Mice with mutations in Trpm1, a gene in the locus of 15q13.3 microdeletion syndrome, display pronounced hyperactivity and decreased anxiety-like behavior.

The 15q13.3 microdeletion syndrome is a genetic disorder characterized by a wide spectrum of psychiatric disorders that is caused by the deletion of a region containing 7 genes on chromosome 15 (MTMR10, FAN1, TRPM1, MIR211, KLF13, OTUD7A, and CHRNA7). The contribution of each gene in this syndrome has been studied using mutant mouse models, but no single mouse model recapitulates the whole spectrum of human 15q13.3 microdeletion syndrome. The behavior of Trpm1-/- mice has not been investigated in relation to 15q13.3 microdeletion syndrome due to the visual impairment in these mice, which may c

A broad phenotypic screen identifies novel phenotypes driven by a single mutant allele in Huntington's disease CAG knock-in mice.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disorder caused by the expansion of a CAG trinucleotide repeat in the HTT gene encoding huntingtin. The disease has an insidious course, typically progressing over 10-15 years until death. Currently there is no effective disease-modifying therapy. To better understand the HD pathogenic process we have developed genetic HTT CAG knock-in mouse models that accurately recapitulate the HD mutation in man. Here, we describe results of a broad, standardized phenotypic screen in 10-46 week old heterozygous HdhQ111 knock-in mice, prob

Riluzole, a glutamate release inhibitor, and motor behavior.

Riluzole (2-amino-6-trigluoromethoxy benzothiazole) has neuroprotective, anticonvulsant, anxiolytic and anesthetic qualities. These effects are mediated by blockade of glutamate transmission, stabilizing of sodium channels and blockade of gamma-aminobutyric acid (GABA) reuptake. The action profile of riluzole is dominated by its effects on glutamate transmission which are predominately mediated by N-methyl-D-aspartate (NMDA) receptor-linked processes in vitro. In vivo studies show that blockade and stimulation of the different NMDA receptor complex binding sites or AMPA receptors modulate moto

Emotion work in the public performances of male-to-female transsexuals.

Much research has shown that entering the public sphere is emotionally taxing yet key to male-to-female transsexuals' status passage. Yet, little is known about how transsexuals actively manage their emotions during this important transitional phase. Taking a dramaturgical approach to emotions, we explored how some male-to-female transsexuals managed their emotions in ways that helped generate self-confidence and commitment to their paths. Interviewees engaged in three primary forms of emotion work: (1) preparatory emotion work mitigated anxiety and bolstered confidence, which motivated them t

The role of intolerance of uncertainty in bipolar disorder.

OBJECTIVES: Intolerance of uncertainty (IU) is a dispositional trait arising from negative beliefs about uncertainty. IU has been found to be associated with various psychological disorders, including anxiety and depression, but little is known about its role in bipolar disorder (BD). The present study investigates the presentation of IU in BD, including exploring its relationship with mania and impulsivity. METHOD: Participants with a self-reported diagnosis of BD (n = 93), unipolar depression (n = 90) and control participants (n = 91) were recruited from a participant recruitment platform t

Contextual fear conditioning leads to hypo-innervation of the left ventricular myocardium in female but not male C57BL/6J mice.

OBJECTIVE: Contextual fear conditioning is an effective way of inducing and modelling anxiety-like states in rodents like mice. We examined the sex-dependent effects of contextual fear conditioning on the autonomic innervation of the left ventricular myocardium in female and male mice. METHODS: Contextual fear conditioning was induced in male and female C57BL/6J mice and compared to male and female controls (n = 5 each). The experimental procedure consisted of two days of habituation, one day of training and one day of context fear memory retrieval. 24 h after context fear memory retrieval, h

Medial Preoptic Nucleus to Periaqueductal Gray GABAergic Pathway Underlies Individual Differences in Consolation-Like Behavior in Mice.

INTRODUCTION: Consolation is a complex empathic behavior that varies considerably across individuals, yet the neural basis of this heterogeneity remains poorly understood. The medial preoptic area is a key hub for instinctive social behaviors, but it remains unclear which of its subregions - the medial preoptic nucleus (MPN) versus the surrounding area (MPA) - or which neuronal subtypes (GABAergic, glutamatergic, or galanin-expressing) contribute to individual differences in consolation. METHODS: We used a social defeat-induced consolation paradigm in male mice and categorized them into low-

How to predict medication adherence and other health-related adherence in patients with inflammatory arthritis?

OBJECTIVE: This study investigated the role of psychological variables (anxiety, depression, beliefs about medication, satisfaction and frustration of basic psychological needs, and mindfulness skills) on adherence in patients with inflammatory arthritis (IA). METHODS: 250 patients with IA completed an online questionnaire at baseline (T0) and three months later (T1). Their adherence was assessed at T1 with the General Adherence for Chronic Disease Profile (GACID-P) exploring patients' daily life and medical domains of adherence. Measures carried out at T0 included sociodemographic variables,

Effectiveness of online mindfulness-based interventions based on the structured curriculum framework in improving mental health: A meta-analysis of randomized controlled trials.

With the rapid development of digital technology, online Mindfulness-Based Interventions (MBIs) are increasingly being used in the mental health field. There is a lack of systematic quantitative synthesis of the effectiveness of online MBIs with a structured curriculum framework. This meta-analysis aims to evaluate the overall impact of online MBIs based on the structured curriculum framework in improving mental health. This study performed a random effects meta-analysis of 38 RCTs. Analysis shows, significant effect sizes were found for symptoms of anxiety (Ncomp = 25, N = 3890, SMD = -0.44,

Mapping the thalamic subnuclei in fibromyalgia: insights into pain modulation and affective amplification.

Fibromyalgia is a chronic disorder characterized by widespread pain, fatigue, sleep problems, and emotional or cognitive difficulties. The thalamus is central to both pain modulation and affective processing, yet structural alterations within its subregions remain unclear. This study examined volumetric characteristics of thalamic subnuclei in individuals with fibromyalgia and explored their relationships with clinical symptoms. Structural magnetic resonance imaging data from 20 female patients with fibromyalgia and 20 age and gender-matched healthy controls were analyzed using automated segme

Infralimbic cortex neurostimulation attenuates nociceptive and affective manifestations in a reserpine-induced fibromyalgia model.

Fibromyalgia (FM) is a chronic syndrome characterized by widespread pain, frequently accompanied by cognitive deficits and psychiatric symptoms. FM has been associated with large-scale functional alterations of brain networks involved in pain processing, particularly the default mode network (DMN) and the pain salience network. The infralimbic cortex (IL), a region of the medial prefrontal cortex (mPFC) considered part of the DMN, is functionally connected to regions involved in salience processing. However, the role of the IL in modulating FM-related manifestations remains poorly understood.

Genome-Wide Association Study of Symptom Change Following Cognitive Behavioral Therapy for Common Mental Disorders.

Cognitive behavioral therapy (CBT) is a well-established, evidence-based treatment for common mental disorders such as depression, anxiety disorders, and obsessive-compulsive disorder (OCD). However, treatment outcomes vary widely, and a substantial proportion of patients do not achieve sufficient improvement. Robust predictors of individual differences in symptom change are currently lacking. Genetic differences have been suggested to play a role, but existing evidence is inconclusive. This study investigated the extent to which common genetic variants-single nucleotide polymorphisms (SNPs)-c
