Dry eye disease in axial spondyloarthritis: heterogeneous ocular surface dysfunction involving tear film instability and tear production from the AIDA network registry.

OBJECTIVE: Dry eye disease (DED) has been described in axial spondyloarthritis (axSpA), but patterns of tear fluid and ocular surface involvement remain incompletely characterized. This study aims to evaluate tear film instability and tear production in axSpA and explore the heterogeneity of DED manifestations. METHODS: Axial spondyloarthritis patients and healthy controls underwent non-invasive tear break-up time (niTBUT), Schirmer test, and Ocular Surface Disease Index (OSDI)- 6 evaluation. Group differences were assessed using niTBUT thresholds (<10 s and <5 s). RESULTS: Mean niTBUT was s

Computational analysis of hub genes associated with sarcopenia: integrative transcriptome insights from an Asian cohort.

Sarcopenia, a progressive loss of skeletal muscle mass and strength, leads to frailty, falls, fractures, and delayed recovery following orthopedic surgery. When combined with osteoporosis, it manifests as osteosarcopenia, exacerbating musculoskeletal fragility. Although chronic inflammation, mitochondrial dysfunction, and impaired autophagy are recognized contributors, the integrated regulation of these processes in Asian populations remains unclear. This study aimed to elucidate molecular mediators and signaling pathways connecting inflammation, autophagy, and muscle-bone degeneration using a

Perspectives on intrinsic articular cartilage interfaces.

INTRODUCTION: According to the current concept the joint is an organ composed of multiple tissues forming manifold communicating interfaces. Moreover, cartilage itself presents typical intrinsic zonal interfaces. Since the healthy joint cartilage is not vascularized, soluble mediators are distributed by the synovia fluid flow in the cartilage extracellular matrix (ECM) of the joint compartments. This distribution is dependent on intermittent mechanical loading and water flow in the joint. Hence, soluble factors play a pivotal role in articular cartilage interfacial communication. These mediato

Loss of type III collagen has limited effect on tendon healing in young adult mice.

Tendon injuries account for considerable and escalating clinical burden. After injury, tendons heal poorly with persistent fibrovascular scar. Unlike healthy tendon which is type I collagen (COL1)-rich and highly-aligned, fibrovascular scar is type III collagen (COL3)-rich and disorganized; this compositional and organizational change has been implicated in the functional deficits resulting from tendon injury. Accordingly, COL3 is historically considered a primary driver of poor tendon healing. Given compelling evidence of COL3's role in regulating matrix structure and cell behavior, we sought

Brain irradiation drives remote liver changes via senescence-independent mechanisms.

BACKGROUND AND PURPOSE: Although radiotherapy is essential for treating brain tumors, it often damages surrounding healthy brain tissue, causing long-term neurocognitive dysfunction. Cellular senescence, a stable state of cell cycle arrest triggered by various stressors including radiation-induced DNA damage, has been implicated in aging- and neurodegeneration-associated cognitive decline via its pro-inflammatory secretome and has been reported to exert paracrine effects on distant organs. However, whether brain irradiation, particularly through the induction of cellular senescence, can trigge

TRECing down the source: naive and memory T cell generation in dirty mice and men.

Healthy aging relies on the maintenance of a diverse T cell pool. This diversity is ensured by balancing thymic output, differentiation of naive into memory T cells, T cell proliferation and cell death. For naive T cells, the balance of these processes differs between standard laboratory mice and humans. This may be a true species difference or, alternatively, result from the vastly different amounts of antigens to which standard laboratory mice and humans are exposed. Using wildlings, that is, laboratory mice born to wild mice, we studied the impact of antigen-exposure through a natural micro…

Female resistance to the metabolic benefits of protein restriction is reversed by ovariectomy in mice.

Dietary protein intake mediates healthy aging in diverse species, with consumption of a low protein (LP) diet improving metabolic health in both humans and mice. In mice, the benefits of LP diets are sex-specific, with males exhibiting a stronger response to a LP diet than females. The reason for this sexually dimorphic response is unknown, but we hypothesized that sex hormones might be responsible for this difference. Here, we tested the role of sex hormones in the response to a LP diet by feeding intact and gonadectomized mice of both sexes either a Control (21% of calorie from protein) or L…

Older Adults share an agreement on experiences that elicit love: A study of consensus and individual differences in judgments.

