Effects of exogenous fibrolytic and amylolytic enzymes on ruminal fermentation and performance of mid-lactation dairy cows.

Lactation diets are composed mostly of carbohydrates that are not fully fermented by rumen microbes. The aim of this study was to evaluate exogenous fibrolytic (Fibrozyme, Alltech Inc., Nicholasville, KY) and amylolytic (Amaize, Alltech Inc.) enzymes on nutrient intake, sorting index, total-tract apparent digestibility, ruminal fermentation, nitrogen utilization, milk yield, and composition of dairy cows in mid-lactation. Thirty-two multiparous Holstein cows (181 ± 35 d in milk, 571 ± 72.7 kg of body weight, and 29.6 ± 5.24 kg/d of milk yield at the start of experiment) were blocked according

Efficacy of Mirtazapine in Patients With Functional Dyspepsia and Weight Loss.

BACKGROUND & AIMS: A subset of patients with functional dyspepsia (FD) present with early satiation and weight loss, for which there are no established therapeutic options. We investigated the efficacy of mirtazapine (an antidepressant and antagonist of the histamine receptor H1, the α2 adrenergic receptor, and the serotonin receptors 5-HT2C and 5-HT-3) in patients with FD and weight loss. METHODS: We conducted a randomized, placebo-controlled pilot trial that studied 34 patients with FD (29 women; mean age, 35.9 ± 2.3 years) with weight loss >10% of original body weight (mean loss, 12.4 ± 2.

Impact of an early-life intervention on the nutrition behaviors of 2-y-old children: a randomized controlled trial.

BACKGROUND: Despite an extensive well-child health service, 30% of New Zealand's 2- to 4-y old children are overweight or obese. This suggests that additional intervention is necessary to establish healthy nutrition behaviors. OBJECTIVE: The aim of this study was to assess the effect of intervention from 0 to 18 mo of age on food and nutrient intake, eating behaviors, and parental feeding practices in 18- to 24-mo-old children. DESIGN: In total, 802 families with healthy infants were randomly allocated to 1 of 4 groups: Usual Care (UC); Food, Activity, and Breastfeeding (FAB); Sleep; or FAB

Consuming the daily recommended amounts of dairy products would reduce the prevalence of inadequate micronutrient intakes in the United States: diet modeling study based on NHANES 2007-2010.

BACKGROUND: A large portion of Americans are not meeting the Dietary Reference Intakes (DRI) for several essential vitamins and minerals due to poor dietary choices. Dairy products are a key source of many of the nutrients that are under consumed, but children and adults do not consume the recommended amounts from this food group. This study modeled the impact of meeting daily recommended amounts of dairy products on population-based nutrient intakes. METHODS: Two-day 24-h dietary recalls collected from participants ≥ 2 years (n = 8944) from the 2007-2010 What We Eat in America, National Heal…

Flow cytometric investigation of filamentation, membrane patency, and membrane potential in Escherichia coli following ciprofloxacin exposure.

Ninety-eight percent of the cells in a population of Escherichia coli in log-phase growth lost colony-forming ability after being exposed for 3 h to the quinolone antibiotic ciprofloxacin at four times the MIC in nutrient broth, a concentration easily reached in vivo. Flow cytometric analysis, however, demonstrated that only 68% of this bacterial population had lost membrane potential, as judged by the membrane potential-sensitive dye bis-(1,3-dibutylbarbituric acid) trimethine oxonol [DiBAC(4)(3)], and only 30% could no longer exclude the nucleic acid-binding dye propidium iodide (PI), reflec

Yeast β-glucan supplementation supports immunometabolic anti-tumor responses and reverses obesity-induced dysfunction via trained hematopoiesis.

Obesity is associated with profound immune dysregulation, driving chronic inflammation while compromising host defense against tumors. While … nction sustained after weight loss. Our work reveals that yeast β-glucans act as functional "immuno-nutrients," which remodel innate immunity and identifies the mucosal/bone-marrow axis as a target for dietary manipulations to restore immune resilience without impacting metabolism.

Cell membranes sustain phospholipid imbalance via cholesterol asymmetry.

Membranes are molecular interfaces that compartmentalize cells to control the flow of nutrients and information. These functions are facilitated by diverse collections of lipids, nearly all of which are distributed asymmetrically between the two bilayer leaflets. Most models of biomembrane structure and function include the implicit assumption that these leaflets have similar abundances of phospholipids. Here, we show that this assumption is generally invalid and investigate the consequences of lipid abundance imbalances in mammalian plasma membranes (PMs). Using lipidomics, we report that cyt…

How leaky is the gut in Parkinson's disease?

The intestinal epithelial barrier (IEB) plays a critical role in health and disease by regulating the absorption of nutrients, electrolytes and water while preventing gut translocation of pathogens. A compromised intestinal barrier has been reported in Parkinson's disease (PD) further reinforcing the assumption that PD is a gut-brain axis disorder and suggesting that gut-derived factors may participate in disease development and/or progression. However, the diversity of methodology between existing studies on gut permeability in PD, especially regarding the methods used for the evaluation of t…

New Trends in Brain Shuttle Peptides.

The pharmacological treatment of central nervous system diseases faces significant challenges due to the presence of the blood-brain barrier (BBB). This barrier naturally protects the brain and prevents therapeutics from reaching their targets efficiently. However, the BBB allows the passage of nutrients and other molecules that guarantee brain homeostasis through selective transport mechanisms present at the BBB. These mechanisms provide an opportunity for delivering therapeutic agents into the central nervous system using brain shuttles. Here we review the progress of brain shuttle peptide d…

Robust Saccharomyces cerevisiae by rational metabolic engineering for effective ethanol production from undetoxified steam-exploded corn stover hydrolysate.

Lignocellulosic bioethanol production by S.cerevisiae is severely hampered by xylose assimilation and inhibitors. Aiming to solve these barriers, the xylose isomerase pathway was heterologously introduced into parental strain, followed by conducting the adaptive laboratory evolution. Meanwhile, the reduced glutathione and NADPH synthesis systems to reduce excess intracellular reactive oxygen species (ROS) were further enhanced. Results indicated the bioethanol production from undetoxified steam-exploded corn stover hydrolysate (SECSH) without any nutrients supplementation was improved using th…

TMAO and the gut microbiome: implications for the CVD-CKD-IBD axis.

Trimethylamine N-oxide (TMAO) is a metabolite produced by the gut microbiome from dietary nutrients such as choline and carnitine. Recent research has found that TMAO is strongly associated with cardiovascular disease (CVD), chronic kidney disease (CKD) and, more recently, inflammatory bowel disease (IBD). Although TMAO is linked to conditions characterized by inflammation, oxidative stress, fibrosis and gut microbiome imbalances, its exact role in disease development remains unclear. This review examines TMAO's potential role as a key link in the IBD-CKD-CVD disease spectrum. Highlighting the…

Determining the expression of vitamin D receptor, regulator of iron metabolism hepcidin and cathelicidin antimicrobial peptide genes for development of future diagnostic and therapeutic options in patients with inflammatory bowel disease.

Patients with inflammatory bowel diseases (IBD) frequently experience malnutrition and develop nutrients deficiencies such as vitamin D or iron. Collectively, 39 patients were recruited to the study - 32 with IBD and 7 patients constituting the control group (CG). Quantitative real-time polymerase chain reaction was used to assess the expression of vitamin D receptor (VDR), hepcidin (HAMP) and cathelicidin antimicrobial peptide (CAMP) in colon mucosa. The mean expression of VDR and CAMP was higher in patients with ulcerative colitis than other groups (VDR vs. CG, p<0.05). However, the mean HAM…
