Efficient myelin repair in the macaque spinal cord by autologous grafts of Schwann cells.

Experimental transplantation in rodent models of CNS demyelination has led to the idea that Schwann cells may be candidates for cell therapy in human myelin diseases. Here we investigated the ability of Schwann cells autografts to generate myelin in the demyelinated monkey spinal cord. We report that monkey Schwann cells derived from adult peripheral nerve biopsies retain, after growth factor expansion and transduction with a lentiviral vector encoding green fluorescent protein, the ability to differentiate in vitro into promyelinating cells. When transplanted in the demyelinated nude mouse sp…

The effect of a repeat bout of exercise on muscle injury in persons with spinal cord injury.

Following an initial bout of damaging exercise, a successive bout of similar exercise typically results in less injury, known as the "protective effect". Unloading due to spinal cord injury (SCI) increases the susceptibility to contraction-induced muscle injury. We tested the hypothesis that two bouts of isometric actions would evoke the same damage in the quadriceps femoris (QF) of patients with SCI. Six male subjects [32 (5) years old, 182 (9) cm, 81 (21) kg, injury level C6-T7, 6 (2) years post-injury, mean (SD)] were tested at two time points (Time1, Time2), separated by 8 weeks. Magnetic …

Roles of serotonin receptor subtypes for the antinociception of 5-HT in the spinal cord of rats.

The contribution of 5-HT (5-hydroxytryptamine) receptor subtypes to the antinociception produced by intrathecal 5-HT in the formalin test was investigated in rats. Intrathecal 5-HT suppressed both phases of behaviors produced by 5% formalin, and this was blocked by antagonists for 5-HT(1B) (3-[3-(Dimethylamino)propyl]-4-hy-droxy-N-[4-(4-pyridinyl)phenyl]benzamide dihydrochloride, GR 55562), 5-HT(2C) (N-ormethylclozapine/8-Chloro-11-(1-piperazinyl)-5H-dibenzo[b,e][1,4]diazepine, D-MC), 5-HT3 (1-Methyl-N-(8-methyl-8-azabicyclo[3.2.1]-oct-3-yl)-1H-indazole-3-carboxamide maleate, LY-278,584) and 5

Magnetic resonance imaging as a guide in the diagnosis and follow-up of spinal cord infarction.

BACKGROUND: Spinal cord infarction (SCI) is a dynamic process; it can present wide-spectrum images simulating a variety of entities. The purpose of this study is to characterize the typical magnetic resonance (MR) findings and to describe evolutions and contrast-enhanced patterns of SCIs at different stages. METHODS: MR examinations of 15 patients (10 men and 5 women) with clinical diagnosis of SCIs were reviewed respectively. MR was performed from 3 hours to 178 days after onset of symptoms. The images studies were reviewed with special attention to the location, cord size and sequential sig…

Effect of ischaemia & aglycaemia on the synaptic transmission in neonatal rat spinal cord in vitro.

BACKGROUND & OBJECTIVES: In vitro models of anoxia have revealed severe changes in neuronal functions after ischaemia but not after aglycaemia, although hypoglycaemia produced severe neuronal dysfunctions sometimes leading to coma. The present study was therefore undertaken to examine and compare the effects of aglycaemia with that of ischaemia on synaptic transmission in vitro. METHODS: Spinal cord from the neonatal rat was isolated, hemisected and placed in a chamber perfused with standard physiological solution. The stimulation of a dorsal root elicited monosynaptic (MSR) and polysynaptic …

Leukocytospermia in spinal cord injured patients is not related to histological inflammatory changes in the prostate.

PURPOSE: Patients with spinal cord injuries have abnormal seminal plasma, which contributes to impaired sperm motility and viability. A common finding in these patients is an elevated leukocyte count in semen. We examined the prostatic tissue of spinal cord injured patients vs young healthy controls to determine whether a pathological process related to the prostate gland is a possible source of leukocytospermia. MATERIALS AND METHODS: Seven men with a mean age of 26.1 years with spinal cord injury and 4 controls with a mean age of 35.0 years underwent standard transrectal ultrasound guided p…

[A case of perineal abscess due to urethral fistula in a patient with spinal cord injury].

