Waterpipe smoke condensate activates autophagy in non-tumorigenic lung epithelial cells: Implications for inflammation and stemness.

Waterpipe smoking has emerged as a major global health concern, with incompletely characterized effects on bronchial epithelial cells. In this study, we investigated the cytotoxic and adaptive responses of non-tumorigenic human bronchial epithelial BEAS-2B cells exposed to waterpipe smoke condensate (WPSC). Increasing concentrations of WPSC reduced cell viability in a dose-dependent manner; however, exposure to 0.1% WPSC caused only a modest, transient reduction in viability that was no longer significant by 72 hrs and did not significantly induce apoptosis over 72 hrs, as determined by MTT (3…

Pseudomonas aeruginosa infection causes lysosomal dysfunction in the cystic fibrosis bronchial epithelium.

Cystic fibrosis (CF) is a genetic disorder that causes chronic airway disease, leading to accelerated aging, inflammation, and susceptibilit … aling. In conclusion, our study implies that PA infection does not induce cell senescence in the CF bronchial epithelium but disturbs lysosomal homeostasis, which can be attenuated by RAPA. These findings highlight lysosomal health as a promising therapeutic target in CF airways, especially as patients live longer yet remain susceptible to recurrent airwa …

TGF-β Receptor-dependent Tissue Factor Release and Proteomic Profiling of Extracellular Vesicles from Mechanically Compressed Human Bronchial Epithelial Cells.

In asthma, tissue factor (TF) concentrations are elevated in the lung. In our previous studies using mechanically compressed human bronchial epithelial (HBE) cells, which are a well-defined in vitro model of bronchoconstriction during asthma exacerbations, we detected TF within extracellular vesicles (EVs) released from compressed HBE cells. Here, to better characterize the potential role of this mechanism in asthma, we tested the extent to which the transcriptional regulation of epithelial cell-derived TF varied between donors with and without asthma. Using RNA in situ hybridization, we detec…

In vitro effects of aged low-density polyethylene micro(nano)plastic particles on human airway epithelial cells.

Airborne micro(nano)plastic (MNP) pollution has emerged as a major global concern due to its increasingly worrying adverse health effects. E … ed for the first time the toxicological effects of environmentally relevant aged LDPE MNPs on human bronchial epithelial cells (BEAS-2B). UV-aged LDPE fragments of irregular sizes and shapes were used to mimic real atmospheric particles, and BEAS-2B cells were exposed to 10-1000 μg/cm2 of LDPE MNPs. Our results showed that MNPs were internalized by BEAS-2 …

Dosimetric evaluation of target and peri-cavity by replacing CT values for cavities within the target in volumetric modulated arc therapy.

Volumetric-modulated arc therapy (VMAT) enables advanced dose modulation and is effective for treating tumors with complex shapes. However, for targets that contain low-density regions such as cavities, issues arise with dose distribution heterogeneity caused by high-fluence beams and excessive doses to the surrounding tissues owing to positional shifts during the treatment period. This study aimed to evaluate a treatment planning method in VMAT that replaces the computed tomography (CT) values of cavities within the target to suppress excessive peribronchial doses caused by interfractional mo…

Arsenic disrupts H3K9me3 and H3K27me3 balance by biasing PRC2.1 and PRC2.2 activity via PALI1 inhibition in carcinogenesis.

Inorganic arsenic (As3+) is a well-established human carcinogen, yet the molecular mechanisms underlying its oncogenic potential remain incompletely understood. Here, we show that exposure to environmentally relevant concentrations of As³⁺ disrupts chromatin architectures in human bronchial epithelial cells (BEAS-2B) by discordantly regulating two key repressive histone modifications: histone H3 lysine 27 trimethylation (H3K27me3) and H3K9me3. Chromatin immunoprecipitation and sequencing (ChIP-seq) reveals a genome-wide gain of H3K27me3 and a marked loss of H3K9me3 following As³⁺ treatment. Me…

Efficacy of Spray Cryotherapy on Airway Secretions in Canine Models of Chronic Bronchitis and Mechanism Insights.

INTRODUCTION: Bronchoscopic spray cryotherapy (SCT) is a novel treatment showing promise for chronic bronchitis (CB), characterized by excessive mucus secretion and productive cough. A large animal model for preclinical research of SCT is lacking, and its treatment's efficacy and mechanisms for CB are not well understood. METHODS: Eight Labradors were exposed to 200 ppm SO2 for 6 months to develop a CB model. Evaluations included pulmonary resistance, bronchoalveolar lavage fluid (BALF), CT images, and pathology. After model validation, 6 dogs received SCT and were observed for short-term (7 …

Airway Mucus Plugging in Chronic Bronchitis and the Impact of Bronchial Rheoplasty.

