Urolithins at the Crossroads of Gut Inflammation and Cancer-A Narrative Review.

Chronic inflammation and the associated dysbiosis of the gut microbiota are increasingly recognized as key factors contributing to the development of inflammatory bowel disease (IBD) and colorectal cancer (CRC). The diet-gut microbiota-immune system-cancer axis is considered a key regulator of these processes. Among the bioactive compounds found in the diet, ellagitannins have garnered significant scientific interest due to their conversion by the gut microbiota into biologically active metabolites known as urolithins. Urolithin A (UroA), one of the best characterized compounds, exhibits broad

Shared immune dysregulation in systemic lupus erythematosus and colorectal cancer: a multi-omics guided discovery of DNASE1L3-centric efferocytosis deficiency.

BACKGROUND: Although immune dysregulation is implicated in both autoimmune diseases and cancer, comparative pathogenesis and immune response mechanisms between systemic lupus erythematosus (SLE) and colorectal cancer (CRC) remain elusive. This study identifies common molecular biomarkers and pathogenic pathways shared between SLE and CRC via multi-omics analysis. METHODS: Integrated datasets including SLE (GSE61635, GSE50772, GSE142016) and CRC cohorts (GSE39582, GSE17536, E-MTAB-8107, COAD, READ), were analyzed. Differential expression analysis identified shared genes, and machine learning s

Colorectal cancer in Greenland.

INTRODUCTION: Distal radius fractures (DRFs) are common, with over 14,000 annual cases in Denmark. Most cases are treated non-surgically and may require closed reduction prior to casting. National guidelines for anaesthesia and reduction technique for closed reduction of DRFs are lacking, impacting clinical practices and potentially optimal care. This survey aimed to describe Danish clinical practice for initial treatment and closed reduction of DRFs. METHODS: Between January and April 2025, the survey was distributed online to the 23 physicians responsible for all 44 emergency departments an

Molecular features and clinical significance of cancer-associated fibroblasts in colorectal cancer.

The elevated occurrence and fatality rates of colorectal cancer (CRC) can be attributed not only to the biological attributes of uncontrolled tumor cell proliferation but also to the intricate interplay between tumor cells and surrounding cells in the microenvironment. Among these, cancer-associated fibroblasts (CAFs) stand out as a predominant type of mesenchymal cell in the microenvironment, with their biological functions in tumor advancement becoming increasingly recognized. CAFs play a significant role in the progression of CRC through diverse mechanisms, such as direct interactions with

Initial management strategies for right-sided malignant colonic obstruction: a systematic review and bayesian network meta-analysis.

BACKGROUND: Right-sided malignant colonic obstruction (RMCO) remains challenging, as the evidence is more limited than for left-sided disease. Although primary resection is the standard approach, emergency surgery carries higher perioperative risks, increasing interest in bridge-to-surgery (BTS) strategies. However, their optimal role remains uncertain. We conducted a systematic review and Bayesian network meta-analysis to compare initial management strategies for RMCO and evaluate short- and long-term outcomes. METHODS: A comprehensive search of PubMed, Embase, and the Cochrane Library was pe

Modulatory role of Blastocystis-derived solubilized antigens in colorectal carcinogenesis: A systematic review.

Colorectal cancer develops through complex interactions among genetic alterations, immune responses, and the intestinal microenvironment. Increasing evidence suggests that intestinal parasites may also contribute to colorectal carcinogenesis (CRC). Blastocystis sp. (Blastocystis) is one of the most common intestinal protozoans worldwide. Although its role in disease remains controversial, it has been associated with chronic inflammation and disruption of normal gut homoeostasis. Therefore, this systematic review aimed to summarise the current experimental evidence on the immunomodulatory effec

Colorectal cancer organoid co-culture systems and their applications in drug screening.

Colorectal cancer (CRC) is a common malignant tumor. The incidence and mortality rates of CRC in China have been rising over the years. Organoids are micro-organs that can self-organize into three-dimensional structures and recapitulate the morphological structures and functions of native organs. Organoid culture technology is developing rapidly and has shown great potential for applications in translational biomedical research such as high-throughput screening, drug development, toxicity assessment, and personalized tumor treatment. In this review, we first introduce the development of patien

A Droplet Digital PCR Approach for the Quantitative Detection of Fusobacterium nucleatum in Formaldehyde-Fixed, Paraffin-Embedded Tumor Tissues.

Fusobacterium nucleatum, a prevalent component of the intratumoral microbiome, has been associated with reduced survival in colorectal cancer. To enable sensitive and specific detection of F. nucleatum in tumor specimens, we developed a droplet digital PCR (ddPCR) assay targeting the transcription termination/anti-termination gene nusG, normalized to host tissue content using the solute carrier organic anion transporter family member 2A1 gene, SLCO2A1. Here, we present a ddPCR protocol optimized for quantifying F. nucleatum DNA in human genomic DNA extracted from formalin-fixed, paraffin-embed

Modeling Fusobacterium nucleatum-Driven Colorectal Cancer in Apc-Mutant Mice.

