The Timing and Magnitude of the Type I Interferon Response Are Correlated with Disease Tolerance in Arbovirus Infection.

Infected hosts possess two alternative strategies to protect themselves against the negative impact of virus infections: resistance, used to abrogate virus replication, and disease tolerance, used to avoid tissue damage without controlling viral burden. The principles governing pathogen resistance are well understood, while less is known about those involved in disease tolerance. Here, we studied bluetongue virus (BTV), the cause of bluetongue disease of ruminants, as a model system to investigate the mechanisms of virus-host interactions correlating with disease tolerance. BTV induces clinica…

Evaluation of frequency of antibodies against Neospora caninum, Toxoplasma gondii and Brucella melitensis, risk factors and spatial distribution of infection in goat and sheep flocks from Argentina.

Neospora caninum, Toxoplasma gondii and Brucella melitensis are pathogens that cause abortion in small ruminants. Besides, B. melitensis and T. gondii are zoonotic pathogens. The aim of this study was to describe the frequency of antibodies against N. caninum, T. gondii and B. melitensis in sheep and goats from three provinces of the center region of Argentina. In addition, the spatial distribution of the infected flocks/herds and risk factors were evaluated. A cross-sectional study was conducted from 2015 through 2016. Serum samples from 4783 goats and 1524 sheep from 186 goat, 51 sheep and 3…

An assessment of Walk-over-Weighing to estimate short-term individual forage intake in sheep.

The main limitation for determining feed efficiency of freely grazing ruminants is measurement of daily individual feed intake. This paper describes an investigation that assessed a method for estimating intake of forage based on changes in BW of ewes. A total of 24 dry and non-pregnant Romane ewes (12 hoggets, HOG; mean±SD 51.8±2.8 kg BW; body condition score (BCS) 2.6±0.2; and 12 adults, ADU; 60.4±8.5 kg BW; BCS 2.7±0.8) were selected for the study and moved from their rangeland system to a confined pen with controlled conditions and equipped with individual automatic feeders. The experiment…

The L, M, and S Segments of Rift Valley Fever Virus MP-12 Vaccine Independently Contribute to a Temperature-Sensitive Phenotype.

UNLABELLED: Rift Valley fever (RVF) is endemic to Africa, and the mosquito-borne disease is characterized by "abortion storms" in ruminants and by hemorrhagic fever, encephalitis, and blindness in humans. Rift Valley fever virus (RVFV; family Bunyaviridae, genus Phlebovirus) has a tripartite negative-stranded RNA genome (L, M, and S segments). A live-attenuated vaccine for RVF, the MP-12 vaccine, is conditionally licensed for veterinary use in the United States. MP-12 is fully attenuated by the combination of the partially attenuated L, M, and S segments. Temperature sensitivity (ts) limits vi…

Creation of Rift Valley fever viruses with four-segmented genomes reveals flexibility in bunyavirus genome packaging.

UNLABELLED: Bunyavirus genomes comprise a small (S), a medium (M), and a large (L) RNA segment of negative polarity. Although the untranslated regions have been shown to comprise signals required for transcription, replication, and encapsidation, the mechanisms that drive the packaging of at least one S, M, and L segment into a single virion to generate infectious virus are largely unknown. One of the most important members of the Bunyaviridae family that causes devastating disease in ruminants and occasionally humans is the Rift Valley fever virus (RVFV). We studied the flexibility of RVFV ge…

Targeting the cytokine-redox axis: dose-dependent immunomodulation by methotrexate in a biomarker-guided ovine endotoxaemia model.

BACKGROUND: Oxidative stress and excessive inflammation are central to the pathophysiology of sepsis and its large-animal analogue, endotoxaemia. Current anti-inflammatory treatments in ruminants lack efficacy in modulating redox balance or acute-phase responses. Low-dose methotrexate (MTX), a known immunometabolic modulator, may offer targeted attenuation of these processes. OBJECTIVE: To evaluate the anti-inflammatory, antioxidant, and organ-sparing effects of two low-dose MTX regimens in a controlled ovine model of lipopolysaccharide (LPS)-induced endotoxaemia. METHODS: 20 clinically heal…

Congenital anomalies in sheep and goats in Iran over ten years.

BACKGROUND: Iran ranks among the world's leading producers of small ruminants, with over 53 million sheep and 25 million goats. Despite this prominence, congenital defects in these animals remain under-documented, presenting a major gap in veterinary epidemiology and herd health management. This study represents the first nationwide effort to systematically classify and analyze congenital anomalies in Iranian sheep and goats. METHODS: Over 10 years (2014-2024), cases of congenital anomalies were recorded across all 31 provinces of Iran. Data were collected through farm-level clinical evaluati…

Neospora caninum infection specifically suppresses the expression of a host lncRNA XR_001919077.1 to facilitate parasite propagation by modulating host cell mitochondrial function and autophagy.

Neospora caninum is one of the most common pathogens causing reproductive failure in ruminants (e.g., cattle and goats) worldwide. However, due to a poor understanding of the pathogenic mechanisms of N. caninum infection, no effective drugs and vaccines are currently available. Long non-coding RNAs (lncRNAs) have been reported to be important regulators involved in a great number of physiological and pathological processes. Our previous study found that N. caninum infection induced significantly aberrant expression of lncRNA profiles in caprine endometrial epithelial cells (EECs). In the prese…

A systematic study of sheep faeces to be used as an alternative inoculum source in the Hohenheim gas test.

