Strategies for the Viral Exploitation of Nuclear Pore Transport Pathways.

Viruses frequently exploit the host's nucleocytoplasmic trafficking machinery to facilitate their replication and evade immune defenses. By encoding specialized proteins and other components, they strategically target host nuclear transport receptors (NTRs) and nucleoporins within the spiderweb-like inner channel of the nuclear pore complex (NPC), enabling efficient access to the host nucleus. This review explores the intricate mechanisms governing the nuclear import and export of viral components, with a focus on the interplay between viral factors and host determinants that are essential for

Cryptococcus neoformans and the EGFR Puzzle: Uncovering a Novel mechanism for blood-brain barrier crossing.

This study aims to investigate the molecular mechanisms by which Cryptococcus neoformans (C. neoformans) crosses the blood-brain barrier (BBB), focusing specifically on the role of the epidermal growth factor receptor (EGFR) and its ligand, HB-EGF. Cryptococcal meningitis, caused by C. neoformans, has a high mortality rate and poses a significant threat to global public health. Research indicates that C. neoformans employs various strategies to cross the BBB, with transcellular transport being particularly critical. We observed that C. neoformans infection significantly upregulates the express

Lamin A/C promotes HBV transcription by modulating histone modification associated with cccDNA minichromosome in an HBx-dependent manner.

BACKGROUND: Chronic hepatitis B virus (HBV) infection remains a serious public health challenge, and primary treatment strategies are unable to cure HBV infection due to the persistence of HBV covalently closed circular DNA (cccDNA) in the nuclei of cells. The HBV X protein (HBx) plays a crucial role in regulating cccDNA transcription. Therefore, targeting HBx to identify host proteins that regulate cccDNA transcription could represent a curative approach. METHODS: Here, we used co-immunoprecipitation (co-IP) and mass spectrometry (MS) to identify proteins that interact with HBx. The candidat

Ubiquitin-specific protease 5 promotes EV-A71 replication by de-ubiquitinating MAVS and IRF3.

Human enterovirus A71 (EV-A71) is a major causative agent of hand, foot and mouth disease (HFMD), which poses a significant public health threat, particularly among young children. Mitochondrial antiviral signaling protein (MAVS) and interferon regulatory factor 3 (IRF3) are vital proteins for the induction of type I interferons (IFN-I) and downstream interferon-stimulated genes (ISGs) during EV-A71 infection. While posttranslational modifications are known to critically influence viral infection processes, the mechanisms by which EV-A71 exploits host deubiquitinases (DUBs) for immune evasion

Silencing of ubiquitin specific protease-15 activates components of innate immune response and inhibits dengue virus infection.

Innate immune responses induced by interferons are one of the key defense mechanisms against virus infections. It is activated by specific sensors in host cells that detect particular features of viruses. Secretion of interferons stimulate interferon-stimulated genes (ISGs) that act as antiviral effectors and inhibit virus infections. Many factors, including post-translational modifications (PTMs) of host cell proteins modulate virus infections. Ubiquitination/deubiquitination is one such PTM that utilizes ligases and deubiquitinases, which viruses exploit to enhance their replication and evad

Adaptor protein complex 1 gamma 1 subunit is an important host factor involved in both Zika virus and dengue virus infections.

Mosquito-borne flaviviruses, such as Zika virus (ZIKV) and dengue virus (DENV), cause diverse severe clinical manifestations including fever, rash, hepatitis, arthralgia, and congenital anomalies. Here, we identified a host factor, the adaptor protein complex 1 gamma 1 subunit (AP1G1), which plays an important role in both ZIKV and dengue virus 2 (DENV2) infections. We explored the role of AP1G1 in ZIKV and DENV2 infections using CRISPR/Cas9 gene editing technology and RNA interference (RNAi) techniques. Knockout or silencing of AP1G1 decreases the replication of ZIKV and DENV2 in multiple hum

Cellular transcription factor TFII-I represses adenovirus gene expression.

