Prioritized memory access explains planning and hippocampal replay.

To make decisions, animals must evaluate candidate choices by accessing memories of relevant experiences. Yet little is known about which experiences are considered or ignored during deliberation, which ultimately governs choice. We propose a normative theory predicting which memories should be accessed at each moment to optimize future decisions. Using nonlocal 'replay' of spatial locations in hippocampus as a window into memory access, we simulate a spatial navigation task in which an agent accesses memories of locations sequentially, ordered by utility: how much extra reward would be earned

Neurocognitive effects of stress in higher education: Mechanisms, challenges, and opportunities.

Stress is a key modulator of cognition with important consequences for learning and memory, particularly in university students. This systematic review integrates evidence from neuroscience, psychology, and selected theoretical contributions to clarify how acute and chronic stress shape cognitive processes relevant to academic performance. Literature published between 2000 and 2025 was examined using PRISMA-aligned procedures, prioritizing empirical studies while drawing on foundational works to contextualize concepts. Across studies, moderate stress levels may facilitate memory consolidation,…

The Zinc-sensing Receptor (GPR39) Modulates Declarative Memory and Age-related Hippocampal Gene Expression in Male Mice.

As a neuromodulator, zinc regulates synaptic plasticity, learning and memory. Synaptic zinc is also a crucial factor in the development of toxic forms of amyloid beta protein and, subsequently, of Alzheimer's dementia (AD). Therefore, efforts to pinpoint mechanisms underlying zinc-dependent cognitive functions might aid AD research, by providing potential novel targets for drugs. One of the most understudied proteins in this regard is a zinc-sensing metabotropic receptor: GPR39. In this study we investigated the impact of GPR39 knock-out (KO) on age-related memory decline in mice of both sexes

Tracing melanin-concentrating hormone function: decades of memory research and emerging links to Alzheimer's disease.

Melanin-concentrating hormone (MCH) is a hypothalamic neuropeptide classically implicated in the regulation of energy homeostasis, sleep-wake cycle and motivated behaviors. Over the last 30 years, an increasing amount of evidence has revealed a complex, non-linear role in memory processing. In this mini-review, we summarize experimental findings on the multifaceted influence of MCH on memory, highlighting how this neuropeptide can promote consolidation by improving synaptic efficacy, induce the forgetting of hippocampus-dependent memories during REM sleep or negatively regulate memory formatio

Astrocyte and Neuronal Panx1 Support Long-Term Reference Memory in Mice.

Pannexin 1 (Panx1) is an ubiquitously expressed protein that forms plasma membrane channels permeable to anions and moderate-sized signaling molecules (e.g., ATP, glutamate). In the nervous system, activation of Panx1 channels has been extensively shown to contribute to distinct neurological disorders (epilepsy, chronic pain, migraine, neuroAIDS, etc.), but knowledge of the extent to which these channels have a physiological role remains restricted to three studies supporting their involvement in hippocampus dependent learning. Given that Panx1 channels may provide an important mechanism for a

The relationship between hippocampal volumes and nonverbal memory in patients with medial temporal lobe epilepsy.

OBJECTIVE: To explore the involvement of medial temporal lobe structures such as the hippocampus, amygdala, and entorhinal cortex (EC) in memory consolidation by volumetric magnetic resonance imaging (MRI). METHODS: Sixty-two consecutive patients with medial temporal lobe epilepsy (MMTLE) were assessed using the Clinical Memory Scale (CMS) and MRI to measure the volumes of the hippocampus, amygdala, and EC. Participants were grouped according to MRI findings into 3 groups: left MRI-positive (abnormal hippocampal formation on the left side; n=17), right MRI-positive (abnormal hippocampal forma…

Visual areas become less engaged in associative recall following memory stabilization.

Numerous studies have focused on changes in the activity in the hippocampus and higher association areas with consolidation and memory stabilization. Even though perceptual areas are engaged in memory recall, little is known about how memory stabilization is reflected in those areas. Using magnetoencephalography (MEG) we investigated changes in visual areas with memory stabilization. Subjects were trained on associating a face to one of eight locations. The first set of associations ('stabilized') was learned in three sessions distributed over a week. The second set ('labile') was learned in o

Phase similarity between similar objects indicates representational merging across retrieval training but not sleep.

