The effect of exogenous substrate concentrations on true and apparent metabolism of hyperpolarized pyruvate in the isolated perfused lung.

Although relatively metabolically inactive, the lung has an important role in maintaining systemic glycolytic intermediate and cytosolic redox balance. Failure to perform this function appropriately may lead to lung disease progression, including systemic aspects of these disorders. In this study, we experimentally probe the response of the isolated, perfused organ to varying glycolytic intermediate (pyruvate and lactate) concentrations, and the effect on the apparent metabolism of hyperpolarized 1-(13)C pyruvate. Twenty-four separate conditions were studied, from sub-physiological to super-ph

Abnormal lactate levels in patients with polymyositis and dermatomyositis: the benefits of a specific rehabilitative program.

BACKGROUND: Polymyositis (PM) and Dermatomyositis (DM) are chronic, inflammatory and autoimmune skeletal muscle disorders characterized by reduced muscle strength, fatigue and myalgia. While inflammation causes muscle damage in the early phase, metabolic alterations such as an impairment of oxidative metabolism seem to be responsible for the disability in the chronic phase of the disease. AIM: To assess muscle oxidative efficiency and to test the effect of aerobic training in a group of PM/DM patients. DESIGN: A case-control study and a within-group comparison. SETTING. Outpatients of the Un

Mitochondrial dysfunction in EEG: high-beta specific metabolic/oxidative coupling with lactate in mania-predominant polarity.

Psychiatric diseases are progressively recognized as disruptions in brain dynamics that can be measured using electrophysiological and metabolic indicators. Conventional EEG investigations utilizing power spectra or standard entropy metrics (sample entropy, multiscale entropy, permutation entropy) have shown diagnostic significance but are constrained in their ability to capture higher-order relationships. This work combines psychiatric ideas with sophisticated mathematical methods to discover new indicators of modified brain complexity. From an engineering standpoint, we provide entropy-doubl

Semaphorin 6A phase separation sustains a histone lactylation-dependent lactate buildup in pathological angiogenesis.

Ischemic retinal diseases are major causes of blindness worldwide and are characterized by pathological angiogenesis. Epigenetic alterations in response to metabolic shifts in endothelial cells (ECs) suffice to underlie excessive angiogenesis. Lactate accumulation and its subsequent histone lactylation in ECs contribute to vascular disorders. However, the regulatory mechanism of establishing and sustaining lactylation modification remains elusive. Here, we showed that lactate accumulation induced histone lactylations on H3K9 and H3K18 in neovascular ECs in the proliferative stage of oxygen-ind

Fear of childbirth among pregnant women in hospital for delivery and its impact on birth outcomes and lactation initiation: a prospective longitudinal study in Wuhan.

BACKGROUND: Fear of childbirth (FOC) has been shown to adversely affect the physical and mental health of pregnant women. However, most of the previous studies have explored only certain factors influencing FOC and have not consistently followed birth outcomes and lactation initiation. This study aimed to understand the incidence of FOC antenatally among pregnant women, analyse the factors influencing this fear, investigate the relationship between generalised anxiety disorder (GAD) and FOC, and explore the influence of this fear on birth outcomes and lactation initiation. METHODS: Between Au

The Δ Anion Gap/Δ Bicarbonate Ratio in Lactic Acidosis: Time for a New Baseline?

KEY POINTS: The mean delta anion gap (ΔAG)/delta bicarbonate (ΔHCO3) calculated using an albumin-corrected anion gap and each patient's individual baseline AG and serum HCO3 was 1.20. The ΔAG/ΔHCO3 using mean normal AG and serum HCO3 was 1.6–1.8; use of mean normal values can result in misdiagnosis of complex acid-base disorders. The elevated ΔAG/ΔHCO3 is likely due to unmeasured anions. BACKGROUND: The ratio of delta anion gap and delta bicarbonate (ΔAG/ΔHCO3) is used to detect coexisting acid-base disorders in patients with high anion gap metabolic acidosis. The ΔAG/ΔHCO3 ratio of 1.6–1.8:1

Lactation and pregnancy in Iran. III. Hormonal factors.

Lactation failure is common in urban areas of industrially developing countries, but little is known about its epidemiology and causality. The study reported here was undertaken to investigate the serum levels of some hormones other than prolactin that have been shown in animal studies to play a role in lactation, and to examine their relationship to adequacy of lactation and to nutritional and socioeconomic status in urban Iranian women. Serum levels of placental lactogen, growth hormone, cortisol, and thyroid hormones were measured under standard conditions in two groups of pregnant women fr

Interplay between N6-methyladenosine RNA methylation and protein lactylation: a novel crosstalk linking metabolism and epigenetic regulation in human diseases.