To successfully replicate, viruses must overcome innate cellular antiviral responses. With human adenovirus (HAdV), a key viral repressor of cellular antiviral responses is the early protein E4ORF3. E4ORF3 relocalizes numerous cellular antiviral proteins, particularly those involved in the interferon (IFN) and DNA damage response (DDR) pathways, to sequester them from viral replication sites in the nucleus. E4ORF3 also directs SUMO modification of a subset of its targets, some of which are subsequently targeted for proteasomal degradation. We previously identified TFII-I, a cellular transcript

Host cell and viral protease targets of human SERPINs identified by in silico docking.

Serine protease inhibitors (SERPINs) are involved in various physiological processes and diseases, such as inflammation, cancer metastasis, and neurodegeneration. Their role in viral infections is poorly understood, as their expression patterns during infection and the range of proteases they target have yet to be fully characterized. Here, we show widespread expression of human SERPINs in response to respiratory virus infections, both in bronchioalveolar lavages from COVID-19 patients and in polarized human airway epithelial cultures. Using in silico docking of 10 SERPINs to 48 host proteases

Restriction of HIV-1 infectivity by interferon and IFITM3 is counteracted by Nef.

The viral accessory protein Nef is a major determinant of HIV-1 pathogenicity in vivo. Nef is a multifunctional, immunomodulatory protein that downmodulates cell surface proteins, including CD4 and MHC class I (MHC-I), which are important for T cell-mediated immunity. In addition, Nef also regulates cell-intrinsic immunity-Nef boosts the infectivity of virions produced and released from HIV-infected cells, at least in part, by counteracting the antiviral activity of transmembrane proteins SERINC3 and SERINC5. Here, we show that Nef proteins derived from many primary isolates of HIV-1 restore i

Chlamydia trachomatis disrupts host metabolism in primary cervical epithelial cells.

Chlamydia trachomatis is a major cause of bacterial sexually transmitted infections and reproductive complications. Despite progress in understanding C. trachomatis pathogenesis, studies using primary cells remain limited. Here, we investigated C. trachomatis infection of cervical epithelial cells and its impact on host cellular processes. Transcriptomic profiling show that C. trachomatis infection in cervical epithelial cells leads to significant changes in host cellular processes, including modulation of immune signaling, attenuation of DNA repair and proliferation-linked signaling pathways,

The cGAS-STING signaling pathway in the regulation of pulmonary infections: a systematic review.

The lungs are constantly exposed to airborne pathogens and depend on robust innate immune surveillance for protection. The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling pathway, a core component of the innate immune system, plays a pivotal role in defending against respiratory infections caused by viruses, bacteria, and mycobacteria, including Mycobacterium tuberculosis. Dysregulation of this pathway has been linked to several chronic lung diseases, such as cystic fibrosis, chronic obstructive pulmonary disease (COPD), idiopathic pulmonary fibrosis, and asthma

IL-32 positively regulates the AEBP1-IκBα-NF-κB-TNF-α axis to inhibit Mycobacterium tuberculosis infection in human macrophages.

Mycobacterium tuberculosis (MTB), the causative agent of tuberculosis, survives in host macrophages, the primary effector and antigen-presenting cells involved in the host immune response. We previously showed that Interleukin-32 (IL-32) is significantly increased in the peripheral blood plasma of tuberculosis patients, can act as an anti-MTB agent. However, the underlying molecular mechanism for its effect remains unknown. Here, we showed that inhibiting IL-32 with monoclonal antibody increases MTB loads that was positively correlated with higher antibody concentrations and longer exposure ti

Mycobacteria trehalose dimycolate interactions with host Mincle remodel blood-brain barrier junctions for brain invasion.

Tuberculous meningitis is unique among bacterial meningitides because it occurs in two temporally separated steps: mycobacteria first invade the brain, then form infected macrophage aggregates called Rich foci, which later erode the meninges. Here, using transparent zebrafish larvae, we detail the first step-brain invasion. We find that whereas elsewhere in the body mycobacteria disseminate within phagocytes, only extracellular mycobacteria reach the brain microvasculature. There, they adhere to the microvascular endothelium and grow into microcolonies. These microcolonies induce endothelial t

Shigella type-III secretion system effectors counteract the induction of host inflammation and cell death.