Retrieval training (i.e., cued recall) is theorised to induce rapid memory consolidation, similarly to sleep. Across consolidation, related neural representations become increasingly similar; yet, representational change has never been directly compared between sleep and retrieval training to test similarities in their underlying mechanisms. In this study, 30 subjects (27F, 18-34, M = 22.17) completed 4 separate sessions in which they (1) learnt object-word pairs, followed by (2) immediate recognition testing, (3) one of four 120-min interventions (retrieval training, restudy, sleep, or wake),…

Acute Restraint Stress Induces Long-Lasting Synaptic Enhancement by Inhibiting AMPK Activation in AD Model Mice.

BACKGROUND: Alzheimer's disease (AD) is characterized by a gradual synaptic loss. The progression of AD severely affects late-phase long-term potentiation (L-LTP), which is essential for long-term memory consolidation. AIM: We have previously demonstrated the beneficial effects of acute restraint stress (ARS) on hippocampal LTP in AD mouse models. This study aimed to verify the effects and potential mechanisms of ARS on the maintenance of hippocampal L-LTP in two AD mouse models. MATERIALS AND METHODS: 5xFAD and Tg2576 mice underwent a 30-min body immobilization protocol to induce ARS, follo…

Visual Training Induced Temporo-Occipital Fast Sleep Spindle Clustering in Humans Revealed by Full-Night HD-EEG Recordings.

This study investigates the impact of extensive visual procedural training on the temporal organisation of sleep spindles in healthy young a … ted temporo-occipital regions. These results might be interpreted in the context of sleep-dependent memory consolidation or potentially by homeostatic-related processes following extensive training. Moreover, here we illustrate that spindle reorganisation occurs not only in motor tasks but also in visual learning. The absence of age-related differences indicates that …

Social fear memory as a framework for understanding social defeat stress and psychopathology.

Social defeat is a potent stressor in both rodents and humans that, under conditions of chronic or severe stress, can lead to persistent social avoidance. This review synthesizes evidence supporting the view that social defeat promotes the formation of social fear memory through associative learning mechanisms that parallel classical fear conditioning. Drawing on preclinical models, we describe how the context of social defeat functions as a conditioned stimulus, while the experience of defeat serves as an unconditioned stimulus, giving rise to fear-related behaviors such as freezing and socia

Single prolonged stress: toward an animal model of posttraumatic stress disorder.

Although selective serotonin reuptake inhibitors (SSRIs) are reported to be effective in decreasing posttraumatic stress disorder (PTSD) symptoms, a subgroup of PTSD patients remain chronically symptomatic and maintain conditioned fear responses to traumatic stimuli. In this context, the establishment of an appropriate animal model of PTSD is necessary to promote better understanding of the mechanisms of the disorder and to facilitate the development of more effective therapeutic alternatives to SSRIs. Although no single widely accepted animal model of PTSD has been established to date, the si

Rapid eye movement sleep: Who needs it? Creativity mechanisms and psychiatric applications of REM sleep enhancement.

Rapid eye movement sleep (REMS) has historically been associated with anecdotal 'creative insights', possibly due to the fantastical and ostensibly illuminating nature of its associated phenomena (dreams). REMS, characterised by rapid eye movements, muscle atonia, and high-energy neuronal activity, has been linked to memory consolidation and information processing, particularly regarding the formation of novel associations or reintegration of consolidated memories into new cognitive networks. However, studies in these domains have largely used methodology which deprived subjects (animal or hum…

Hydrogen sulfide in the brain as a silent neuroprotector in Alzheimer's disease.

Hydrogen sulfide (H2S) is an endogenously produced gasotransmitter that has garnered growing attention for its critical roles in cellular signalling and brain function. It regulates NMDA receptors during long-term potentiation, a fundamental mechanism underlying memory consolidation and influences neurotransmission and essential neurophysiological functions. H2S is synthesized by three enzymes: cystathionine γ-lyase (CSE) and cystathionine β-synthase (CBS) and 3-mercaptopyruvate sulfurtransferase (MST) within the cell. CBS is suggested to be the primary source of H2S in the brain parenchyma, w…

Rodents' episodic-like memory: concepts, ageing, neurodegeneration, future.

Episodic memory (EM), traditionally defined as the conscious recollection of what, where, and when events occurred, was long considered uniquely human due to its reliance on autonoetic awareness. However, behavioural criteria allow memory assessment based on observable features, enabling the study of EM-related processes in non-human species and leading to the concept of episodic-like memory (ELM) in animal models. We review conceptual advances and methodological challenges in ELM research, focusing on rodent models that provide mechanistic insights into neural circuits and molecular pathways
