Secondary organizing pneumonia associated with sertraline: A case report.

Cryptogenic organizing pneumonia is a rare idiopathic interstitial lung disease, with a well-defined clinical-radiological and pathological entity. It may also be secondary to several causes. Rapid clinical and imaging improvement is usually obtained with corticosteroid therapy. We report here, to the best of our knowledge, a unique case of organizing pneumonia associated with Sertraline, a selective serotonin reuptake inhibitor, commonly used in antidepressant therapy.

Fluvoxamine-induced intracranial hypertension in a 10-year-old boy.

Drug-induced intracranial hypertension is a well-established entity. We report a rare case of intracranial hypertension with papilledema in a 10-year-old boy following use of fluvoxamine, a selective serotonin reuptake inhibitor. On discontinuing the drug, the papilledema resolved over 4 months without any residual visual anomalies. To the best of our knowledge, this is the first report of fluvoxamine-induced intracranial hypertension with papilledema.

Upper gastrointestinal bleeding in an end-stage renal disease patient treated with fluoxetine.

The defective platelets of end-stage renal disease patients might be further incapacitated by the antiplatelet effect of selective serotonin reuptake inhibitor, and bleeding tendency in these patients could be aggravated. Here we demonstrate an end-stage renal disease patient who was admitted for duodenal ulcer bleeding during the treatment with fluoxetine, and the bleeding did not stop by various active treatments until the cessation of fluoxetine.

Anorectic effect of dehydroepiandrosterone combined with dexfenfluramine or thionisoxetine.

Free feeding rats given supplementary 1 h access per day to a palatable dessert test meal were tested for the anorectic effect of dehydroepiandrosterone alone or in combination with either the serotonin releasing agent dexfenfluramine or the norepinephrine uptake inhibitor thionisoxetine (LY 368975). Isobolographic analysis showed that the effect of dehydroepiandrosterone combined with either dexfenfluramine or thionisoxetine was within the range predicted for additivity of action.

Tachycardia, orthostatic hypotension and profound weakness due to concomitant use of fluoxetine and nifedipine.

The use of the selective serotonin reuptake inhibitor fluoxetine is associated with only minor cardiovascular effects. However, due to a possible inhibitory effect on the metabolism of calcium channel blockers it may potentiate the activity of nifedipine, causing profound adverse cardiovascular effects. This report describes the appearance of profound weakness, orthostatic hypotension and tachycardia following the initiation of fluoxetine treatment in a nifedipine-treated 80-year-old patient.

[The role of acetaldehyde in the pathogenetic mechanisms of the development of chronic alcoholism].

Experiments on white rats were carried out to confirm an important role of acetaldehyde in pathogeny of alcoholism. It is evidenced by results of experiments when animals were given acetaldehyde and medichronal (a drug which combines acetaldehyde) and by data obtained in the course of studies of changes in the content of endogenic ethanol and acetaldehyde, activity of alcohol dehydrogenase and aldehyde dehydrogenase, concentration of biogenic amines (catecholamines and serotonin) in the blood and brain structures.

Pharmacologic treatment of anxiety disorders.

The benzodiazepines and the beta-adrenergic blockers are effective in the treatment of anxiety neurosis (generalized anxiety disorder and panic disorder) but offer little help in the treatment of agoraphobia and obsessive-compulsive disorders. The antidepressants are often effective in agoraphobia and obsessive-compulsive disorders, and the evidence suggests that their antipanic and antiobsessional effects may be independent of their antidepressant properties. There is suggestive evidence linking the antiobsessional effect to a specific neurotransmitter (serotonin) system.

Methysergide in the treatment of narcolepsy.

Five patients with narcolepsy (four with the allied symptom of cataplexy) were treated with the serotonin antagonist methysergide. All patients had as good control of their sleep attacks while on methysergide therapy as on a control period of dextroamphetamine therapy. The cataplexy was less well controlled by methysergide than by dextroamphetamine, but improved when compared to a period without medication. Two patients developed severe calf claudication while on methysergide.

Sertraline-induced galactorrhea: case report and review of cases reported with other SSRIs.

There is limited literature reporting galactorrhea with antidepressants including selective serotonin reuptake inhibitors (SSRIs). In this case report, I present a case of a young female who developed galactorrhea with sertraline, which improved on discontinuation of sertraline. Computer-assisted searches on galactorrhea with SSRIs yielded 23 cases, mostly with escitalopram and paroxetine and rarely with fluoxetine, fluvoxamine and sertraline, and it may be much more frequent than recognized.

The emergence of tics during escitalopram and sertraline treatment.

The selective serotonin reuptake inhibitors (SSRIs) have well-established antidepressant activity and have also demonstrated efficacy in a number of anxiety disorders, including panic disorder. The SSRIs, escitalopram and sertraline, are well-tolerated and movement disorders are rarely associated with their use. We present a case of the emergence of tics associated with escitalopram and sertraline treatment, which might be due to an imbalance in the dopaminergic neurotransmitter system.

Alcohol, the reward system and dependence.

