A plant vesicle-dendritic cell chimera for enhancing cancer immunotherapy.

The tumour microenvironment (TME) causes mitochondrial dysfunction in resident dendritic cells (DCs), resulting in inadequate antigen presentation and weak T cell priming. Herein, we identify hypoxia as a key factor for causing pathological mitochondrial fission in tumour-associated DCs, and develop a plant vesicle-DC chimera to relieve hypoxia-induced mitochondrial dysfunction for enhancing cancer immunotherapy. The biohybrid chimera is fabricated by loading algae-derived nanovesicles (ANVs) with genetically engineered CCR2 overexpressing-DCs. The CCR2-DC-ANVs target tumour by leveraging the

Dendritic Cells in the Immunoregulation of Myocardial Infarction: Mechanisms and Therapeutic Strategies.

Myocardial infarction (MI) remains a leading cause of mortality and long-term disability worldwide. Its pathological process involves multiple interconnected stages, including atherosclerosis, ischemia-reperfusion injury, ventricular remodeling, and fibrosis. Dendritic cells (DCs), as key antigen-presenting cells bridging innate and adaptive immunity, exert bidirectional immunoregulatory functions in the pathogenesis of MI. During atherogenesis, DCs actively participate in lipid deposition, foam cell formation, and inflammatory regulation. Following MI, DCs influence myocardial injury and repa

Restoring Immunological Tolerance via Dendritic Cells in Rheumatoid Arthritis.

Rheumatoid arthritis (RA) represents a major global public health challenge, with approximately 1% of the world's population suffering from this disease. In the absence of a cure, patients require ongoing and very often lifelong treatment. While environmental, genetic, and epigenetic factors have all been linked to the development of RA, a key, universally accepted initiating factor in disease development is the loss of immunological tolerance to self-antigens. Currently, most treatment approaches utilise agents that suppress the immune system or inflammatory response. However, there is no cur

FDG PET/CT in Duodenal Follicular Dendritic Cell Sarcoma.

Follicular dendritic cell sarcoma (FDCS) of the duodenum is extremely rare. We describe the contrast-enhanced CT and FDG PET/CT findings in a case of duodenal FDCS. Contrast-enhanced CT revealed a large, irregular mass originating from the horizontal portion of the duodenum. The mass demonstrated heterogeneous enhancement, with internal areas of low-attenuation necrosis and coarse calcifications. The mass showed significantly increased FDG uptake. The patient underwent surgical resection, and histopathologic examination revealed a spindle cell tumor positive for CD21 and CD35, confirming the d

Primary hepatic interdigitating dendritic cell sarcoma: A rarity unveiled.

Interdigitating dendritic cell sarcoma (IDCS) is a rare malignancy originating from dendritic cells, which participate in immune response activity as antigen-presenting cells by stimulating T lymphocytes. We report the case of a 24-year-old man who experienced an involuntary 20 kg weight loss over 2 months, combined with a steadily growing, painless lump in his right upper abdomen. Upon examination, a hard, nontender, immobile mass measuring 16 × 16 cm was found in the right hypochondrium. A massive, well-defined hypodense lesion covering all segments of the liver's right lobe was excised and

Defining the Role of Dendritic Cells in Red Blood Cell Alloimmunization.

Storage of red blood cells (RBCs) increases the incidence of alloimmunization following transfusion. Mouse transfusion models have revealed the central role that splenic dendritic cells (DCs) play in storage-dependent RBC alloimmunization. The use of flow cytometry, immunofluorescence imaging, and T-cell proliferation assays combined with various genetically modified mouse strains has provided a detailed understanding of the processes by which DCs instruct the generation of alloantibodies. We and others have established a sequence of events in which transfused stored RBCs are taken up by splen

Overcoming the limitations of motion sensor models by considering dendritic computations.

The estimation of motion is an essential process for any sighted animal. Computational models of motion sensors have a long and successful history but they still suffer from basic shortcomings, as they disagree with physiological evidence and each model is dedicated to a specific type of motion, which is controversial from a biological standpoint. In this work, we propose a new approach to modeling motion sensors that considers dendritic computations, a key aspect for predicting single-neuron responses that had previously been absent from motion models. We show how, by taking into account the

Crystal Growth Engineering for Dendrite-Free Zinc Metal Plating.

The practical implementation of aqueous zinc-ion batteries (AZIBs) is limited by uncontrolled zinc (Zn) dendrite growth during anode plating, compromising both safety and cycle life. Typically, Zn plating proceeds via 2D growth along the six equivalent prismatic [ 10 1 ¯ 0 ] directions of the hexagonal close-packed (HCP) Zn lattice, forming hexagonal platelets that promote dendrite formation. Here, an effective electrolyte engineering strategy is presented using rare-earth ions to regulate Zn plating. Combined multiscale experimental analyses and computational modeling reveal that these ions

Relapsing Blastic Plasmacytoid Dendritic Cell Neoplasm With Neurologic Involvement.

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare and aggressive hematologic malignancy characterized by cutaneous, bone marrow, and central nervous system (CNS) involvement. Despite advances in therapy, prognosis remains poor, particularly in relapsed or refractory disease. We present the case of a woman in her 30s with a history of BPDCN diagnosed three months prior at an outside hospital following symptoms of blurry vision, headaches, night sweats, and weight loss. She achieved initial remission after AML-type induction chemotherapy (7+3 with cytarabine and idarubicin) but was

Follicular Dendritic Cell Sarcoma of the Spleen: A Report of a Rare Case.