Colorectal cancer (CRC) typically initiates with mutations in the adenomatous polyposis coli (Apc) gene, followed by the influence of environmental factors. Among these, Fusobacterium nucleatum has emerged as an important CRC-enriched microbial species with demonstrated tumor-promoting potential. To investigate the contribution of F. nucleatum in the context of oncogenic mutations, physiologically relevant mouse models are essential. While Apcmin/+ mice have been employed to study F. nucleatum-associated CRC, prior studies have not systematically evaluated the effects of different bacterial in

A network toxicology and machine learning approach to uncover the molecular machinery of bisphenol A-induced colorectal cancer.

The carcinogenic relevance of environmental contaminant bisphenol A (BPA) to colorectal cancer (CRC) has gained growing attention, yet the molecular networks potentially linking BPA exposure to CRC remain incompletely characterized. This study integrated network toxicology and machine learning to predict candidate molecular targets and putative signaling networks associated with BPA-correlated colorectal carcinogenesis. The study's methodology involved an initial differential expression screening across several CRC transcriptomic datasets to establish a disease-specific gene signature. Subsequ

Bayesian Integration of Tumor Mutational Signatures and Somatic Features Refines Pathogenicity Assessment of Germline Mismatch Repair Variants.

Variants of uncertain significance (VUS) in mismatch repair (MMR) genes represent a persistent bottleneck in germline interpretation for Lynch syndrome, creating a critical opportunity to leverage tumor biology to refine pathogenicity assessment. Although tumor features such as microsatellite instability (MSI) and immunohistochemistry (IHC) are routinely evaluated, they are typically interpreted separately from germline classification, and their quantitative contribution within ACMG/AMP frameworks remains poorly defined. We therefore analyzed paired germline and tumor sequencing data from 1110

UBE2D4 Upregulation Promotes Cuproptosis Sensitivity in Colorectal Cancer.

BACKGROUND: Cuproptosis, a copper-dependent form of regulated cell death, represents a potential therapeutic vulnerability in colorectal cancer (CRC). However, the regulatory mechanisms governing cuproptosis in CRC remain largely unknown. METHODS: UBE2D4 expression was analyzed in the TCGA-COAD cohort and validated in CRC cell lines (HCT116, HT29) and normal colon epithelial cells (FHC) by qRT-PCR and western blot. Paired CRC and adjacent normal tissues (n = 5) were also examined by western blot. UBE2D4-knockdown HT29 cells were generated by transient siRNA transfection to assess cell viabili

Three-year risk of colorectal cancer by faecal haemoglobin concentration in subjects with a negative FIT.

INTRODUCTION AND AIM: To evaluate whether a risk-stratified colorectal cancer (CRC) screening approach based on a single faecal immunochemical test (FIT) performed better than one using two consecutive tests. METHODS: Approximately 420,000 negative FIT results (positivity cut-off: 20 µg faecal haemoglobin, f-Hb/g) were collected between 2000 and 2015 within the Florence screening program. RESULTS: The 3-year CRC risk was 15.9‰ in individuals with 15-20 µg/g on a single test, and 9.36‰ for those with a cumulative f-Hb concentration of 20-40 µg/g across two tests. DISCUSSION: The two-tests ap

Heterogeneity of treatment preferences in the absence of guideline recommendations - a case vignette study in colorectal cancer tumor boards in Germany, Austria and Switzerland.

BACKGROUND: For the treatment of colorectal cancer, the S3-Guideline of the German Guideline Program in Oncology supports clinical decision-making. Centers certified by the German Cancer Society are required to implement the guideline recommendations as comprehensively as possible. When guidelines provide insufficient or ambiguous evidence, heterogeneity of treatment preferences is likely to emerge across individual centers. The aim of this study is to describe the preferences of the centers' tumor boards for treatment decisions when clear, evidence-based guideline recommendations are lacking.

Inactivation of necroptosis-promoting protein MLKL creates a therapeutic vulnerability in colorectal cancer cells.

Mortality from colorectal cancer (CRC) is significant, and novel CRC therapies are needed. A pseudokinase MLKL typically executes necroptotic cell death, and MLKL inactivation protects cells from such death. However, we found unexpectedly that MLKL gene knockout enhanced CRC cell death caused by a protein synthesis inhibitor homoharringtonine used for chronic myeloid leukemia treatment. In an effort to explain this finding, we observed that MLKL gene knockout reduces the basal CRC cell autophagy and renders such autophagy critically dependent on the presence of VPS37A, a component of the ESCRT
