Metabolic Rewiring and Post-Translational Modifications: Unlocking the Mechanisms of Bone Turnover in Osteoporosis.

Osteoporosis is a metabolic disease characterized by low bone density resulting from abnormal bone metabolism, caused by impaired osteogenesis and/or excessive bone resorption. The coordinated differentiation of osteoblasts (originating from mesenchymal stem cells) and osteoclasts (derived from hematopoietic progenitor cells) is necessary for maintaining normal bone remodeling and homeostasis. Metabolites have been confirmed to regulate cellular behavior through post-translational modifications (PTMs), including acetylation, lactylation, and succinylation. During osteoblast and osteoclast diff…

Mesenchymal stem cells prevent ovariectomy-induced osteoporosis formation in mice through intraosseous vascular remodeling.

Mesenchymal stem cells (MSCs) can promote osteogenesis and are a promising therapy for postmenopausal osteoporosis. However, the relationship between improved intraosseous microcirculation and increased bone mass induced by MSCs in postmenopausal osteoporosis remains unclear. After the primary MSCs were characterized, they were transplanted into ovariectomized mice. MSCs transplantation enhanced the trabecular number, trabecular bone volume/total volume, and trabecular bone mineral density in ovariectomized mice. To determine the role of MSCs in vascular repair, mice were subjected to femoral …

How faithfully does intramembranous bone regeneration recapitulate embryonic skeletal development?

Postnatal intramembranous bone regeneration plays an important role during a wide variety of musculoskeletal regeneration processes such as fracture healing, joint replacement and dental implant surgery, distraction osteogenesis, stress fracture healing, and repair of skeletal defects caused by trauma or resection of tumors. The molecular basis of intramembranous bone regeneration has been interrogated using rodent models of most of these conditions. These studies reveal that signaling pathways such as Wnt, TGFβ/BMP, FGF, VEGF, and Notch are invoked, reminiscent of embryonic development of mem…

What If the Prenatal Diagnosis of a Lethal Anomaly Turns Out to Be Wrong?

Advances in prenatal diagnosis create a unique set of clinical ethics dilemmas. Doctors routinely obtain genetic screening, radiologic image … ed based on prenatal assessment. We present a case in which a prenatal diagnosis was made of severe osteogenesis imperfecta, leading to a decision to induce delivery at 31 weeks. On postnatal evaluation, the infant's disease did not appear to be as bad as had been anticipated. We discuss the ethical implications of such diagnostic and prognostic errors.

Implant bone rings. One-stage three-dimensional bone transplant technique: a case report.

A variety of techniques and materials has been used to provide the structural base of bone and soft tissue support for dental implants. Alveolar bone augmentation techniques include different surgical approaches such as guided bone regeneration, onlay grafting, interpositional grafting, distraction osteogenesis, ridge splitting, and socket preservation. In the case presented, a technique was used to augment the alveolar bone three-dimensionally with autologous "bone rings" and immediate implant placement in a 1-stage procedure following teeth extraction. Bone rings (circular osteotomies) were …

Macrophage immunometabolism in osteoporosis: an emerging regulatory axis linking metabolic-polarization coupling to therapeutic targets.

Osteoporosis has long been understood, and treated, as an imbalance between osteoblastic bone formation and osteoclastic resorption. Yet many patients continue to lose bone, pointing to upstream regulators of the remodeling unit that remain poorly defined. Macrophages are developmentally continuous with osteoclasts and functionally coupled to bone-forming cells, and are now recognized as central immune regulators of skeletal homeostasis. What sets their polarization state, however, has been comparatively neglected in bone. This review adopts metabolic-polarization coupling as a unifying framew

Sensory-Skeletal Integration Underlies Diverse Bone Fusion Patterns in a Complex Demography of Cavefish.

Over the course of development, sensory systems must integrate specialized cells and tissues to generate a functional unit. A robust example of this type of integration is the lateral line, a mechanosensory system in fishes and amphibians that senses changes in water movement. Over the past century, numerous reports noted that the principal organs of the lateral line-neuromasts-co-localize to the position of ossification centers for bones of the facial skeleton. More recent work in the freshwater model system, Astyanax mexicanus, revealed facial bones of the suborbital complex frequently fuse

Divergent Mechanisms of Cranial Suture Ossification in Normal Development and Pathologic Fusion.

