Engineering an extracellular matrix-functionalized, load-bearing tendon substitute for effective repair of large-to-massive tendon defects.

A significant clinical challenge in large-to-massive rotator cuff tendon injuries is the need for sustaining high mechanical demands despite limited tissue regeneration, which often results in clinical repair failure with high retear rates and long-term functional deficiencies. To address this, an innovative tendon substitute named "BioTenoForce" is engineered, which uses (i) tendon extracellular matrix (tECM)'s rich biocomplexity for tendon-specific regeneration and (ii) a mechanically robust, slow degradation polyurethane elastomer to mimic native tendon's physical attributes for sustaining

Pathogenesis-Guided Rational Engineering of Nanotherapies for the Targeted Treatment of Abdominal Aortic Aneurysm by Inhibiting Neutrophilic Inflammation.

Abdominal aortic aneurysm (AAA) remains a fatal disease in the elderly. Currently, no drugs can be clinically used for AAA therapy. Considering the pivotal role of neutrophils in the pathogenesis of AAA, herein we propose the targeted therapy of AAA by site-specifically regulating neutrophilic inflammation. Based on a luminol-conjugated α-cyclodextrin material (LaCD), intrinsically anti-inflammatory nanoparticles (NPs) were engineered by simple nanoprecipitation, which were examined as a nanotherapy (defined as LaCD NP). After efficient accumulation in the aneurysmal aorta and localization in

Engineering atomically dispersed Mn and Fe sites on hollow nitrogen-doped carbon for high-performance Zn-air batteries.

Zinc-air batteries (ZABs) have garnered significant interest due to their environmental friendliness, low cost, and high energy density. However, their practical application is significantly hindered by high overpotential and sluggish reaction kinetics. Here, we propose a hollow nitrogen-doped carbon supported atomically dispersed Fe and Mn sites (FeMn-HNC) through a facile NaCl-assisted pyrolysis strategy. The synthesized FeMn-HNC catalyst possesses a hollow porous structure, resulting in exceptional oxygen reduction reaction (ORR) catalytic activity, remarkable durable stability, and good to

Probing Deep Hydrogen Polysulfides Fluctuations In Vivo by Engineering Activatable Fluorescence Reporters with Second Near-Infrared Window Emission.

Deep and comprehensive understanding of the intricate biochemical processes mediated by H2Sn (n ≥ 1), but not H2S, is of paramount importance. A few fluorescent probes have been developed with the intention of addressing this issue. However, there is currently no evidence of any activatable NIR-II fluorescent probes for H2Sn-specific imaging over H2S. In this study, we developed the inaugural H2Sn-activated NIR-II probe for specific and deep imaging of H2Sn through a series of molecular engineering strategies. Strong electron-absorbing moieties enabled the redshift of the emission of the desig…

A Multifunctional Tissue-Engineering Hydrogel Aimed to Regulate Bacterial Ferroptosis-Like Death and Overcoming Infection Toward Bone Remodeling.

Infection is the most common complication after orthopedic surgery and can result in prolonged ailments such as chronic wounds, enlarged bone defects, and osteomyelitis. Iron, which is essential for bacterial metabolism and immune cell functions, is extremely important. Bacteria harness iron from nearby cells to promote biofilm formation, ensuring their survival. Iron deficiency within the infection microenvironment (IME) consequently hampers macrophage function, enabling further dissemination of the infection and hindering macrophage polarization to the M2 phenotype. Therefore, a novel approa
