Hypophosphatasia is rare enzymopathy that normally presents within the first few years of life and often has profound effects upon the periodontium. It is a heritable disorder characterised by defective mineralisation of the skeletal and dental structures of the body and a deficiency in the liver/bone/kidney (L/B/K) isoenzyme of alkaline phosphatase (ALP). There has been a tremendous advance in our knowledge of this condition over the last decade due to the advent of highly specific DNA probes and novel microanalytical techniques. This paper aims to review current literature about hypophosphat

This study aims to investigate the action mechanism by which Stellera chamaejasme extract(SCLE) overcomes multidrug resistance(MDR) in breast cancer through the mitogen-activated protein kinase kinase(MEK)/extracellular signal-regulated kinase(ERK) signaling pathway. Sensitive cell line MCF-7 of human breast cancer and its multidrug resistance counterpart MCF-7/adriamycin(ADR) were employed as models. The effect of SCLE on cell proliferation was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay. The transcriptional level of P-glycoprotein(P-gp) was analyzed by

Development requires the fusion of two gametes each with a haploid complement of DNA and the necessary components (RNA, proteins, organelles) needed for early cellular processes and divisions. The C. elegans paternal-effect embryonic lethal gene, spe-11, is one of the few known sperm components required for normal development. Sperm-provided SPE-11 forms a complex with the oocyte protein OOPS-1 at fertilization that mediates egg activation events. In this manuscript, we identified SPSP-1, a sperm protein that is associated with SPE-11. We found that SPSP-1 co-localizes with SPE-11 during sperm

Pancreatic cystic lesions have been increasingly detected with the widespread use of cross-sectional imaging. As these lesions are heterogeneous, differentiating benign from (potentially) malignant cystic lesions is important as well as challenging. Endoscopic ultrasound assessment, combined with cyst fluid analysis, forms the cornerstone of the diagnostic algorithm. Biochemical markers such as carcinoembryonic antigen (CEA), amylase, and glucose have been shown to distinguish mucinous from non-mucinous cystic lesions. Each parameter has its own advantages and its own limitations. Novel molecu

Aspergillosis is a group of different invasive and non-invasive diseases affecting the lung and other organs, caused by species of the genus Aspergillus. Additionally, complications associated with treatment and the increasing emergence of antifungal-resistant strains of Aspergillus spp. are high-impact problems. For these reasons, a combined approach of diagnostic tests is necessary to reach an accurate and timely diagnosis. This review aims to describe some biomarkers and their usefulness for the diagnosis of aspergillosis. Among the findings obtained in different studies, the nature, analyt

The presence of amyloid-beta (1-42) in CSF and plasma has been implicated as a pathological hallmark of Alzheimer's disease. Here, we demonstrate a high-sensitivity method of detection and quantification for total Aβ42 utilizing two-antibody functionalized microparticles interfaces. Our method of biomarker detection utilizes two types of microbeads: (1) magnetic particles functionalized with monoclonal antibodies and (2) polystyrene particles dual functionalized with monoclonal antibodies and a unique DNA "tag". The bead complex that forms in the presence of the target analyte is detected usin

Recent discoveries have indicated that biofilm communities may play a role in natural drug tolerance of Mycobacterium tuberculosis. A transposon-based mutation library of a closely related species, Mycobacterium marinum, was used to identify clones in which the relative amount of extracellular DNA (eDNA), an important component of the extracellular matrix of biofilms, is altered. The disruption of a putative glycosyl transferase gene QDR78 11175, epsH, caused a substantial increase of the eDNA content of biofilms, and increased the growth rate and the biomass/cell in biofilm-forming conditions

Esophageal squamous cell carcinoma (ESCC) is one of the most common forms of malignancy worldwide. However, there is currently a lack of effective chemotherapeutic drugs for ESCC. Ivermectin is a broad-spectrum antiparasitic drug with notable antitumor activity. However, the cellular and molecular mechanisms by which ivermectin inhibits cancer growth remain unclear. In this study, we elucidate the role of ivermectin in ESCC suppression by activating the endoplasmic reticulum (ER) stress and autophagy pathways. In transcriptome analyses, we find that activating transcription factor 4 (ATF4) and

Iron is a crucial element required to sustain multiple biological processes, including oxygen transport, DNA synthesis, and electron transport. In living cells, iron exists as either ferrous iron (Fe2+) or ferric iron (Fe3+), and its redox forms are regulated by the labile iron pool. Both iron deficiency and excess can lead to a range of pathological conditions, such as anemia, cancer, neurodegenerative disorders, and ischemia and reperfusion injury. Iron overload can cause oxidative damage and even cell death, especially via ferroptosis. Impaired ferroptosis pathways are implicated in the pat

