primarily based on controlling the infection and restoring cardiovascular/respiratory homeostasis. Unfortunately, except for systemic corticosteroid therapy, no approved immunomodulatory treatment for sepsis exists. However, the ability of nanoparticles to cross biological barriers and deliver therapeutic agents in a targeted manner makes them particularly attractive to hinder a number of mechanisms within sepsis pathology. While histori

Fighting cancer smarter: Using hydrogel delivery systems to target chemokines.

Chemokines organize immune cell locomotion and trafficking in the tumor milieu, exerting dual roles by either boosting antitumor immunity (e.g., CXCL9/10/11 recruit effector T-cells) or promoting tumor progression (e.g., CCL2 supports immunosuppressive myeloid cells). Short half-lives, off-target effects, and tumor microenvironment (TME) barriers such as hypoxia and acidity hinder systemic administration of chemokines. Hydrogel-based delivery systems provide a biocompatible and tunable platform for controlled, localized chemokine release, thereby improving cargo stability and facilitating effe

Advances in therapeutic strategies for acute kidney injury.

Acute Kidney Injury (AKI) is a complex clinical syndrome marked by a rapid decline in renal function, most commonly triggered by ischemia, hypoxia, or nephrotoxic insults, and is associated with significant morbidity and mortality. Despite advances in understanding its underlying mechanisms, current treatments remain predominantly supportive, with no effective therapies available to reverse or repair renal damage. Although biomarker-guided precision medicine holds promise, its clinical utility requires further validation through large-scale trials. This review summarizes recent progress in AKI

Transformative potential of plant-based nanoparticles in cancer diagnosis and treatment: bridging traditional medicine and modern therapy.

Cancer is a primary global health concern, with an estimated 35.3 million cancer cases expected worldwide, representing a 76.6% increase in 2022, and 20 million by 2050, resulting from genetic mutation and environmental factors that cause uncontrolled cell growth. Other factors including smoking, unhealthy diets, physical inactivity, exposure to carcinogens, UV radiation, and aging increase DNA damage. Current cancer treatments like chemotherapy, radiation therapy, immunotherapy, and surgery are effective, but those have significant effects like lack of specificity, development of drug resista

A Novel Retrievable Drug Infusion Stent and Blood-Brain Barrier Portal for Antibody Therapies to the Spinal Cord.

BACKGROUND AND OBJECTIVES: The blood-brain barrier (BBB) is prohibitive to intravascular drug delivery. We hypothesized that isolation of spinal cord perfusion would focus on a higher effective dose while minimizing systemic toxicity and that a histamine portal would increase BBB permeability and facilitate large molecule therapies to the spinal cord. METHODS: A dumbbell-shaped, retrievable drug infusion stentgraft (RDIS) was constructed from nitinol and polytetrafluorethylene. A porcine model under anesthesia underwent percutaneous femoral insertion of the sheathed RDIS and deployment in the

Targeted nanoliposomes for precision rheumatoid arthritis therapy: a review on mechanisms and in vivo potential.

Rheumatoid arthritis (RA) is an inflammatory immune-triggered disease that causes synovitis, cartilage degradation, and joint injury. In nanotechnology, conventional liposomes were extensively investigated for RA. However, they frequently undergo rapid clearance, reducing circulation time and therapeutic efficacy. Additionally, their stability in the bloodstream is often compromised, resulting in premature drug release. The current review explores the potential of targeted liposomal-based nanosystems in the treatment of RA. It highlights the pathophysiology of RA, explores selective targeting

Olfaction and drug delivery to the human olfactory airspace: current challenges and recent advances.

INTRODUCTION: Olfactory function, despite its critical role in human survival and quality of life, is often underappreciated. This could be associated with the fact that symptoms of olfactory dysfunction rarely occur in isolation as they are frequently concomitant with comorbidities. Furthermore, effective treatments for olfactory dysfunction largely remain elusive, and no standardized clinical practice for treating this dysfunction currently exist, thus complicating the initiation of appropriate therapeutic modalities. Intranasal administration of topical medication targeting the olfactory cl

Biological Nanoparticles for Enhancing Chronic Wound Regeneration.

Chronic wounds (CWs) represent a growing global health concern with profound clinical and socioeconomic implications. Studies indicate that approximately 15% of CWs remain unhealed one year after the initial treatment. At the same time, it is assumed that from 1% to 2% of the population of developed countries will suffer from chronic wounds during their lifetime. CWs severely impair patients' quality of life. Current therapies (compression bandages, antibiotics, hyperbaric oxygen, and skin grafts) face limitations, including toxicity, contraindications, inefficacy in patients with comorbiditie

A promising strategy for ocular noninvasive protein delivery: The case in treating corneal neovascularization.

Current clinical treatment of corneal neovascularization (CNV), a leading cause of visual impairment worldwide, by a class of glucocorticoids suffers from the ineffective and numerous adverse effects. Bevacizumab (Beva), an anti-neovascularization protein, is a promising therapeutic option but limited by subconjunctival injection due to its poor penetration across ocular bio-barriers, which significantly reduces patient compliance and increases the risk of infection. Herein, a CmA@Beva nanomedicine was developed, based on the co-assembly of novelly designed peptide, (Cysteine-Histidine-Arginin