Many enteric bacterial pathogens deliver virulence effectors to counteract host innate immune responses, such as inflammation and cell death, and colonize the intestinal epithelium. However, host cells recognize the disruption of their innate immune signaling by bacterial effectors and induce alternative immune responses, collectively termed "effector-triggered immunity", to clear bacterial pathogens. Here, we describe a mechanism of cell death induction via effector-triggered immunity and the bacterial countermeasures of the pathogen Shigella flexneri. Shigella delivers the OspI effector to i

The emergence of multidrug-resistant Proteus vulgaris infection in cage reared Pangasianodon hypophthalmus: Molecular characterization and host-pathogen response.

The study investigates the causative agent responsible for massive mortality in Pangasianodon hypophthalmus cage farms. The infected pangasius were lethargic, not taking feed, and had exophthalmia, deep ulceration, and hemorrhage on the ventral body surface. Pathogens were isolated from infected pangasius tissue samples, and the strain was preliminarily identified as Proteus vulgaris based on morphology, biochemical tests, 16S rRNA PCR sequencing, and phylogenetic analysis. The microbiological analysis revealed that P. vulgaris was associated with the disease outbreak and mortality of P. hypop

Ubiquitin-Mediated Effects on Oncogenesis during EBV and KSHV Infection.

The Herpesviridae include the Epstein-Barr Virus (EBV) and the Kaposi Sarcoma-associated Herpesvirus (KSHV), both of which are oncogenic gamma-herpesviruses. These viruses manipulate host cellular mechanisms, including through ubiquitin-mediated pathways, to promote viral replication and oncogenesis. Ubiquitin, a regulatory protein which tags substrates for degradation or alters their function, is manipulated by both EBV and KSHV to facilitate viral persistence and cancer development. EBV infects approximately 90% of the global population and is implicated in malignancies including Burkitt lym

Signaling events driving Aspergillus fumigatus-induced eosinophil activation.

Allergic bronchopulmonary aspergillosis is an incurable disease caused by the environmental mold Aspergillus fumigatus. This hypersensitivity pneumonia is characterized by an inflammatory type 2 immune response, accompanied by influx of eosinophils into the lung. To investigate the mode of action of eosinophils and the signaling events triggered by A. fumigatus, we used an in vitro coculture system of murine bone marrow-derived eosinophils confronted with conidia. Using small-molecule inhibitors, we identified signaling modules of eosinophils in the course of A. fumigatus confrontation. Eosino

Cryptococcus neoformans trehalose-6-phosphate synthase (tps1) promotes organ-specific virulence and fungal protection against multiple lines of host defenses.

Trehalose-6-phosphate synthase (TPS1) was identified as a virulence factor for Cryptococcus neoformans and a promising therapeutic target. This study reveals previously unknown roles of TPS1 in evasion of host defenses during pulmonary and disseminated phases of infection. In the pulmonary infection model, TPS1-deleted (tps1Δ) Cryptococci are rapidly cleared by mouse lungs whereas TPS1-sufficent WT (H99) and revertant (tps1Δ:TPS1) strains expand in the lungs and disseminate, causing 100% mortality. Rapid pulmonary clearance of tps1Δ mutant is T-cell independent and relies on its susceptibility

Functional genomics screens reveal a role for TBC1D24 and SV2B in antibody-dependent enhancement of dengue virus infection.

Under some conditions, dengue virus (DENV) can hijack IgG antibodies to facilitate its uptake into target cells expressing Fc gamma receptors (FcgR)-a process known as antibody-dependent enhancement (ADE) of infection. Beyond a requirement for FcgR, host dependency factors for this unusual IgG-mediated infection route remain unknown. To identify cellular factors exclusively required for ADE, here, we performed CRISPR knockout (KO) screens in an in vitro system poorly permissive to infection in the absence of IgG antibodies. Validating our approach, a top hit was FcgRIIa, which facilitates the
