We examined the effects of ruscogenin glycoside (Lm-3), isolated from Liriope muscari, on lymphocyte adhesion to extracellular matrix. Adhesion of Jurkat cells activated by anti-CD3 to type I collagen was inhibited by Lm-3 in a concentration- and time-dependent manner. Lm-3 also inhibited the cell attachment to fibronectin and laminin. However, the saponin did not influence anti-CD3-induced cell proliferation and Mn2+-induced adhesion. Protein kinase C activator, phorbol 12,13-dibutyrate, significantly enhanced, while its inhibitor, chlorpromazine, almost completely blocked, the adhesion of an

Present studies emphasize the pharmacological, toxicological, and molecular simulation studies of the potent phytoconstituents in the Sanchezia nobilis leaf extract (SNE) and its nonpolar and polar fractions. We identified 27 phytoconstituents in SNE using Gas Chromatography Mass Spectrometry (GC-MS), where aniline, dibutyl phthalate, and isoquinoline are noteworthy. Anxiolytic assessments by hole board test (HBT) and elevated plus maze test (EPMT) demonstrated dose-dependent effects of nonpolar and polar fractions of SNE. Both fractions also exhibited minimal cytotoxicity in brine shrimp leth

Anaphylaxis is an acute, potentially life-threatening reaction, often triggered by foods and largely mediated by immunoglobulin (Ig)E. The human microbiota is known to influence oral tolerance, but the microbial mechanisms directly involved in IgE-mediated anaphylaxis remain unknown. Here, we demonstrate that human saliva and jejunum harbor peanut (PN)-degrading bacteria that metabolize immunodominant allergens (Ara h 1 and 2), reducing IgE binding and anaphylaxis. Isolated Rothia and Staphylococcus species degraded PN allergens in vitro, generating proteins with reduced IgE binding and limite

Riboflavin (vitamin B₂; RF) and its derivatives flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD) are indispensable cofactors for redox reactions in plants. While higher plants possess a conserved pathway for de novo RF biosynthesis, how flavins are transported and spatially distributed between tissues remains unresolved. In particular, the molecular identity of plasma membrane (PM) transporters involved in flavin transport in plants remains largely unknown. Here, we identify Arabidopsis purine permease 5 (AtPUP5) as a PM-localized protein associated with RF transport and its d

Roots adapt to growth-restricting but nonlethal high temperatures. Although lethal heat shock and moderately elevated temperatures have been extensively studied, the effects of nonlethal thermal stress on root development remain unclear. We defined 31°C as a nonlethal thermal stress condition in Arabidopsis thaliana and examined its effects using phenotypic and transcriptomic analyses. Compared with 22°C, growth at 31°C reduced primary root growth, meristem size, and superoxide (O2˙-) accumulation, accompanied by altered redox states, restricted distribution of the meristem regulator PLETHORA2

A phytochemical investigation of the roots of Euphorbia fischeriana led to the isolation of four tigliane-type diterpenoid glucosides (1-2, 4, and 5) and two daphnane-type diterpenoid glucosides (3 and 6). Compounds 1-3 represent previously undescribed structures. Their chemical structures were fully elucidated through comprehensive spectroscopic analyses, including HRESIMS, IR and NMR experiments. A plausible biosynthetic pathway for 1-6 is also proposed. In bioactivity tests, compounds 2, 4, and 5 exhibited notable inhibitory effects on lipopolysaccharide (LPS)-induced nitric oxide (NO) prod

This study aimed to investigate the inhibitory effect of the chloroform fraction (CF) from Vitis vinifera root extract on LPS-induced neuroinflammation in BV-2 microglia cells and a C57/BL6J mouse model. CF significantly suppressed LPS-induced proinflammatory cytokines, including nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in BV-2 microglia cells. Mechanistically, CF inhibited LPS-induced activation of nuclear factor-κB (NF-κB) by blocking the p65 subunit and preventing the phosphorylation of NF-kappa-B inhibitor α (IκBα), while its effect was independent of th

Plants have evolved an elaborate cell wall integrity (CWI) sensing system to monitor and modify cell wall formation. LRR-extensins (LRXs) are cell wall-anchored proteins that bind RAPID ALKALINIZATION FACTOR (RALF) peptide hormones and induce compaction of cell wall structures. LRXs also form a signaling platform with RALFs and the transmembrane receptor kinase FERONIA (FER) to maintain cell wall integrity. LRX1 of Arabidopsis thaliana is predominantly expressed in root hairs, and lrx1 mutants develop defective root hairs. Here, we identify a regulator of LRX1-RALF-FER signaling as a suppresso
