Study on the role and function of RRS1 in cutaneous squamous cell carcinoma.

RRS1 (regulator of ribosome synthesis 1) has been identified as a critical factor in the proliferation, metastasis, and invasion of some cancer's cells. In this study, immunohistochemical analysis and GEO database studies demonstrated significantly higher RRS1 expression in cutaneous squamous cell carcinoma(cSCC) tissues and neck squamous cell carcinoma (HNSCC) compared to normal skin. To further investigate RRS1's role, a lentiviral transduction method was used to establish an RRS1 knockdown model in A431 cells. Following RRS1 knockdown, MTT, Celigo scratch, and Transwell assays revealed a si

Trk Signaling Inhibition Reduces cSCC Growth and Invasion in In Vitro and Zebrafish Models and Enhances Photodynamic Therapy Outcome.

Cutaneous squamous cell carcinoma (cSCC) is the second most common skin cancer, with a rising global incidence. Neurotrophins (NTs) and their receptors, including TrkA and CD271, play key roles in epidermal homeostasis and tumor progression. We showed that CD271 expression and function are critical for low- to high-risk progression of cSCC, while TrkA is highly expressed in poorly differentiated tumors. Although NTRK fusions are recognized as oncogenic drivers, the functional impact of TrkA signaling in cSCC remains underexplored. In this study, we investigated the effects of TrkA inhibition,

In Silico Screening of Phytochemicals as Potential Inhibitors of the JAK/STATs Pathway in Psoriasis.

The skin is a dynamic tissue that consists of different layers such as stratum corneum, the site for keratinocyte development and maturation for the natural changeover of skin. In psoriasis, this natural development of keratinocytes gets disturbed and aggregation of nucleated keratinocytes takes place in the epidermis of the skin, leading to the presence of scaly skin, which makes the patient physically, socially, and psychologically ill. Various natural, semisynthetic, and synthetic treatments are available. Still, semisynthetic or synthetic are mainly used to treat psoriasis with side effect

Growth hormone-releasing hormone receptor (GHRH-R) and its signaling.

The hypothalamic polypeptide growth hormone-releasing hormone (GHRH) stimulates the secretion of growth hormone (GH) from the pituitary through binding and activation of the pituitary type of GHRH receptor (GHRH-R), which belongs to the family of G protein-coupled receptors with seven potential membrane-spanning domains. Various splice variants of GHRH-R (SV) in human neoplasms and other extrapituitary tissues were demonstrated and their cDNA was sequenced. Among the SVs, splice variant 1 (SV1) possesses the greatest similarity to the full-length GHRH-R and remains functional by eliciting cAMP

Limited Proteolysis as a Regulator of Lymphatic Vessel Function and Architecture.

Recent advances have highlighted the multifaceted roles of the lymphatic vasculature in immune cell trafficking, immunomodulation, nutrient transport, and fluid homeostasis. Beyond these physiological functions, lymphatic vessels are critically involved in pathologies such as cancer metastasis and lymphedema, rendering their structural and functional regulation of major interest. Emerging evidence suggests that limited proteolysis is a key regulatory mechanism for lymphatic vascular function. In dyslipidemic conditions, dysregulated calpain activity impairs lymphatic trafficking and destabiliz

Targetable pathways for drug repurposing in gastric cancer.

Gastric cancer (GC) is both the fifth most common cancer worldwide and the fifth in mortality. Owing to a lack of symptoms in the early stages and unspecific clinical presentation in the later stages, GC is usually diagnosed at advanced stages. This means that only approximately 60% of patients are eligible for curative treatment, and overall, GC patients have a 5-year survival rate of only 28.3%, underscoring the importance of developing new treatment strategies. Drug repurposing involves identifying new therapeutic uses for approved drugs and is a promising strategy for cancer treatment beca

Discovery of the roles of SmMAPK4 in regulating tanshinone biosynthesis in Salvia miltiorrhiza.

Tanshinones, the primary bioactive components found in Salvia miltiorrhiza, are widely utilized in the treatment of cardiovascular diseases; however, the molecular mechanisms that govern their biosynthesis remain unclear at the protein phosphorylation level. The proteomic profiles of roots, stems, leaves, and flowers of S. miltiorrhiza identified a total of 8301 proteins. A novel MITOGEN ACTIVATED PROTEIN KINASE 4 (SmMAPK4) was identified to be positively correlated with the accumulation of tanshinones in S. miltiorrhiza through tissue-specific metabolomics and proteomics analyses. Meanwhile,

p38γ modulates ferroptosis in brain injury caused by ethanol and cerebral ischemia/reperfusion by regulating the p53/SLC7A11 signaling pathway.