Sepsis is a life-threatening organ dysfunction caused by infection that lead to dysregulation of the host response. Sepsis and septic shock with a high mortality threaten human health at present, which are important medical and health problems. Early diagnosis and treatment decision-making for sepsis and septic shock still need to be improved. Exosomes are extracellular vesicles with a diameter of 30-150 nm formed by the fusion of multi-vesicle bodies and cell membranes. Exosomes can effectively transport a variety of bioactive substances such as proteins, lipids, RNA, DNA, and participate in

Mutations in a broad variety of genes can provoke the severe childhood disorder trichothiodystrophy (TTD) that is classified as a DNA repair disease or a transcription syndrome of RNA polymerase II. In an attempt to identify the common underlying pathomechanism of TTD we performed a knockout/knockdown of the two unrelated TTD factors TTDN1 and RNF113A and investigated the consequences on ribosomal biogenesis and performance. Interestingly, interference with these TTD factors created a nearly uniform impact on RNA polymerase I transcription with downregulation of UBF, disturbed rRNA processing

Karyotyping remains an invaluable tool to researchers exploring the cause and consequence of genomic instability in biologic systems. It allows investigators to survey the entire chromosome complement in individual cells and in a single experiment, visualize, and measure different forms or features of instability such as aneuploidy, ongoing chromosomal instability, DNA damage/mis-repair, telomere erosion, chromosome mis-segregation, or defects in cell cycle progression. This chapter describes the combined use of conventional (DAPI-banding) and spectral karyotyping (SKY) to characterize genomic

Hemoglobin (Hb) Hammersmith, formed by serine substitution for phenylalanine at residue 42 in the beta-globin chain, is a very rare variant of unstable hemoglobin with low oxygen affinity. For patients with hemoglobinopathies, it is well-established that hematopoietic stem cell transplantation provides a complete cure, but the literature on its role for those with Hb Hammersmith is limited. A seven-month-old girl who was examined for anemia and splenomegaly was followed up for congenital hemolytic anemia. The patient with visible cyanosis of the lips and whose p50 was low in blood gas was diag

Developing effective vaccines against viral infections have significant impacts on development, prosperity and well-being of human populations. Thus, successful vaccines such as smallpox and polio vaccines, have promoted global societal well-being. In contrast, ineffective vaccines may fuel arguments that retard scientific progress. We aim to stimulate a multilevel discussion on how to develop effective vaccines against recent and future pandemics by focusing on acquired immunodeficiency syndrome (AIDS), coronavirus disease (COVID) and other viral infections. We appeal to harnessing recent ach

BACKGROUND: A newly discovered zoonotic infection carried by ixodid ticks, Anaplasma capra, affects a wide variety of hosts, including numerous mammals. A. capra most likely infects erythrocytes or endothelial cells in mammals. This study aimed to investigate the A. capra pathogen in goats in Türkiye's Van province.
METHODS: A total of 200 goat blood samples were examined. Goat samples were subjected to partial amplification of the gltA gene fragment using a nested polymerase chain reaction.
RESULTS: A. capra DNA was detected in 0.5% of goat blood samples. Phylogenetic analysis of a partial

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by motor neuron loss. ALS is now associated with mutations in numerous genes, many of which cause disease in part through toxic gain-of-function mechanisms. Antisense oligonucleotides (ASOs) are small sequences of DNA that can reduce expression of a target gene at the post-transcriptional level, making them attractive for neutralizing mutant or toxic gene products. Advancements in the medicinal chemistries of ASOs have improved their pharmacodynamic profile to allow safe and effective delivery to the central

Leishmaniasis is a neglected tropical disease with two main clinical forms: cutaneous and visceral leishmaniasis. Diagnosis of leishmaniasis is still a challenge, concerning the detection and correct identification of the species of the parasite, mainly in endemic areas where the absence of appropriate resources is still a problem. Most accessible methods for diagnosis, particularly in these areas, do not include the identification of each one of more than 20 species responsible for the disease. Here, we summarize the main methods used for the detection and identification of leishmaniasis that

Mitochondrial stress releases mitochondrial DNA (mtDNA) into the cytosol, thereby triggering the type Ι interferon (IFN) response. Mitochondrial outer membrane permeabilization, which is required for mtDNA release, has been extensively studied in apoptotic cells, but little is known about its role in live cells. We found that oxidatively stressed mitochondria release short mtDNA fragments via pores formed by the voltage-dependent anion channel (VDAC) oligomers in the mitochondrial outer membrane. Furthermore, the positively charged residues in the N-terminal domain of VDAC1 interact with mtDNA