In vitro studies of gas production (GP) using rumen fluid obtained from rumen-cannulated animals are common in feed evaluation for ruminants. The objective was to compare sheep faeces with rumen fluid from dairy cows as inocula in the Hohenheim gas test (HGT) using a large number of different-quality feeds. It then was evaluated whether GP obtained by using rumen inoculum (RI-GP) can be estimated from GP using faecal inoculum (FI-GP). Ninety feeds for ruminants including roughages (n = 27), total mixed rations (n = 6), commercial compound feeds for dairy cows (n = 18), cereal grains (n = 10), …

Genotypic and phylogeographic insights into a pre-epidemic variant of Wesselsbron virus detected in sylvatic Aedes mcintoshi from Semuliki Forest, Uganda.

Wesselsbron virus (WSLV) is a neglected mosquito-borne virus within the yellow fever subgroup in the genus Orthoflavivirus of the Flaviviridae family. Despite being primarily a veterinary pathogen able to cause stillbirths, congenital malformations, and mortality in ruminants, WSLV also infects humans, causing a usually self-limiting febrile illness, or may lead to neurological complications in rare cases. WSLV causes sporadic outbreaks in Southern Africa, but findings in mosquitoes from other African countries suggest a wider distribution. Here, we report the detection and isolation of WSLV f…

Should nonsuicidal self-injury be a putative obsessive-compulsive-related condition? A critical appraisal.

Nonsuicidal self-injury (NSSI) has many behavioral and cognitive features that would make it appear to be closely tied to obsessive-compulsive disorder (OCD). Obsessive-compulsive-related disorders (OCRDs) have been described in the literature as conditions that share a common phenomenology, neurobiology, and treatment response. The authors reviewed the literature describing the degree that NSSI is similar to, and distinct from, OCRDs based on these hypothesized common areas. They conclude with recommendations for conceptualization that draws partly from the OCRD literature and from cognitive-

Outbreak of botulism in a dairy herd in Turkey.

In this study, the clinical findings and results of haematological and biochemical analyses of 26 cattle with botulism were evaluated. The most important clinical signs in the affected cattle included: decreased appetite, ataxia, difficulty to rise, loss of tongue tone, salivation and bradycardia. A definitive diagnosis of botulism was based on demonstration of the preformed toxin in ruminal and intestinal contents and feed materials including poultry litter, by mouse inoculation test. This study is the first confirmation, by direct toxin isolation, of Clostridium botulinum type C and Clostrid

Response styles theory: downward extension to children.

Describes partial support found for the downward extension of Nolen-Hoeksema's response styles theory of depression to preadolescents (N = 201). A self-report measure of response styles was developed. Evidence for the multidimensionality and independence of rumination and distraction is provided, as well as support for the reliability and validity of the new measure. Results supported 2 primary tenets of the theory: sex differences in ruminatory tendencies and associations between response styles and self-reported depressive symptoms. Implications of the findings for understanding depression i

Secondary indigestion as a cause of functional pyloric stenosis in the cow.

In 23 cows suffering from a secondary indigestion, in most cases with septicaemia, the syndrome of functional pyloric stenosis or vagal indigestion developed. The signs were anorexia, ruminal distension with fluid material, abomasal reflux into the ruminoreticulum, dehydration, hypochloraemic, hypokalaemic metabolic alkalosis and uraemia. These signs often disappeared after treatment of both the primary causative disease and the secondary indigestion. The importance of recognising this condition is emphasised, because the serious signs of the secondary indigestion may dominate the causative di

Chlorimipramine therapy for obsessive-compulsive neurosis.

Twenty patients with a history of treatment-resistant obsessional neurosis underwent a 4-week clinical trial of chlorimipramine. Scores on the Psychiatric Questionnaire for Obsessive Compulsive Neurosis, the Leyton Obsessional Inventory, and 3 self-assessments indicated that although there was no significant change in obsessive ruminations, obsessive rituals, horrific temptations, or pervading doubt, there was a substantial significant improvement in the obsessive symptom item and severity of obsessions, as well as anxiety, depression, and phobia. Side effects were mild. The authors believe th

In vitro culture of the parasitic stage larvae of hematophagous parasitic nematode Haemonchus contortus.

Current research on common parasitic nematodes is limited because their infective stages cannot be propagated in vitro. Here, we report a culture system for developing L4s of Haemonchus contortus, a blood-feeding nematode of ruminants. Our results demonstrated that a proportionate mixture of NCTC-109 to Luria-Bertini (1:2) media promoted the formation of early L4s and then into late L4s upon inclusion of 12.5% (v/v) defibrinated blood, albeit with a decline in survival. Adding antioxidants (0.3 mg/mL of L-glutathione or 200 nmol of vitamin C) improved survival of L4s, with approximately 90% de…

Molecular Diagnosis of Phlebovirus riftense Infection Using an L-Segment-Based Real-Time RT-PCR Method.

Rift Valley fever (RVF) is one of the main vector-borne zoonotic diseases that affects a wide range of ruminants and humans in Africa and the Arabian Peninsula.Several techniques involving cell culture and molecular biology methods have been developed to diagnose RVF infection. Success partly relies on sending samples to a national or reference laboratory in good conditions and having the capacity to perform the appropriate diagnostic test in the matrix of interest during the period of viremia where high loads of viral particles are present. Conventional and duplex/multiplex real-time reverse …

Rift Valley Fever Virus: An update on current status and future prospects.

Rift Valley fever is a mosquito-borne disease caused by the Rift Valley Fever Virus (RVFV) belonging to the genus Phlebovirus. This virus causes febrile or hemorrhagic illness in humans and ruminants, such as abortion, and death; especially in young sheep, cattle, and goats resulting in devastating epidemics in Africa and the Arabian Peninsula. The WHO has included this virus in Bluepoint's list of eight pathogens. This virus is a crucial health concern in the Kingdom of Saudi Arabia (KSA), as the Kingdom is regularly exposed to this virus from the original source of East African countries. A …
