Alterations in mood, cognitive behaviour and functional connectivity between brain regions in healthy adult males with acute total sleep deprivation.

OBJECTIVES: Although acute total sleep deprivation (TSD) is known to harm health and daily activities, how it specifically alters the link between distinct brain regions and emotional‑cognitive behaviors remains poorly understood. This study therefore investigated mood changes, cognitive performance, and functional connectivity in healthy adult males after 36 h of TSD. METHODS: Sixty healthy adult males participated in a within‑subject protocol. Assessments were performed before and after 36 h of TSD using multiple measures: mood rating scales, behavioral tests of sustained attention and aler

Medial prefrontal cortex and ventrolateral orbitofrontal cortex inputs into the ventrolateral periaqueductal grey region differentially regulate pain and anxiety like behavior.

Distinct cortical inputs to the ventrolateral periaqueductal gray (vlPAG) have been implicated in pain modulation, however, the effect of their simultaneous activation on pain and anxiety remains unclear. Here, we examined the roles of medial prefrontal cortex (mPFC) and ventrolateral orbitofrontal cortex (vlOFC) projections to the vlPAG in a neuropathic pain model. Using optogenetic stimulation in spared nerve injury (SNI) mice, activation of either mPFC or vlOFC inputs to the vlPAG partially reduced mechanical hypersensitivity, whereas simultaneous activation produced a complete analgesic ef

Rare variants in FAT3 as possible contributors to non-syndromic orofacial cleft risk.

Non-syndromic orofacial clefts (NSOFCs) represent the most common human craniofacial malformations, yet the majority of their genetic causes remain unclear. This study, by analyzing whole-exome sequencing (WES) data from multiple patients with NSOFCs, identified eight de novo low-frequency missense variants in the FAT3 gene, all of which were located within the extracellular cadherin domains of the FAT3 protein. Sanger sequencing confirmed the authenticity of these variants, while bioinformatic predictions and protein structural modeling suggested potential disruption of FAT3 function.Using in

Right hippocampal volume and attenuated psychotic symptoms distinguish subgroups of youth with common patterns of temporoparietal effective connectivity.

Associations between patterns of brain connectivity and brain structural features have transdiagnostic relevance to psychopathology. There is considerable evidence for disruptions to the hippocampus and temporoparietal brain systems in psychotic disorders. The present study examines structure-function relationships-specifically, the relation of hippocampal volume with patterns of temporoparietal effective connectivity-in youth at clinical high-risk for psychosis (CHR-P) and healthy controls (HCs). Participants at CHR-P and HCs completed clinical symptom measures and magnetic resonance imaging

Distinct cerebellar networks underpin clinical improvement in adolescent Tourette disorder.

Adolescence is frequently called the second period of brain maturation. In Tourette disorder (TD), the clinical trajectory of tics and associated psychiatric co-morbidities varies significantly across individuals during the transition from adolescence to adulthood. In this study, we aimed to identify patterns of resting-state functional connectivity that differentiate adolescents with Tourette disorder from their neurotypical peers, and to monitor symptom-specific functional changes over time. We employed a multivariate analysis based on a support vector machine (SVM) to predict patterns of re

Modeled Long-Term Effects of Psilocybin on Dynamic Activity and Effective Connectivity of Fronto-Striatal-Thalamic Circuits.

Psilocybin has been shown to induce fast and sustained symptoms improvements across various psychiatric conditions, yet its long-term mechanisms of action are not fully understood. Initial evidence suggests that longitudinal functional and structural brain changes implicate fronto-striatal-thalamic (FST) circuitry, a broad system involved in goal-directed behavior and motivational states. Here, we performed secondary analyses and applied computational modeling to resting-state fMRI data from a within-subject longitudinal psilocybin trial in psychedelic-naïve healthy volunteers. We first showed

Corticostriatal connectivity in monozygotic twin pairs discordant for obsessive-compulsive disorder.

Traditional case-control studies show that obsessive-compulsive disorder (OCD) is associated with disturbances in cortico-striatal-thalamo-cortical (CSTC) brain circuits. Whether these findings represent underlying familial vulnerabilities (i.e. shared genetic and environmental factors) or are the result of acquired non-shared environmental influences, such as chronic stress or lifestyle factors, is unknown. Using a unique sample of monozygotic twin pairs discordant for lifetime OCD diagnoses (14 pairs, 28 twins) and resting-state functional MRI, we examined the contribution of non-shared envi

Role of PBN-CeA Circuit in Paclitaxel-Induced Neuropathic Pain and Negative Emotions.

BACKGROUND: Paclitaxel produces a chronic, glove-stocking neuropathy that is frequently accompanied by anxiety and depression. However, the neural circuits and molecular signals driving this pain-affect comorbidity remain unclear. METHODS: Chemotherapy-induced neuropathic pain (CINP) was established by paclitaxel injections in male rats. Mechanical and thermal sensitivity and negative emotional behaviors were assessed using behavioral tests. Parabrachial nucleus (PBN)-central amygdala (CeA) projections were traced with viral vectors, and optogenetics was used to manipulate PBN terminals. Prod

Activity-Dependent Regulation of Basal Forebrain Cholinergic Neurons by Striatal Spiny Projection Neurons.

The basal forebrain cholinergic neurons (BFCNs) in substantia innominata (SI) and nucleus basalis magnocellularis (nBM) provide dense innervation to multiple cortical areas, shaping cognition, attention, and the sleep-wake cycle. However, the afferent connectome controlling the spiking of BFCNs is poorly characterized. Although biased toward the nearby ventral striatum, monosynaptic rabies virus mapping revealed that spiny projection neurons (SPNs) throughout the striatum innervated BFCNs in the SI/nBM. Roughly 1-5% of SPNs were retrogradely labeled, with both D1 dopamine (DA) receptor-express

Gastrodin enhances stress resilience through promoting Wnt/β-Catenin-dependent neurogenesis.

