The potential roles of intraovarian peptides in normal and abnormal mechanisms of reproductive physiology.

Normal and abnormal follicular growth and steroidogenesis depend on gonadotropins as well as intraovarian peptide and polypeptide growth factors, which may mediate or potentiate gonadotropin action. Epidermal growth factor is mitogenic to ovarian granulosa and is a potent inhibitor of granulosa aromatase. It may be involved in the apparent arrest of follicular development commonly seen in women with polycystic ovarian syndrome as well as in the blunted response to gonadotropins seen in this syndrome. Insulin-like growth factors are also mitogenic to ovarian granulosa, but in contrast to epider

Genetic risk factors for perinatally acquired HIV-1 infection.

This study evaluates genetic influence on susceptibility to perinatal HIV-1 infection among 106 Black infants from New York and San Francisco born to mothers infected with HIV-1. Genes tested by molecular techniques are HLA class II loci DRB1, DPB1 and DQA1; HLA class III loci complement C4A and C4B; alpha and beta interferons; and the constant region of the T-cell receptor beta chain. Of the 106 infants analysed, 54 are infected with HIV and 52 remain uninfected at age 15 months and older. Genotypes in the HLA region appear to influence risk of HIV infection. Specifically, infants with the am

A regulatory system for soluble immune response suppressor production in steroid-responsive nephrotic syndrome.

Patients with nephrotic syndrome frequently have suppressed immune responses. Previously, it has been determined that subjects with steroid-responsive nephrotic syndrome (SRNS) produce the lymphokine, soluble immune response suppressor (SIRS). In the present group of experiments, a potential pathway of suppressor cell activation was investigated. Sera from patients with SRNS stimulated normal CD8+ lymphocytes to produce SIRS. Serum SIRS-inducing activity was abrogated by treatment with proteinase K or boiling, but was not affected by dialysis, acidification to pH 2, or heating to 56 degrees C.

Chronic hepatitis: pathogenesis and treatment.

Current therapies for chronic viral hepatitis, autoimmune "lupoid" chronic active hepatitis, and drug-induced chronic hepatitis are discussed in the context of recent advances in our understanding of the pathophysiology of chronic active liver disease. Accurate diagnosis is the cornerstone of proper treatment; the limitations and pitfalls of conventional techniques are discussed. Current theories of the pathogenesis of chronic hepatitis B are reviewed to provide a framework for the use of antiviral drugs. Data from the early results of therapy with adenine arabinoside, acyclovir, and immunomod

[Treatment of various malignancies with recombinant IFN-gamma (S-6810). The IFN-gamma Study Group].

Recombinant interferon-gamma (rIFN-gamma) was given to patients with malignancies by continuous daily administration or by intermittent high doses. Local administration was performed for patients with skin malignancy, and bladder and hepatic carcinoma. Among 239 cases eligible for evaluation (144 cases treated by systemic administration and 95 cases by local injection), complete response was observed in one case each of renal cell carcinoma, malignant lymphoma and mycosis fungoides, all treated by systemic administration. Intermittent high doses of rIFN-gamma induced a response rate of 21.4% i

Incidence and prevalence of salivary gland tumours in Valparaiso, Chile.

BACKGROUND/AIMS: A simple non-invasive tape (Sebutape) adsorption method was used to recover inflammatory proteins from normal and compromised human scalp (i.e. dandruff and seborrheic dermatitis) in order to assess the inflammatory and immunologic changes relevant to these clinical conditions. METHODS: The scalps of subjects identified by a dermatologist as having either dandruff (n = 18), seborrheic dermatitis (n = 19) or normal scalp (n = 16) were visually graded to obtain total adherent scalp flaking scores (TASFS). Sebutape samples were then collected from both the high and low TASFS sca

Generation of polarized antigen-specific CD8 effector populations: reciprocal action of interleukin (IL)-4 and IL-12 in promoting type 2 versus type 1 cytokine profiles.

We have generated primary effector populations from naive CD8 T cells in response to antigen and determined their patterns of cytokine secretion upon restimulation. The effect of exogenous factors on the effector generation was examined and compared with responses of antigen-specific CD4 effectors generated under comparable conditions. CD8 cells from bm1 mice were stimulated with C57BL/6 (B6) antigen presenting cells (APCs) bearing allogeneic class I and CD8 cells from female severe combined immunodeficiency (SCID) B6 mice, transgenic for a T cell receptor alpha/beta (TCR-alpha/beta) that reco

Kaposi's sarcoma: its 'oncogenes' and growth factors.