We report a case of perineal subcutaneous abscess due to urethral fistula in a patient with spinal cord injury. A 39-year-old male visited our hospital complaining of left scrotal swelling and fever. The left scrotum and perineal skin were swollen to the size of a goose egg, and pus was discharged from the perineal swollen bump. Magnetic resonance imaging (MRI) suggested an urethral fistula with a large subcutaneous abscess. The abscess was resected with debridement of necrotic tissue, and a cystostomy was placed. Endoscopy revealed a fistula in the bulbar urethra. The characteristics of this …

The contribution made by Schistosoma infection to non-traumatic disorders of the spinal cord in Malawi.

In Malawi, although schistosomal myelopathy has been reported in visitors from overseas who have swum in Lake Malawi, the incidence of this disorder in local residents has never been investigated. Consecutive patients with non-traumatic disorders of the spinal cord were therefore recruited in a hospital and a rehabilitation centre in Blantyre. Of the 33 patients investigated, 16 were presumed to be cases of schistosomal myelopathy as they had the markers of past or current schistosomiasis and apparently no other conditions that could explain their clinical features. There was microscopical and…

Effects of abdominal trussing on breathing and speech in men with cervical spinal cord injury.

Abdominal trussing constitutes an inward fixation of the abdominal wall. In the context of speech habilitation/rehabilitation, it is sometimes used to supplant, in part, the function of a paralyzed or paretic abdominal wall. A paralyzed or paretic abdominal wall may be found in individuals with cervical spinal cord injury or in individuals with other neurologic disorders, such as muscular dystrophy. This research was designed to study the effects of trussing as a dinical treatment. A study was designed to systematically and precisely place the abdominal wall at one position (50% inward) to stu…

Evidence for pathological involvement of the spinal cord in motor neuron disease-inclusion dementia.

Fronto-temporal dementia (FTD), characterised pathologically by ubiquitinated inclusions in the hippocampal dentate fascia and fronto-temporal cortex, may develop in association with motor neuron disease (MND). However, FTD with identical pathological hallmarks may occur in isolation and has been termed motor neuron disease-inclusion dementia (MND-ID). We studied the pathology of three cases of MND-ID including the spinal cord, which has not previously been examined in this condition. The ages of the patients at death were 53, 70 and 68 years and the onset of FTD 10, 15 and 9 years before deat…

Afferent pathways of sympathetic skin response in spinal cord: a clinical and electrophysiological study.

BACKGROUND: Sympathetic skin response (SSR) recording is an established test of sudomotor autonomic functions. However, knowledge of its pathways in spinal cord is putative. OBJECTIVE: This study involved subjects with isolated spinal cord lesions to evaluate the afferent pathways of SSR. METHODS: Clinical examination was done according to standard neurological classification of spinal cord injury. Electrophysiological evaluation included: (1) conventional nerve conduction studies to exclude peripheral nerve lesions, (2) scalp somato-sensory-evoked potentials (SEP) with posterior tibial nerv…

Absence of echovirus sequences in brain and spinal cord of amyotrophic lateral sclerosis patients.

The role of enteroviruses in pathogenesis of amyotrophic lateral sclerosis (ALS) is controversial. A recent study, based on reverse transcription-polymerase chain reaction (RT-PCR) analysis of spinal cord, reported identification of a novel echovirus in 15 of 17 French subjects with ALS and only 1 of 29 subjects with other neurologic diseases. We established a real-time RT-PCR method based on this novel echovirus sequence and used this method and that previously employed for analysis of the French subjects to determine the prevalence of echoviral sequences in spinal cord and motor cortex of sp…