BACKGROUND: Enhancing stress resilience constitutes a pivotal strategy in mitigating the risk of depression, making it a critical component of both prevention and treatment. In the current work, we identified a compound, gastrodin (GAS), as capable of enhancing stress resilience, as demonstrated by its ability to protect against depression following chronic stress exposure. OBJECTIVES: To elucidate the potential of GAS to promote neurogenesis under chronic stress, along with the associated cellular and molecular processes involved. METHOD: We evaluated the effect of GAS on NSPC proliferation

DRN-SNc serotonergic circuit drives stress-induced motor deficits and Parkinson's disease vulnerability.

Stress is a recognized risk factor for Parkinson's disease (PD), but the mechanisms by which stress exacerbates PD symptoms through the serotonergic system are not fully understood. This study investigates the role of serotonergic (5-HT) neurons in the dorsal raphe nucleus (DRN) in mediating stress-induced motor deficits and PD progression. Acute and chronic stress were induced in mice using an elevated platform (EP) and combined with MPTP administration to model early-stage PD. Acute EP stress caused transient motor deficits and significant activation of DRN5-HT neurons projecting to substant

A Novel Long-range Excitatory Neural Circuit from the Magnocellular Red Nucleus to Spinal Dorsal Horn Facilitates Neuropathic Pain-like Behaviors in Male Mice.

Neuropathic pain arises from a primary lesion or disease affecting the somatosensory system; however, the brain circuitry underlying the regulation of neuropathic pain remains unclear. The red nucleus (RN) is a critical brain region involved in regulating muscle tension, coordinating movement, and facilitating sensorimotor integration. While some evidence has shown that RN regulates pain, its specific role in neuropathic pain and associated neural circuits remains elusive. In this study, we found that CaMKIIα-positive neurons in the magnocellular red nucleus (RMCCaMKIIα) were activated followi

Cerebellar Activity Affects Distal Cortical Physiology and Synaptic Plasticity in a Human Parietal-Motor Pathway Associated with Motor Actions.

Voluntary movement control depends on plasticity in several interconnected brain regions, including the cerebellum (CB), primary motor cortex (M1), and posterior parietal cortex (PPC). It is thought that one role of the CB is to regulate communication between PPC and M1, but causal evidence for this regulatory role in humans remains lacking. Here, we evaluated how transiently altering activity in CB via intermittent theta burst stimulation (iTBS) affects PPC-M1 connectivity and plasticity by assessing the effectiveness of subsequent Hebbian-like cortical paired associative stimulation (cPAS) t

Brain activity and functional connectivity patterns associated with loneliness: A resting-state fMRI study.

Loneliness is an unpleasant subjective experience associated with significant psychological and physical health problems. With increasing urbanization and aging populations, loneliness is becoming a global public health concern. Thus, understanding the neural correlates of loneliness is crucial for developing targeted intervention approaches. In the current study, we collected resting-state fMRI data from 238 young adults (ages 17-26; 59 males, 179 females) and used fractional amplitude of low-frequency fluctuations (fALFF) and functional connectivity (FC) analyses to investigate the neural co

The structural-functional connectivity coupling in the right caudal hippocampus mediates the effects of body mass index on cognitive function.

BACKGROUND: Studies have demonstrated an association between cognitive decline and body mass index (BMI). However, the neural mechanisms that underlie this association remain unclear. This study aimed to investigate the relationship between BMI, cognitive function, and brain structural-functional connectivity (SC-FC) coupling. METHODS: This study included 490 healthy subjects undergoing resting-state functional magnetic resonance imaging (rs-fMRI) and diffusion tensor imaging (DTI) scans and cognitive function tests. Structural connectivity (SC) was constructed using DTI, and functional conne

Locus Coeruleus Microstructure and Connectivity as Novel Markers of Depression and Cognitive Dysfunction in Older Adults.

BACKGROUND: Late-life depression (LLD) is a risk factor for age-related cognitive decline. Postmortem studies have highlighted pathological changes in the locus coeruleus (LC) and its projections as potential early cognitive vulnerability markers. Here, we used a novel individualized multimodal magnetic resonance imaging (MRI) approach to characterize the cognitive correlates of LC microstructure and connectivity in participants with LLD and age-matched never-depressed (ND) control participants. METHODS: Diffusion-weighted and LC-sensitive MRI were acquired for 52 participants (LLD: n = 26, 1

Gamma oscillatory transcranial direct current stimulation of motor cortex enhances corticospinal excitability and brain connectivity in healthy individuals.

Cortical excitability, the tendency of neurons to respond to various stimuli, is impaired in most neuropsychiatric conditions. Non-invasive brain stimulation can exert therapeutic effects by modulating the cortical excitability. Transcranial direct current stimulation (tDCS) and transcranial alternating current stimulation (tACS) have shown promise in various neuropsychiatric disorders, including improving cognitive abilities and motor function following stroke. Oscillatory transcranial direct current stimulation (otDCS), as a novel stimulation paradigm, combines tDCS and tACS to simultaneousl

Cognitive prediction using regional connectivities and network biomarkers in Alzheimer's disease.

Achieving a deep understanding of brain mechanisms requires multi-scale perspectives to capture the architecture of complex networks. In this study, we focused on patients with cognitive impairment and constructed individual brain networks from neuroimaging data. We introduced a Significant Edges Selection (SES) method, which effectively extracts the most informative connections while suppressing noise. Using these refined features, we computed network characteristics and applied machine learning models to predict cognitive performance, achieving a prediction accuracy of correlation r = 0.683
