Near-infrared-responsive silk/dopamine/fucoidan hydrogel incorporating multilayered silk-based nanoparticles as a reservoir for sequential and triggered NAC/NT delivery in diabetic wound healing.

Chronic diabetic wounds are characterized by excessive oxidative stress and impaired angiogenesis, presenting significant challenges for effective treatment. Herein, we developed a near-infrared (NIR)-responsive reservoir hydrogel (SD-NFA@SD/FC) capable of sustained and sequential drug delivery to promote diabetic wound healing. Free fucoidan (FC) was incorporated into a silk fibroin/dopamine (SD) hydrogel for early antioxidant and anti-inflammatory effects. Neurotensin (NT) and acetylcysteine (NAC) were sequentially assembled onto SD nanoparticles (224.61 ± 2.89 nm; zeta potential 1.36 ± 0.43

Microneedle patches deliver targeted GLP-1RAs-loaded nanoparticles for the treatment of rheumatoid arthritis.

Despite the strong immune-modulatory benefits, there has been a lag in using glucagon-like peptide-1 receptor agonists (GLP-1RAs) in rheumatoid arthritis (RA) due to inefficient articular delivery and poorly understood direct cartilage protection. In this paper, we addressed these critical issues through the mechanism-informed design of a hierarchical drug delivery system. We elucidated previously unnoted ways in which GLP-1RAs curbed RA pathology via direct suppression of the cGAS-STING signaling pathway to inhibit chondrocyte ferroptosis and concomitantly induced macrophage M2 polarization.

Smart Stings of Nature: Harnessing Microneedles Assisted Targeted Phytoconstituent Delivery for Dermatological Disorders.

Microneedle-mediated transdermal delivery of phytoconstituents represents a sophisticated and emerging paradigm in dermatological therapeutics, bridging the pharmacological efficacy of plant-derived bioactive with the precision of advanced biomedical engineering. Conventional topical dosage forms are often constrained by the formidable barrier function of the stratum corneum, instability of phytochemicals, and suboptimal patient adherence. Microneedle arrays circumvent these limitations by creating transient microchannels that enable minimally invasive, painless, and controlled delivery of the

Fibrous Biomaterial Scaffold for Tympanic Membrane Repair: Microarchitectural Engineering and Structure Function Performance.

Tympanic membrane (TM) perforations, arising from infections, injuries, or chronic otitis media, remain a frequent clinical finding and can lead to hearing problems when the tissue does not regenerate adequately. Although autologous grafts are still the standard option for repairing persistent defects, they come with well-known limitations. Beyond the need for additional harvesting procedures, these grafts rarely reproduce the intricate, fibrous layering of the native TM, which can compromise sound transmission after healing. In search of alternatives, fibre-based scaffolds have attracted cons

Bioinspired Unidirectional 3D Scaffold for Synergistic Enhanced Solar Evaporation and Efficient VOCs Degradation.

3D design has effectively enhanced photothermal materials water evaporation performance via utilizing side surfaces. However, for photothermal/photocatalytic composite evaporators which can effectively degrade volatile organic compounds (VOCs) to prevent their contamination in condensate, introducing 3D design should promote VOCs escaping from the evaporator sides surface, reducing efficiency of VOCs photocatalytic degradation. Herein, a bioinspired unidirectional silk fibroin (SF)-Ag2S/Ag3PO4 3D evaporator is reported to achieve synergy between enhancing water evaporation and maintaining VOCs

In Vivo and In Vitro Study of a Multifunctional SF/nHAp Corrosion-Resistant Bio-Coating Prepared on MAO Magnesium Alloy via Ultrasonic Spraying.

Magnesium alloys are often used in bone repair surgeries due to their biodegradability and excellent elastic modulus, making them a promising alternative to traditional nondegradable implants like titanium alloys. However, their rapid degradation rate limits their use as implants in the body. To enhance the corrosion resistance and bioactivity of magnesium alloys, we applied an ultrasonic spray coating on microarc oxidized (MAO) AZ31 magnesium alloy, using a mixture of silk fibroin (SF) and nanohydroxyapatite (nHAp). This SF/nHAp composite embeds directly into the micropores on the MAO-treated

Biomimetic 3D-printed bilayered scaffold with stem cell delivery for rabbit vaginal regeneration.

Traditional biological scaffolds for vaginal reconstruction often fail to restore the physiological bilayered structure of vaginal tissue, leading to post-operative complications such as contraction, stenosis, and fibrosis. To address this, we developed a biomimetic bilayered vaginal scaffold using decellularized porcine vaginal extracellular matrix (vECM), gelatin methacryloyl (GelMA), and silk fibroin (SF) hydrogels. The optimized bioink (vECM/GelMA/SF) was characterized via SEM, rheology, FTIR, swelling tests, and H&E staining, confirming its morphology, stability, and biocompatibility. Rab

Adipose-derived mesenchymal stem cells inhibit hepatic stellate cells activation to alleviate liver fibrosis via Hippo pathway.