Viral genes capable of inducing vascular tumors in the skin of transgenic mice are the tat gene of HIV-1 and polyoma virus' middle T antigen gene. Instead of vascular tumors, the tat gene of HTLV-I causes thymic atrophy and mesenchymal tumors in transgenic mice. No proof exists that any of these genes contribute to the induction of KS but HIV-1 tat is a strong suspect. The gene product K-FGF of the oncogene K-fgf/hst (int) uses bFGF receptors, is homologous with bFGF and acts as a mitogen for fibroblasts, endothelial cells and melanocytes. The overexpression of the K-fgf gene in KS is not prov

Mice with type I interferon signaling deficiency are prone to epilepsy upon HSV-1 infection.

Viral encephalitis continues to be a significant public health concern. In our previous study, we discovered a lower expression of antiviral factors, such as IFN-β, STING and IFI16, in the brain tissues of patients with Rasmussen's encephalitis (RE), a rare chronic neurological disorder often occurred in children, characterized by unihemispheric brain atrophy. Furthermore, a higher cumulative viral score of human herpes viruses (HHVs) was also found to have a significant positive correlation with the unihemispheric atrophy in RE. Type I IFNs (IFN-I) signaling is essential for innate anti-infec

A proline deletion in IFNAR1 impairs IFN-signaling and underlies increased resistance to tuberculosis in humans.

Type I interferons (IFN), best known for their anti-viral functions, have been shown to impair host resistance to intracellular bacteria in mice. However, the precise role of type I IFN signaling in bacterial infection in humans is unclear. Here, we show that genetic variation in the human IFNAR1 gene is associated with decreased susceptibility to tuberculosis and an increased risk of viral hepatitis in Chinese populations. Receptor mutagenesis and cell signaling studies establish that the IFNAR1 mutation corresponding to a proline deletion in the hinge region of the membrane-proximal domain o

Chimeric viruses between Rocio and West Nile: the role for Rocio prM-E proteins in virulence and inhibition of interferon-α/β signaling.

Mosquito-transmitted flavivirus Rocio (ROCV) was responsible for an outbreak of encephalitis in the Ribeira Valley, located in the south coast of Sao Paulo State, Brazil, in 1975-1976. ROCV also causes fatal encephalitis in adult mice. Seroprevalence studies in humans, horses and water buffaloes in different regions of Brazil have suggested that ROCV is still circulating in the country, indicating the risk of re-emergence of this virus. West Nile virus (WNV) is also a mosquito-transmitted encephalitic flavivirus, however, WNV strains circulating in Australia have not been associated with outbr

Role of the suppressor of cytokine signaling 2 (SOCS2) during meningoencephalitis caused by Bovine herpesvirus 5 (BoHV-5).

The role of suppressors of cytokine signaling (SOCS) in meningoencephalitis caused by Bovine herpesvirus 5 (BoHV-5) was evaluated by intracranial infection in C57BL/6 wild-type mice (WT) and SOCS2 deficient mice (SOCS2(-/-)). Both infected groups presented weight loss, ruffled fur and hunched posture. Additionally, infected SOCS2(-/-) mice showed swollen chamfer and progressive depression. Infected WT animals developed mild meningitis, characterized by infiltration of mononuclear cells. Moreover, viral DNA was detected in liver and lung from infected WT group. This group also showed elevated b

Poly (I:C) downregulates platelet production and function through type I interferon.

Thrombocytopenia is a frequent complication of viral infections; the underlying mechanisms appear to depend on the identity of the virus involved. Previous research, including reports from our group, indicates that as well as having antiviral activity type I interferons (IFN I) selectively downregulate platelet production. In this study we extended understanding of the role of endogenous IFN I in megakaryo/thrombopoiesis by evaluating platelet and megakaryocyte physiology in mice treated with polyinosinic:polycytidylic acid [poly (I:C)], a synthetic analogue of double-stranded RNA, Toll-like r

Targeting ERO1A enhances antitumor immune response by recruiting CD8+T cells and synergizing with PD-L1 blockade in liver cancer.