Follicular dendritic cell sarcoma (FDCS) is a rare neoplasm that is composed of cells with morphological and immunophenotypic features of follicular dendritic cells. A minority of these tumors are found on extranodal sites. We describe the case of a 58-year-old woman referred to the hospital emergency department by her general practitioner due to new-onset bicytopenia and petechial rash. The diagnostic workup showed an iron and folic acid-deficient normocytic normochromic anemia, and she was admitted to hospitalization. Further investigation revealed Helicobacter pylori infection, later eradic

Dendritic cell vaccines: Current research progress, challenges, and opportunities.

This review examines the advancements in cancer immunotherapies, particularly focusing on dendritic cell (DC)-based vaccines developed through in vitro methods. DCs are essential for connecting innate and adaptive immunity and serve as powerful antigen-presenting cells. They play an essential role in the anti-tumor immune response by activating cytotoxic T lymphocytes and natural killer cells. DC vaccines, which involve engineering DCs to express tumor-associated antigens and administering them to patients, potentially enhance the T-cell-mediated destruction of tumor cells. The review details

The MCPH7 Gene Product STIL Is Essential for Dendritic Spine Formation.

Dendritic spine formation/maintenance is highly dependent on actin cytoskeletal dynamics, which is regulated by small GTPases Rac1 and Cdc42 through their downstream p21-activated kinase/LIM-kinase-I/cofilin pathway. ARHGEF7, also known as ß-PIX, is a guanine nucleotide exchange factor for Rac1 and Cdc42, thereby activating Rac1/Cdc42 and the downstream pathway, leading to the upregulation of spine formation/maintenance. We found that STIL, one of the primary microcephaly gene products, is associated with ARHGEF7 in dendritic spines and that knockdown of Stil resulted in a significant reductio

Dendritic fibromyxolipoma of the male breast: case report and literary review.

BACKGROUND: Dendritic fibromyxolipoma (DFML) represents a benign lipomatous tumor. Its rarity and distinctive morphology can lead to misinterpretation of other mesenchymal malignancies, which may result in unnecessary aggressive treatments. CASE PRESENTATION: A 39-year-old male presented with a left mammary mass that had been noticed 1 year earlier. Chest computed tomography identified a nodule characterized by fat density in the left mammary region. The initial clinical impression was gynecomastia. A local excision of the left mammary mass was subsequently performed. Histopathological analys

Functional and Metabolic Heterogeneity of Dendritic Cells in Self-Tolerance and Autoimmunity.

Dendritic cells (DCs) demonstrate remarkable functional and metabolic heterogeneity that governs the balance between immune tolerance and autoimmune pathogenesis. Under homeostatic conditions, tolerogenic DC subsets maintain immunological equilibrium through distinct metabolic programs and the production of immunoregulatory metabolites, promoting T cell anergy and regulatory T cell (Treg) differentiation. In contrast, autoimmune conditions trigger pathogenic metabolic rewiring, shifting DCs toward glycolysis and enhanced lipid synthesis, which drives DC hyperactivation and breakdown of self-to

Guardians on call: neutrophils, macrophages and dendritic cells in arthritis pathogenesis.

Several immune/inflammatory components have been associated with arthritis. The role of monocytes/macrophages in inflammatory arthritis has been explored over the last years; however, the role of other myeloid cells, such as neutrophils and dendritic cells, in driving the pathophysiology of arthritis is largely overlooked. In this article, we aim to discuss literature pointing to the role of these immune cells in inflammatory arthritis and emphasize the multiple and dynamic phenotypic roles these cells can hold either in the persistence or in the resolution of inflammation. We also highlight t

The role of targeted therapies in blastic plasmacytoid dendritic cell neoplasm.

INTRODUCTION: Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is an ultra-rare disease that arises from malignant precursor dendritic cells. Historically, there was no uniform standard-of-care chemotherapy. In this setting, outcomes were substandard, especially for patients not eligible for intensive chemotherapy, clinical trials, and/or hematopoietic stem cell transplantation, which constituted the majority of patients. The development of targeted therapy, namely the anti-CD123 agent tagraxofusp, marked the beginning of a new era in the treatment of BPDCN. AREAS COVERED: This review sum

Dendritic cell heterogeneity and its role in connective tissue diseases.

By balancing immunity and tolerance, dendritic cells (DCs) are key regulators of immune responses. Recent studies have highlighted the crucial role of these cells in connective tissue diseases. Conventional DCs (cDCs) and plasmacytoid DCs (pDCs) exhibit distinct contributions to disease progression. In systemic lupus erythematosus and systemic sclerosis, pDCs primarily drive pathogenesis via excessive type I interferon production, whereas in rheumatoid arthritis (RA), cDCs play a major role in promoting autoreactive T cell activation. Emerging DC subsets, such as inflammatory DC3s and LAMP3+ D

Macrophages form dendrite-like pseudopods to enhance bacterial ingestion.

Macrophages are critical innate immune cells that exhibit remarkable adaptability during pathogen infections. However, the relationship between their morphological plasticity and physiological functions remains largely elusive. Here, we discovered an unprecedented paradigm of macrophage adaptation within a few hours upon severe Gram-negative bacterial infections, characterized by the formation of dendrite-like pseudopods (DLPs). Using in vitro, microfluidic, and in vivo infection models, we demonstrate that these pseudopods enhance bacterial uptake by expanding the macrophage searching radius,

Dendritic Cells and the Establishment of Fetomaternal Tolerance for Successful Human Pregnancy.

Pregnancy is a remarkable event where the semi-allogeneic fetus develops in the mother's uterus, despite genetic and immunological differences. The antigen handling and processing at the maternal-fetal interface during pregnancy appear to be crucial for the adaptation of the maternal immune system and for tolerance to the developing fetus and placenta. Maternal antigen-presenting cells (APCs), such as macrophages (Mφs) and dendritic cells (DCs), are present at the maternal-fetal interface throughout pregnancy and are believed to play a crucial role in this process. Despite numerous studies foc
