Immunoguided cutaneous regeneration: a theoretical framework linking extracellular matrix, immunity, and skin allografts.

Adult human skin healing is predominantly characterized by fibrosis rather than true regeneration, despite the conserved regenerative principles observed in fetal wound healing, liver regeneration, and other biological systems. Historical and contemporary observations in skin allotransplantation, including studies describing the differential behavior of the epidermis and dermis and recent experience with Cryopreserved Total Skin Allografts (CTSA), reveal a recurring biological pattern in which the epidermis is progressively eliminated while the dermal extracellular matrix remains relatively pr

Central nervous system and cancer: mechanistic insights and therapeutic opportunities CNS and cancer: a review.

Cancer is increasingly recognized as a systemic disease whose biology is shaped by reciprocal interactions with its microenvironment, including the central and peripheral nervous systems. Recent evidence demonstrates that neural inputs can directly promote tumour growth through synaptic, paracrine, and neuroendocrine mechanisms, and that these inputs intersect with canonical drug-resistance pathways, creating new opportunities and challenges for precision oncology. Primary brain tumours such as gliomas form bona fide functional synapses with neurons, hijacking both excitatory (AMPA/NMDA) and i

Histological Transformation and Progression in Follicular Lymphoma: A Clonal Evolution Study.

BACKGROUND: Follicular lymphoma (FL) is an indolent, yet incurable B cell malignancy. A subset of patients experience an increased mortality rate driven by two distinct clinical end points: histological transformation and early progression after immunochemotherapy. The nature of tumor clonal dynamics leading to these clinical end points is poorly understood, and previously determined genetic alterations do not explain the majority of transformed cases or accurately predict early progressive disease. We contend that detailed knowledge of the expansion patterns of specific cell populations plus

An Immediate Innate Immune Response Occurred In the Early Stage of E. granulosus Eggs Infection in Sheep: Evidence from Microarray Analysis.

BACKGROUND: Cystic Echinococcosis(CE), caused by infection with the larval stage of the cestode Echinococcus granulosus (E. granulosus), is a chronic parasitic zoonosis, with highly susceptible infection in sheep. However, the comprehensive molecular mechanisms that underlie the process of E. granulosus infection in the early stage remain largely unknown. The objective of this present study was to gain a cluster of genes expression profiles in the intestine tissue of sheep infected with CE. METHODS: Nine healthy sheep were divided into infection group and healthy controls, with six infected p

Philadelphia chromosome positive adult acute lymphoblastic leukemia: characteristics, prognostic factors and treatment outcome.

Philadelphia chromosome positive (Ph+) acute lymphoblastic leukemia (ALL) is an aggressive form of acute leukemia that represents about one third of all adult ALL. Between 1984 and 1996, forty-three cases of Ph+ ALL (22 males and 21 females) were diagnosed in our institution by successful cytogenetic studies and/or molecular biology. Median age was 42 years (range, 20-71 years) with 28 patients aged below 50 years. Median leukocyte count was 39.7 x 10(9)/l on admission. Tumoral syndrome was seen only in 21 patients (49%) of which 4 cases presented with central nervous system (CNS) involvement.

In Vitro Anticancer Activity of the Light Stable Zinc Isotope (64Zn) Compounds.

BACKGROUND/AIM: Today, stable isotopes of zinc are actively used for diagnostic purposes in oncology. However, there is extremely limited data on the attempts to apply stable zinc isotopes in cancer therapy or about the molecular mechanisms of their effects on the biology of tumor cells. Therefore, in this in vitro research, we evaluated the cytotoxic activity of stable zinc isotope (64Zn) enriched compounds against malignant cells and determined the mechanisms of their action. MATERIALS AND METHODS: Malignant and non-malignant cells of different histogenesis were used as objects of the study

Nanobodies as Versatile Next-Generation Biologics.

Nanobodies, also known as single-domain antibodies or variable domains of heavy-chain-only antibodies (VHHs), are the antigen-binding domains derived from heavy-chain-only antibodies that naturally occur in camelids. Since their discovery in the early 1990s, Nanobodies have attracted significant attention in the fields of biotechnology, structural biology, diagnostics, and therapeutics. Their unique structural and biochemical properties distinguish them from conventional antibodies and offer several practical advantages as versatile modular units in next-generation biologics. We discuss the mo

C3 in Pomacea canaliculata: a conserved effector with developmental and immune roles.

C3 is the central effector of the complement system and one of the most conserved molecules of metazoan innate immunity. Despite its relevance, little is known about complement components in Lophotrochozoa. Here, we characterise the C3 ortholog of the freshwater gastropod Pomacea canaliculata (PcC3) using long-read transcriptomics, phylogenetics, expression profiling and immunohistochemistry. PcC3 displays a fully conserved domain architecture and clusters phylogenetically in agreement with metazoan relationships. Following LPS challenge, PcC3 is up-regulated in haemocytes and unexpectedly in

Exosome-Derived Circular RNAs in Colorectal Cancer: Emerging Roles in Tumorigenesis, Diagnosis, and Therapy.

Circular RNAs (circRNAs) are a class of non-coding RNAs characterized by their covalently closed loop structures, which confer remarkable stability. Recent studies have highlighted the role of exosomal circRNAs, which are encapsulated within extracellular vesicles known as exosomes, in intercellular communication and tumor progression. In colorectal cancer (CRC), exosomal circRNAs have been implicated in various oncogenic processes, including cell proliferation, migration, invasion, epithelial-mesenchymal transition (EMT), and drug resistance. These molecules exhibit dysregulated expression pa

Extracellular Vesicles in Ophthalmology: From Natural Nanocarriers to Engineered Therapeutics.