Despite the availability of various treatment modalities for liver cancer, its mortality rate remains high. Immunotherapy represents an emerging approach for liver cancer. However, the response rate to single-agent immune checkpoint blockade therapy is suboptimal. Endoplasmic reticulum oxidoreductase 1-alpha (ERO1A), a potential therapeutic target associated with endoplasmic reticulum (ER) stress, is implicated in impeding protective antitumor immunity. In this study, we found that targeting ERO1A in liver cancer, while activating the PERK/EIF2α/ATF4/CHOP-mediated apoptosis and autophagy signa

Influenza A virus derived NS1 enhances translation of HPLC purified mRNA and interferon adjuvanted mRNA vaccination.

Non-structural protein (NS1) derived from influenza A virus is a potent immune evasion protein capable of enhancing translation of nucleoside-modified mRNA via host gene expression inhibition (HGEI) effect. In this study, we show that the HGEI effect on NS1 transfected cells reduces de novo synthesis and polyadenylation of host RNA. Co-delivering NS1 mRNA also enhances translation of non-inflammatory nucleoside-modified and HPLC-purified mRNA in NIH3T3 cells. Co-delivering NS1 mRNA also enhances translation of nucleoside-modified mRNA in DCs by 100% based on GFP expression, but completely inhi

Tuberculosis Diagnosis and Prevention in Children on Biological Treatment: Clinical Challenges and Considerations.

Tuberculosis (TB) remains a major global health concern, especially for children with rheumatic diseases on biologic therapies. While these agents have improved disease management and quality of life, they also pose new challenges, mainly an increased risk of TB, particularly with tumor necrosis factor‑alpha (TNF‑α) inhibitors like infliximab, adalimumab, and etanercept. Since TNF‑α plays a crucial role in granuloma formation and controlling Mycobacterium TB, its inhibition can allow latent TB infection (LTBI) to develop into active disease. To reduce this risk, screening for LTBI before start

Design, synthesis, characterization and exploration of anti-HBV activity of matrine-adenine hybrids.

Hepatitis B virus (HBV) is a pathogen that often causes progressive liver disease, leading to acute, chronic hepatitis, cirrhosis, and ultimately the development of hepatocellular carcinoma. Although several antiviral drugs, including interferon alpha and nucleotide derivatives, have been approved for clinical treatment of HBV, key issues such as low to moderate efficacy, adverse side effects, and drug-resistant strains remain unresolved. In this study, based on the results of molecular docking experiments, twenty matrine-adenine hybrids were synthesized and their antiviral effect against HBV

IFI27 and BAX are Essential for GSDME-Mediated Myeloma Cell Pyroptosis.

Induction of pyroptosis is a novel strategy for multiple myeloma (MM) treatment, but the underlying mechanism remains elusive. In the analysis of the transcriptomic profile in pyroptotic MM cells, we find the interferon-alpha inducible protein IFI27 is strikingly upregulated. IFI27 is downregulated in MM cells in association with poor prognosis, but its overexpression displays great potency to trigger pyroptosis in both MM cell lines and newly diagnosed MM cells in a GSDME-dependent manner. Moreover, ectopic IFI27 impairs mitochondrial structure and function. Interestingly, when mitochondria a

Overcoming Immunotolerance in Chronic Hepatitis B: Efficacy of Granulocyte-Macrophage Colony-Stimulating Factor-Based Immunotherapy in Achieving Hepatitis B Surface Antigen Seroclearance.

Chronic hepatitis B (CHB) remains incurable due to the immune system's tolerance toward the hepatitis B virus (HBV) surface antigen (HBsAg). This study aimed to achieve a functional cure by breaking HBV tolerance through immunotherapy. CHB patients were treated with either standard nucleotide analog (NA) therapy (Adefovir Dipivoxil, ADV) (Cohort 1) or ADV combined with interferon-alpha (IFN-α) (Cohort 2). Additionally, a third cohort received the THRIL-GM-Vac regimen: three low-dose GM-CSF injections followed by one dose of the HBV vaccine, alongside standard treatment. THRIL-GM-Vac treatment

Restraint of Powassan virus replication by TRIM5α facilitates viral avoidance of antiviral immunity.

TRIpartite Motif (TRIM) protein 5 alpha (TRIM5α) is a well characterized cellular inhibitor of lentivirus replication that limits transmission of related viruses between primates. We previously reported that TRIM5α derived from humans and rhesus macaques inhibits replication of orthoflaviviruses belonging to the tick-borne encephalitis virus (TBEV) serocomplex, including TBEV, Kyasanur forest disease virus and Langat virus (LGTV), but interestingly not the tick-borne Powassan virus (POWV). To further characterize the primate TRIM5α and orthoflavivirus interface, we screened TRIM5α variants fro