Evidence is presented to show that multiple neurotransmitter systems of the brain reward systems including GABA, glutamate, dopamine, serotonin and opioid peptides are involved in alcohol reinforcement. Dependence is associated with changes in many of these same systems, but also with changes in other neurotransmitters, such as brain corticotropin releasing factor. A midbrain forebrain circuitry that involves parts of the nucleus accumbens and amygdala is hypothesized to be the focus for the neuropharmacology of alcohol reinforcement.

Changes in the level of biogenic amines in experimentally infected and DEC treated proxy host mice.

The levels of biogenic amines, viz., epinephrine, histamine and dopamine were significantly altered in the blood and brain of the proxy host mice which were infected with the mf of Setaria digitata, a filarial parasite. When DEC, the drug of choice was administered to the infected mice, serotonine level was enhanced while the histamine level was decreased. The results were similar in normal mice which were given DEC.

Methysergide-induced psychosis: case report with long-term follow-up.

Methysergide, a congener of LSD and potent serotonin (5-HT) antagonist, induced a florid transient psychosis resembling an LSD psychosis in a patient with spasmodic torticollis with a family history of schizophrenia. Thirteen years later he developed a major depression. A variety of other drug challenges and treatments for his torticollis had no psychotomimetic effect. Blockade of 5-HT receptors may subserve methysergide-induced psychosis.

[Patterns of the formation of endocrine reactions in relation to the nature and manifestation of a pathological process in the gastroduodenal area].

Correlation between a degree of activation of hormonal secretion and the expression of a pathological process in the gastroduodenal area was established in patients with peptic ulcer, chronic gastroduodenitis with raised secretory gastric function and in patients after vagotomy. A sequence of involvement in the reaction to disease of the regulatory systems of the local (gastrin, serotonin and histamine mechanisms), systemic (enteroinsular axis) and organic (glucocorticoid and adrenal function of the adrenal glands) levels was revealed.

[Effect of activation of alpha- and beta-adrenergic receptors, M-cholinergic receptors and H1-histamine receptors on mechanical tension in the frog subclavian vein].

The effects of adrenaline, noradrenaline, isoproterenol, dopamine, histamine, acetylcholine, and serotonin on the wall tension in the isolated frog subclavian vein and modification of these effects by appropriate antagonists were examined. The existence of alpha- and beta-adreno-, M-choline- and H1-histamine receptors was demonstrated. Stimulation of alpha-adreno- and H1-histamine receptors augments and that of beta-adreno- and M-cholinoreceptors diminishes active wall tension in the vein.

Treatment of depressive-like behaviour in Huntington's disease mice by chronic sertraline and exercise.

BACKGROUND AND PURPOSE: Depression is the most common psychiatric disorder in Huntington's disease (HD) patients. Women are more prone to develop depression and such susceptibility might be related to 5-hydroxytryptaminergic (serotonergic) dysregulation. EXPERIMENTAL APPROACH: We performed tests of depression-related behaviours on female R6/1 HD mice that had been chronically treated with sertraline or provided with running-wheels. Functional assessments of 5-HT(1A) and 5-HT(2A) receptors were performed by measuring behavioural and physiological responses following administration of specific

Neurobiological changes mediating the effects of chronic fluoxetine on cocaine use.

Acute SSRI (selective serotonin reuptake inhibitor) treatment has been shown to attenuate the abuse-related effects of cocaine; however, SSRIs have had limited success in clinical trials for cocaine abuse, possibly due to neurobiological changes that occur during chronic administration. In order to better understand the role of serotonin (5HT) in cocaine abuse and treatment, we examined the effects of chronic treatment with the SSRI fluoxetine at clinically relevant serum concentrations on cocaine-related neurobiology and behavior. Rhesus macaques self-administering cocaine underwent a 6-week …

Differential involvement of 5-HT(1B/1D) and 5-HT6 receptors in cognitive and non-cognitive symptoms in Alzheimer's disease.

Growing evidence suggests that a compromised serotonergic system plays an important role in the pathophysiology of Alzheimer's disease (AD). We assessed the expression of 5-HT(1B/1D) and 5-HT(6) receptors and cholinacetyltransferase (ChAT) activity in post-mortem frontal and temporal cortex from AD patients who had been prospectively assessed for cognitive function using the Mini-Mental State Examination (MMSE) and behavioral changes using the Present Behavioral Examination (PBE). 5-HT(1B/1D) and 5-HT(6) receptor densities were significantly reduced in both cortical areas. 5-HT(1B/1D) receptor

Central 5-HT(4) receptors and dopamine-dependent motor behaviors: searching for a functional role.

In this study, we evaluated the role of central 5-HT(4) receptors in the control of motor behaviors related to change of nigrostriatal dopamine (DA) transmission, namely, stereotyped behavior and catalepsy in rats. Indeed, given that 5-HT(4) receptors indirectly modulate nigrostriatal DA neuron activity, we hypothesized that these receptors would regulate nigrostriatal DA transmission in the basal ganglia, and consequently, associated motor responses. Stereotypy was induced either by an acute administration of apomorphine (0.3 and 1.5 mg/kg sc), or by a single morphine administration (15 mg/kg