BACKGROUND: Liver fibrosis is a common pathological process of chronic liver disease, characterized by excessive deposition of extracellular matrix (ECM). Mesenchymal stem cells (MSCs) have been found to have potential therapy effect on liver fibrosis, but the mechanism involved was still unclear. The objective of this study is to investigate the therapeutic efficacy of adipose-derived mesenchymal stem cells (ADMSCs) on the treatment of liver fibrosis, with particular emphasis on elucidating the underlying mechanism of action through which ADMSCs inhibit the activation of hepatic stellate cell

Selenium-modified bone cement promotes osteoporotic bone defect repair in ovariectomized rats by restoring GPx1-mediated mitochondrial antioxidant functions.

Over-accumulation of reactive oxygen species (ROS) causes mitochondrial dysfunction and impairs the osteogenic potential of bone marrow-derived mesenchymal stem cells (BMMSCs). Selenium (Se) protects BMMSCs from oxidative stress-induced damage; however, it is unknown whether Se supplementation can promote the repair of osteoporotic bone defects by rescuing the impaired osteogenic potential of osteoporotic BMMSCs (OP-BMMSCs). In vitro treatment with sodium selenite (Na2SeO3) successfully improved the osteogenic differentiation of OP-BMMSCs, as demonstrated by increased matrix mineralization and

Enhancing Osseointegration of TC4 Alloy by Surficial Activation Through Biomineralization Method.

Titanium (Ti) alloys have been applied to biomedical implants for a long time. Although Ti alloys are biocompatible, efforts have been continuously made to improve their bone conductivity and osteogenesis for enhancing their performance. Silk fibroin (SF) is a natural biomaterial with excellent biomedical and mechanical properties, and hydroxyapatite (HAP) nanocomposites derived from SF are promising for producing "artificial bone" owing to their biomedical applicability and strong mechanical functions. Therefore, we built an SF coating on the surface of Ti-6Al-4V alloy, and then the incubated

Development of Dual Neurotrophins-Encapsulated Electrosupun Nanofibrous Scaffolds for Peripheral Nerve Regeneration.

Nerve growth factor (NGF) is widely used for repairing peripheral nerve injury because of its capability in dominating the survival, migration, proliferation, and differentiation of nerve cells. Monosialoganglioside (GM1), as another kind of nerve growth factor, works for regulating NGF function. In this study, GM1 and NGF were incorporated into the Poly(l-lactic acid-co-ε-caprolactone)/silk fibroin (PLCL/SF) nanofibers by the coaxial electrospinning. The fibers morphology and core–shell structure were characterized by SEM and TEM. The scaffolds demonstrated high tensile stress with good flexi

Wounds refer to physical injuries in which the integrity of the skin or other body organs is disturbed. Wound care includes proper management and treatment of the injuries to promote healing while avoiding infection. Here, a core-shell scaffold is developed comprising polyethylene glycol/silk fibroin-chitosan nanoparticles loaded with curcumin. Chitosan nanoparticles and PEG/Silk fibrous scaffold were synthesized by a microfluidic system and electrospinning technique, respectively. TEM, DLS, and FTIR techniques were used to examine the nanoparticles; whereas nanofibers were characterized by SE

Study on the microstructure of African wild silk cocoon shells and fibers.

Silk fibers and cocoon shells from four African wild silkmoths Gonometa postica, Anaphe panda, Argema mimosae and Epiphora bauhiniae-were studied to gain insight into the structure-property-function relations and potential commercial application. The surface and cross-section of cocoon shells and fibers revealed the presence of prominent structural variations. Cocoon shells were multilayered and porous structures constructed from highly cross-linked fibers that are densely packed within the sericin/gum. Fibers had fibrillar sub-structures running along the fiber axis and with greater number an

Approaches to scarless burn wound healing: application of 3D printed skin substitutes with dual properties of anti-infection and balancing wound hydration levels.

BACKGROUND: Severe burn wounds face two primary challenges: dysregulated cellular impairment functions following infection and an unbalanced wound hydration microenvironment leading to excessive inflammation and collagen deposition. These results in hypertrophic scar contraction, causing significant deformity and disability in survivors. METHODS: A three-dimensional (3D) printed double-layer hydrogel (DLH) was designed and fabricated to address the problem of scar formation after burn injury. DLH was developed using methacrylated silk fibroin (SFMA) and gelatin methacryloyl (GelMA) for the up

Polymers for Pelvic Floor Repair: Current State and Future Prospects.

IMPORTANCE: Surgical repair of pelvic organ prolapse and stress urinary incontinence frequently relies on polymeric biomaterials in sutures, meshes, and membranes to provide structural support during tissue healing and beyond. Polypropylene has long served as the default material owing to its strength, durability, and processability, but the chronic foreign body response and mechanical mismatch between stiff implants and viscoelastic pelvic tissue have been linked to complications, including mesh contraction, exposure, and dyspareunia. These limitations, together with regulatory actions on tra