Cranial sutures are dynamic growth sites that balance bone growth with mesenchymal patency to accommodate cranial expansion during development. While intramembranous ossification has traditionally been considered the default mechanism of suture fusion, accumulating evidence demonstrates that endochondral pathways might also play a significant role under both physiological and pathological conditions. In this review, we contrast normal developmental ossification processes with premature fusion in craniosynostosis, integrating histological, molecular, and imaging data. We highlight the context-d

Antioxidant scaffolds for enhanced bone regeneration: recent advances and challenges.

Bone regeneration is integral to maintaining bone function and integrity in the body, as well as treating bone diseases, such as osteoporosis and defects. However, oxidative stress often poses a significant obstacle during bone regeneration, leading to cell damage, inflammatory responses, and subsequent impediment of normal bone tissue formation. Therefore, to maintain bone regeneration, antioxidant therapy is essential. Bone scaffolds, serving as a temporary support for bone tissue, can provide an ideal microenvironment for cell proliferation and differentiation, effectively promoting bone ti

Luteolin's Potential in Managing Osteoporosis and Bone Metabolism Disorders: Preclinical Insights.

Bone homeostasis is a process of dynamic regulation between bone resorption and bone formation, and is the foundation for normal skeletal development in the human body. Once this balance is disrupted, it will lead to bone loss and decreased bone density, profoundly affecting the occurrence and development of bone metabolism-related diseases. For example, common clinical bone metabolic disorders such as osteoporosis, avascular necrosis of the femoral head, and alveolar bone resorption. However, current treatments for bone metabolism-related diseases often have certain limitations, which has dri

A Systematic Review on the Molecular Mechanisms of Resveratrol in Protecting Against Osteoporosis.

Osteoporosis is a prevalent metabolic bone disorder characterized by decreased bone mineral density and increased fracture risk, particularly among aging populations. While conventional pharmacological treatments exist, they often have adverse effects, necessitating the search for alternative therapies. Resveratrol, a naturally occurring polyphenol, has gained significant attention for its potential osteoprotective properties through various molecular mechanisms. This systematic review aims to comprehensively analyze the molecular pathways through which resveratrol protects against osteoporosi

Tall stature and gigantism in adult patients with acromegaly.

OBJECTIVES: Increased height in patients with acromegaly could be a manifestation of growth hormone (GH) excess before epiphysis closure. The aim of this study was to evaluate the relationship between the height of adult patients with GH excess related to mid-parental height (MPH) and population mean and to find whether taller patients with acromegaly come from tall families. METHODS: This is a single-centre, observational study involving 135 consecutive patients with acromegaly diagnosed as adults and no family history of GH excess. We established three categories for height for patients wit

Developmental toxicity of pyriproxyfen induces changes in the ultrastructure of neural cells and in the process of skull ossification.

Some studies relate the use of pyriproxyfen (PPF) in drinking water with damage to embryonic neurodevelopment, including a supposed association with cases of microcephaly. However, the effects on neural cells and skull ossification in embryos remain unclear. This study aims to investigate the effects of PPF on the structure and ultrastructure of brain cells and its influence on the skull ossification process during embryonic development. Chicken embryos, used as an experimental model, were exposed to concentrations of 0.01 and 10 mg/l PPF at E1. The findings demonstrated that PPF led to notabl

In Situ Construction of Morphologically Different Hydroxyapatite-Mineralized Structures on a Three-Dimensional Bionic Chitin Scaffold.

Slow healing at the tendon-bone interface is a prominent factor in the failure of tendon repair surgeries. The development of functional biomaterials with 3D gradient structures is urgently needed to improve tendon-bone integration. The crystalline form of hydroxyapatite (HAP) has a crucial impact on cell behavior, which directly influences protein adsorption, such as bone morphogenetic protein 2, the adhesion, proliferation, and osteogenic differentiation with cells. This work aimed to generate gradient mineral structures in situ by stabilizing calcium and phosphate ions using a polymer-induc