OBJECTIVES: Positive social connections are key ingredients for healthy aging. We studied older adults' shared agreement on loving experiences and individual differences in their decisions-making to unlock critical information about how this developmental group connects socially. METHODS: To explore shared agreement on experiences of love, we surveyed a representative sample (N=400) of adults aged 65 years and older in the United States. They evaluated 61 experiences to determine whether older adults believe they typically evoke feelings of love in most people. We used a Bayesian decision tre…

Association of brain structure and motor function in older persons with multiple sclerosis and healthy controls: A cross-sectional study.

INTRODUCTION: Brain structure deteriorates with aging as well as with neurodegenerative diseases such as multiple sclerosis (MS). While these brain changes are accompanied by deterioration of motor function, the association between brain structures and motor function in older people with MS (pwMS) has been scarcely investigated, especially across a broad range of outcomes. OBJECTIVE: To investigate the association between a broad range of brain structures and motor function outcomes in older pwMS as well as in older, healthy controls (HC). METHODS: The present cross-sectional study included

Longitudinal Analysis of Infraorbital Aging Over a 15-Year Period: An MRI-based Study in Asian Patients.

BACKGROUND: Palpebral bags are commonly managed with infraorbital fat excision during lower blepharoplasty. This surgical approach is based on the assumption of age-related anterior herniation of infraorbital fat. However, controversy remains regarding the true volumetric changes of infraorbital fat with aging. OBJECTIVES: The authors of this study aimed to investigate longitudinal anatomical changes in the infraorbital region. METHODS: A retrospective longitudinal analysis of serial MRI images from 50 healthy individuals (20 males and 30 females) aged 20 to 60 years was conducted; the mean

Detecting artificially impaired balance in human locomotion: metrics, perturbation effects and detection thresholds.

Measuring balance is important for detecting impairments and developing interventions to prevent falls, but there is no consensus on which method is most effective. Many balance metrics derived from steady-state walking data have been proposed, such as step-width variability, step-time variability, foot placement predictability, maximum Lyapunov exponent and margin of stability. Recently, perturbation-based metrics such as center of mass displacement have also been explored. Perturbations typically involve unexpected disturbances applied to the subject. In this study we collected walking data

Closed-loop control of split belt treadmill speed induces targeted bilateral adaptations in ankle kinetics.

On a conventional split-belt treadmill, one leg walks exclusively on a fast belt while the other is on a slow belt. While this setup effectively induces asymmetrical adaptations beneficial for unilateral deficits like post-stroke hemiparesis, its design is less ideal for symmetrical impairments such as bilateral plantarflexor (PF) weakness common in aging. We introduce a novel split-belt treadmill paradigm with two distinct conditions that vary each belt's speed based on its corresponding limb's phase of gait. In the 'TrailFast' condition, each belt accelerated across the stance phase and dece

Mapping the neural substrate of high dual-task gait cost in older adults across the cognitive spectrum.

The dual task cost of gait (DTC) is an accessible and cost-effective test that can help identify individuals with cognitive decline and dementia. However, its neural substrate has not been widely described. This study aims to investigate the neural substrate of the high DTC in older adults across the spectrum of cognitive decline. A total of 336 individuals from the GAIT study cohort were analyzed, including cognitively healthy (N = 122, 71 ± 3.6 years), those with mild cognitive impairment (N = 168, 71 ± 5.3 years), and those with dementia (N = 46, 80 ± 5.7 years). A DTC of 20% or greater was

Can we detect cognitive "super-agers" in Parkinson's disease? Cognitive, neuropsychiatric and motor outcomes in the first 10 years of Parkinson's disease.

INTRODUCTION: Superagers are older individuals who resist age-related cognitive decline and who perform just as well or better than younger people in cognitive tasks. In healthy aging studies, superagers not only experience positive cognitive outcomes (e.g. lower risk of future mild cognitive impairment), but may also experience fewer depressive and anxiety symptoms as they age. We seek to identify superagers in Parkinson's disease (PD) and potential outcomes associated with superager status. METHODS: Data was gathered from the Parkinson's Progression Markers Initiative (PPMI). 689 individual…

An integrated bioinformatics and machine learning approach to identifying biomarkers connecting parkinson's disease with purine metabolism-related genes.

BACKGROUND: Parkinson's disease (PD), a prevalent neurodegenerative disorder in the aging population, poses significant challenges in unraveling its pathogenesis and progression. A key area of investigation is the disruption of oncological metabolic networks in PD, where diseased cells display distinct metabolic profiles compared to healthy counterparts. Of particular interest are Purine Metabolism Genes (PMGs), which play a pivotal role in nucleic acid synthesis. METHODS: In this study, bioinformatics analyses were employed to identify and validate PMGs associated with PD. A set of 20 candid