BACKGROUND: Chronic bronchitis (CB) is characterized by excessive airway mucus production. Mucus plugs occluding the airway are common in CB, are associated with increased all-cause mortality, and can be identified on CT imaging. RESEARCH QUESTION: Does bronchial rheoplasty (BR) reduce mucus plugging (MP) evident on CT imaging by basket catheter expansion and delivery of pulsed electric fields to the airway mucosa? STUDY DESIGN AND METHODS: Fifty-five patients with symptomatic CB (COPD Assessment Test cough and phlegm scores, ≥ 7 of 10) underwent bilateral BR procedures with CT performed bef…

Missing piece: tracheobronchial foreign body aspiration during an elective thyroidectomy.

Airway management during thyroid surgery is challenging, particularly in patients with altered upper airway anatomy that may impair laryngoscopic visualisation. Bougies are commonly used to aid difficult intubation, but in rare cases, they can fracture and lead to tracheobronchial aspiration. We report the case of a woman in her mid-50s with multinodular goitre suspicious for malignancy who underwent elective thyroidectomy. Difficult intubation necessitated bougie use, but postoperatively, its tip was found missing. Chest CT confirmed the fragment lodged in the right lower lobe bronchus. The p…

Molecular cargo of small EVs from NSCLC patient BALF and plasma: unveiling their role in airway inflammation and immune regulation in a novel human 3D bronchial model.

BACKGROUND: Tumor-derived small extracellular vesicles (EVs) play a crucial role in modulating immune responses and shaping the tumor microenvironment; however, their functional impact on airway immunity in NSCLC remains largely unexplored. This study represents the first attempt to investigate the immunomodulatory and tumor-promoting effects of NSCLC-derived EVs in a human 3D bronchial airway model, which closely mimics the human lung microenvironment. METHODS: EVs were isolated from the plasma and bronchoalveolar lavage fluid (BALF) of NSCLC patients and analyzed via nanoparticle tracking a…

Identification of bronchial epithelial genes associated with type 2 eosinophilic inflammation in asthma.

BACKGROUND: Airway inflammation plays a critical role in asthma pathogenesis and pathophysiology, but the molecular pathways contributing to airway inflammation are not fully known, particularly type 2 (T2) inflammation characterized by both eosinophilia and higher fractional exhaled nitric oxide (Feno) levels. OBJECTIVE: We sought to identify genes whose level of expression in epithelial brushing samples were associated with both bronchoalveolar lavage (BAL) eosinophilia and generation of Feno. METHODS: We performed segmental allergen bronchoprovocation (SBP-Ag) in participants with asthma,

Nanoplastics Penetrate Human Bronchial Smooth Muscle and Small Airway Epithelial Cells and Affect Mitochondrial Metabolism.

Micro- and nanoplastic particles, including common forms like polyethylene and polystyrene, have been identified as relevant pollutants, potentially causing health problems in living organisms. The mechanisms at the cellular level largely remain to be elucidated. This study aims to visualize nanoplastics in bronchial smooth muscle (BSMC) and small airway epithelial cells (SAEC), and to assess the impact on mitochondrial metabolism. Healthy and asthmatic human BSMC and SAEC in vitro cultures were stimulated with polystyrene nanoplastics (PS-NPs) of 25 or 50 nm size, for 1 or 24 h. Live cell, la…

Infant bronchial tree simulator: Success of a built-from-scratch model for single lung isolation.

BACKGROUND: One-lung ventilation in infants is a high-risk procedure. Complications include endotracheal tube occlusion, with grave consequences. Although there are commercially available bronchoscopy simulators, there are no realistic models of infant patients. This limits access to training opportunities that would ensure safe and efficient lung isolation. To bridge this gap, we developesd a realistic infant bronchial tree model for single lung intubation and evaluated preliminary validity evidence of its features and clinicians' ability to perform critical skills associated with pediatric o…

FGF2 is secreted in extracellular vesicles from lung cells.

The 18-kDa isoform of basic fibroblast growth factor (bFGF/FGF2) lacks a conventional signal peptide sequence and is exported by a novel membrane-associated transport pathway. Extracellular vesicles (EVs) are increasingly recognized as mediators of intercellular communication in the lung, and our prior work demonstrates that EVs carry cargo that contributes to hyperoxic lung injury and are biomarkers for bronchopulmonary dysplasia. We used primary human bronchial epithelial (HBE), pulmonary artery endothelial (HPAE), and fibroblast (HNF) cells to determine whether FGF2 was secreted in EVs. EVs…