N6-methyladenosine (m6A) RNA methylation is the most prevalent internal modification in eukaryotic mRNA and precisely regulates gene expression by regulating the RNA life cycle. Lactate, a central glycolytic metabolite, can transduce cellular metabolic states into epigenetic signals through protein lactylation, an emerging post-translational modification. Because both modifications are highly responsive to cellular metabolic status, they have emerged as important regulators of the metabolic-epigenetic interface. Increasing evidence supports a bidirectional regulatory crosstalk between m6A modi

Considerations for lactation with Ehlers-Danlos syndrome: a narrative review.

BACKGROUND: Ehlers-Danlos syndrome (EDS) is a rare genetic connective tissue condition that is poorly understood in relation to lactation. As diagnostic methods improve, prevalence has increased. EDS, a disorder that impacts connective tissue, is characterized by skin extensibility, joint hypermobility, and fragile tissue which can affect every organ and body system leading to complications during pregnancy, delivery, and the postpartum period. Traits of this disease can cause mild to severe physiologic and functional obstacles during lactation. Unfortunately, there is little clinical evidence

Incoordination of sucking, swallowing, and breathing as an etiology for breastfeeding difficulty.

This case report describes an infant who presented with coughing and choking during breastfeeding. Comprehensive clinical evaluation of these difficulties included assessment of sucking, swallowing, and breathing. Stridor and difficulty interspersing breaths during sucking bursts were observed, although sucking mechanics were normal. Further evaluation of airway structure revealed asymmetric laryngomalacia. This structural problem, exacerbated by the breastfeeding position, impeded respiration and resulted in poor coordination of sucking, swallowing, and breathing. This case underscores the ne

Lactate Dehydrogenase B Expression in Cerebellum of Adult Ube3a m-/p+ Mice.

Angelman syndrome (AS) is a neurodevelopmental disorder caused by the loss of the maternal allele of the UBE3A gene, which encodes a protein essential for ubiquitin-mediated protein degradation. AS mouse models exhibit elevated lactate and acetate levels and increased Ldha gene expression in fibroblasts. We hypothesize that maternal Ube3a loss alters the protein levels of Ldha, Ldhb, Mct2, and Mct1, contributing to elevated brain lactate levels. Western blot analysis of the cerebellum from adult Ube3a m-/p+ AS and wild-type mice reveals significant sex- and genotype-specific differences in Ldh

Melatonin use during pregnancy and lactation: A scoping review of human studies.

OBJECTIVE: The prevalence of sleep disorders during the perinatal period is high and large health administrative database surveys have shown that the use of exogenous melatonin in pregnant populations is quite common, about 4%. Much of the concern about using melatonin during pregnancy and breastfeeding stems from animal research. Thus, the objective of this article is to provide a critical review of human studies related to exogenous melatonin use during pregnancy and breastfeeding. METHODS: The electronic databases Ovid, MEDLINE, Embase, and the Cochrane Library were searched using terms an

Maternal fructose intake predisposes rat offspring to metabolic disorders via abnormal hepatic programming.

Given that fructose consumption has increased by more than 10-fold in recent decades, it is possible that excess maternal fructose consumption causes harmful effects in the next generation. This study attempted to elucidate the mechanism of the harmful effects of excessive maternal fructose intake from the perspective of offspring liver function. Female rats during gestation and lactation were fed water containing fructose, and their offspring were fed normal water. We attempted to elucidate the mechanism of fructose-induced transgenerational toxicity by conducting a longitudinal study focusin

Lactoferrin Deficiency During Lactation Causes Adult Obesity-Related Metabolic Disease Through Persistent Adipose Dysfunction Driven by Impaired Adipocyte Development.

Obesity is associated with metabolic disorders due to unhealthy white adipose tissue (WAT) failing to sustain energy homeostasis, highlighting the importance of adipose development. The lactation period is critical for epididymal WAT (eWAT) development and metabolic programming. However, the role of lactoferrin (LF) in the early development of adipose remains unclear. Using a mouse model of lactational LF deficiency and single-nucleus RNA sequencing, we assessed the long-term impact of LF deficiency on eWAT plasticity and metabolic homeostasis at weaning, adulthood, and under a high-fat diet (