Ischemic stroke, a neurological condition with a complicated etiology that is accompanied by severe inflammation and oxidative stress, and ethanol (EtOH) may aggravate ischemia/reperfusion (I/R)-induced brain damage. However, the effect of prolonged alcohol intake on acute brain injury remains ambiguous. As part of the mitogen-activated protein kinase (MAPK) family, p38γ is involved in ferroptosis and inflammation in various diseases. This study explored how p38γ is involved in the effects of chronic EtOH consumption and brain injury caused by cerebral I/R. Brain damage was induced in the mice

A toxic gain-of-function variant in MAPK8IP3 provides insights into JIP3 cellular roles.

Mitogen-activated protein kinase 8 interacting protein 3 (MAPK8IP3) gene encoding a protein called JIP3 is an adaption protein of the kinesin-1 complex known to play a role in axonal transport of cargo. Mutations in the gene have been linked to severe neurodevelopmental disorders, resulting in developmental delay, intellectual disability, ataxia, tremor, autism, seizures, and visual impairment. A patient who has a missense mutation in the MAPK8IP3 gene (c. 1714 C>T, Arg578Cys) (R578C) manifests dystonia, gross motor delay, and developmental delay. Here, we showed that the mutation was a toxic

Rhodium(III)-Nitroxyl Radical Complex Triggers Dual-Pronged Disulfidptosis-Apoptosis in Hepatocellular Carcinoma via Metabolic Sabotage and Redox Catalysis.

Hepatocellular carcinoma (HCC) resists apoptosis-targeting therapies, necessitating the development of agents targeting alternative cell death pathways. Here, we report the discovery of a nitroxyl radical-conjugated Rh(III) complex (OG-Rh) that triggers dual disulfidptosis and apoptosis via synergistic metabolic sabotage and redox catalysis. OG-Rh inhibited glucose uptake, depleted NADPH, and induced disulfidptosis, a novel disulfide-stress-mediated death, by inducing actin cytoskeleton collapse via pathogenic disulfide over-crosslinking. Simultaneously, its tumor-selective superoxide dismutas

FGFR antagonists restore defective mandibular bone repair in a mouse model of osteochondrodysplasia.

Gain-of-function mutations in fibroblast growth factor receptor (FGFR) genes lead to chondrodysplasia and craniosynostoses. FGFR signaling has a key role in the formation and repair of the craniofacial skeleton. Here, we analyzed the impact of Fgfr2- and Fgfr3-activating mutations on mandibular bone formation and endochondral bone repair after non-stabilized mandibular fractures in mouse models of Crouzon syndrome (Crz) and hypochondroplasia (Hch). Bone mineralization of the calluses was abnormally high in Crz mice and abnormally low in Hch mice. The latter model presented pseudarthrosis and i

Neuronal p38 MAPK Signaling Contributes to Cisplatin-Induced Peripheral Neuropathy.

This study investigates the role of p38 mitogen-activated protein kinase (MAPK) activation in dorsal root ganglion (DRG) neurons in the development and progression of chemotherapy-induced peripheral neuropathy (CIPN). This research evaluates whether inhibiting activation of p38 MAPK could reduce neuropathic outcomes in a transgenic breast cancer mouse model (C3TAg) and wild-type mice (FVB/N) treated with cisplatin. Cisplatin treatment stimulated p38 MAPK phosphorylation and nuclear translocation in DRG neurons. Neflamapimod, a specific inhibitor of p38 MAPK alpha (p38α), proven to be safe in c

E3 Ubiquitin Ligase TRIM21 Exacerbates Pathological Cardiac Hypertrophy Through ASK1 K63-Linked Polyubiquitination.

Pathological cardiac hypertrophy is a major risk factor for myocardial ischemia, heart failure, and sudden cardiac death, yet its underlying mechanisms remain incompletely understood. Here, we identify Tripartite Motif Containing 21 (TRIM21) as a novel driver of pathological cardiac hypertrophy. TRIM21 was significantly upregulated in mouse hearts following transverse aortic constriction (TAC) and in neonatal rat cardiomyocytes stimulated with phenylephrine (PE). In vitro, TRIM21 knockdown attenuated PE-induced cardiomyocyte hypertrophy, whereas TRIM21 overexpression exacerbated this effect. C