Extracellular vesicles (EVs) are increasingly recognized as programmable bioactive carriers in non-viral gene delivery and adaptable bioengineering platforms. Beyond their roles as natural nanocarriers in intercellular communication, EVs can promote ocular surface repair and retinal neuroprotection with potential for low immunogenicity and high biocompatibility. Bioengineering now enables cargo encapsulation, surface targeting, and integration of EVs with biomaterial platforms to enhance tissue penetration, retention, and precision delivery. A xeno-free human iPSC-derived prostate organoid platform for multilineage differentiation and genetic manipulation.

Current prostate organoid models rely on tissue-derived material or animal components and lack epithelial and stromal complexity. We defined a xeno-free system to generate human prostate organoids from induced pluripotent stem cells with consistent multilineage differentiation. Organoids formed as free-floating 3D aggregates self-organized into the epithelial and stromal domains with basal, luminal, neuroendocrine, fibroblast, and smooth muscle markers. In an alternative modular co-culture system, engineered epithelial progenitors aggregated with wild-type mesenchymal progenitors, enabling com

Renal Dysfunction Complicating the Management of Hematologic Diseases.

Renal dysfunction is common in patients with hematologic diseases and frequently emerges at diagnosis or during treatment. Acute kidney injury, chronic kidney disease, and glomerular involvement may arise from disease biology, antineoplastic therapy, infection, transplantation-related injury, or cumulative supportive exposures. In clinical practice, impaired kidney function rarely represents an isolated comorbidity; rather, it modifies therapeutic feasibility, dose intensity, supportive management, and long-term outcomes. Renal impairment affects delivery of conventional chemotherapy, targeted

PtoHsfB1 regulates growth and salt response by affecting ABA biosynthesis in Populus tomentosa.

Poplar is an important commercial timber species and a model organism for forest molecular biology. Here, we identified PtoHsfB1, a subgroup B heat shock transcription factor from Populus tomentosa that is homologous to AtHsfB1 and PtrHsfB1, and contains the conserved repression motif '-LFGV-'. To elucidate its function, transgenic poplars overexpressing PtoHsfB1 were generated. Overexpression significantly enhanced growth rate and biomass accumulation. Histological analyses revealed increased cambial cell layers and enlarged phloem caps, indicating a positive role in cambial activity and phlo

Advances in m6A Modification in Pregnancy-Related Disorders and Female Reproductive Endocrine Diseases.

N6-methyladenosine (m6A) is the most abundant internal RNA modification and has emerged as an important regulator of reproductive biology. Dysregulation of m6A regulators has been implicated in pregnancy-related disorders, including recurrent implantation failure, miscarriage, preeclampsia, and gestational diabetes, as well as female reproductive endocrine diseases such as polycystic ovary syndrome, premature ovarian insufficiency, and endometriosis. In this review, we summarize current evidence on the roles of m6A writers, erasers, and readers in these disorders, with particular attention to

Aging reprograms microglia toward an inflammasome-linked response to traumatic brain injury.

Traumatic brain injury (TBI) disproportionately kills and disables older adults, yet the biology driving this vulnerability remains unresolved. In this issue of the JCI, Lu et al. combined single-cell transcriptomics, metabolomics, and chromatin profiling in mice, validated in human TBI tissue, to define an age-dependent microglial dichotomy. They report that an NLRP3+/IL-1β-linked state dominates the aged brain, while a Lysozyme+/Lyz2+ state predominates in the young. Microglia-targeted perturbation of NLRP3 and ELF1 each shifted the balance and improved survival in mouse models of TBI, and t

Personalized Zebrafish Models for Fusion-Positive Pediatric Sarcomas.

Clinical sequencing efforts have revolutionized our approaches to categorizing pediatric cancers in real time. This has dramatically improved our ability to profile pediatric tumors, identify actionable vulnerabilities, and influence clinical care. However, often many of these genetic drivers are indicators of poor prognosis with no available targeted therapies. This is especially true for fusion-positive pediatric sarcomas, which are driven by defining oncogenic fusions that are often intractable therapeutically. Additionally, many of these fusions have no cell, animal, or patient-derived xen

Hodgkin Lymphoma: Management of Patients Who Fail Primary Therapy.

Management of relapsed/refractory (R/R) classic Hodgkin lymphoma (cHL) has evolved substantially with the integration of novel targeted and immune-based therapies. Agents such as brentuximab vedotin (BV) and immune checkpoint inhibitors (CPIs) have demonstrated high response rates in multiple settings and are increasingly used as therapeutic strategies for frontline, salvage, and multiply relapsed cHL. Consequently, the traditional roles of cytotoxic chemotherapy and stem cell transplantation are being redefined, with greater emphasis on disease biology and individualized risk-benefit assessme

Systems Biology Identifies TARS2 as a Cardiomyocyte Regulator of Mitochondrial Oxidative Stress in Dilated Cardiomyopathy.

Dilated cardiomyopathy (DCM), a leading cause of heart failure, is characterized by progressive cardiomyocyte (CM) loss and mitochondrial dysfunction; yet, the molecular drivers of mitochondrial oxidative stress (MitOS) remain unclear. By integrating bulk, single-cell, and spatial transcriptomics with machine learning, we identified threonyl-tRNA synthetase 2 (TARS2) as a CM-enriched regulator of MitOS. TARS2 was consistently up-regulated in human DCM hearts and associated with apoptotic signaling and enhanced macrophage crosstalk. Functional studies demonstrated that TARS2 overexpression disr
