The bioprospecting potential of insect venoms as antibiotics: a mini review.

The global rise of antimicrobial resistance has intensified the search for new antibiotic candidates from unconventional biological sources. Insect venoms, although underexplored compared to other venomous taxa, harbor a chemically diverse array of antimicrobial peptides (AMPs) with significant therapeutic promises. This mini review synthesizes evidence from 15 original studies published over the past 15 years that examined the antimicrobial potential of insect venom components. Most investigations have focused on Hymenoptera-wasps, bees, and ants-where peptides such as mastoparans, polydim-I,

Four-electron reduction of benzene by a samarium(II)-alkyl without the addition of external reducing agents.

Benzene reduction by molecular complexes remains an important synthetic challenge, requiring harsh reaction conditions involving group I metals. Reductions of benzene, to date, typically result in a loss of aromaticity, although the benzene tetra-anion, a 10π-electron system, has been calculated to be stable and aromatic. Due to the lack of sufficiently potent reductants, four-electron reduction of benzene usually requires the use of group I metals. Here we demonstrate the four-electron reduction of benzene and some of its derivatives using a samarium(II) alkyl reagent, with no requirement for

Trinuclear Copper(I) and Silver(I) Complexes of a Pentafluorosulfanyl-Decorated Pyrazolate.

This study reports the synthesis and characterization of the first fluorinated coinage metal pyrazolates with SF5 groups, specifically {[3-(SF5)Pz]M}3 (M = Cu, Ag), and compares them to trifluoromethylated analogs. X-ray analysis reveals different pyrazolyl ligand orientations and metallophilic interactions, as well as supramolecular structures influenced by fluorinated substituents and metals. The {[3-(SF5)Pz]M}3 complexes form face-to-face sandwich adducts with benzene. Photophysical investigations show that copper complexes exhibit long-lived phosphorescence, while silver analogs display fa

Predictive modeling of copper iodide properties using graph-theoretical descriptors.

Metal halides, inorganic compounds formed through the combination of metals and halogens, play a significant role in diverse applications. Copper iodide (CuI, Marshite) stands out as a particularly versatile metal halide with important uses in organic synthesis, semiconductor technology, catalytic processes, and atmospheric modification through cloud seeding. This study employs graph-theoretical approaches to analyze the structural properties of CuI, focusing on degree-based topological indices such as the Zagreb indices, Randić index, harmonic index, Sombor index, and atom-bond connectivity i

Cocrystal Network-Based Recommendation: A Multi-Objective Random Walk-Based Network Model for Virtual Screening of Drug-Drug Cocrystals.

Drug-drug cocrystals (DDCs) are composed solely of active pharmaceutical ingredients, offering traditional cocrystal benefits and potential intercomponent synergistic therapeutic effects. However, systematic, efficient computational methods for DDC screening are lacking. To address this challenge, here we introduced CCNBR, a multiobjective random walk network model that combines cocrystal network topology with molecular structural features. By leveraging a third-order path-based weighted random walk algorithm, CCNBR effectively captures key supramolecular interactions that drive cocrystal form

Biological Evaluation of Molecular Spherical Nucleic Acids: Targeting Tumors via a Hybridization-Based Folate Decoration.

Folate receptors (FRs), membrane-bound proteins that bind specifically to folate with high affinity, are overexpressed by various cancer types and are therefore used as targets for delivery of therapeutic agents. Molecular spherical nucleic acids (MSNAs) are dendritic formulations of oligonucleotides (ONs) that may have advantages over linear parent ONs with respect to delivery properties. Here, we assembled folate-decorated MSNAs, site-specifically radiolabeled them, and then biologically evaluated their effects in mice bearing HCC1954 breast cancer xenograft tumors. The biodistribution of in

The multifunctionality of the brainstem breathing control circuit.

Subconscious breathing is generated by a network of brainstem nodes with varying purposes, like pacing breathing or patterning a certain breath phase. Decades of anatomy, pharmacology, and physiology studies have identified and characterized the system's fundamental properties that produce robust breathing, and we now have well-conceived computational models of breathing that are based on the detailed descriptions of neuronal connectivity, biophysical properties, and functions in breathing. In total, we have a considerable understanding of the brainstem breathing control circuit. But, in the l

SpaOmicsVAE: A deep learning framework for integrative analysis of spatial multi-omics data.

Spatial multi-omics technologies have revolutionized our understanding of biological systems by enabling simultaneous measurement of multiple molecular features while preserving spatial information. However, the effective integration and analysis of these complex datasets remain challenging. Here we introduce SpaOmicsVAE, a novel computational framework that combines variational autoencoder architecture with a dual graph neural network to address these challenges. The framework uniquely integrates spatial and feature information through an attention-based mechanism, effectively handling data s

Mapping the Role of Monomer Conformation in the Amyloid Formation of α-Synuclein Splice Variants.

Amyloid formation of the protein α-synuclein (αSyn) is a hallmark of pathogenesis in Parkinson's disease, multiple system atrophy, and dementia with Lewy bodies. Research has predominantly focused on the 140-amino acid αSyn sequence, yet the SNCA gene can be alternatively spliced to generate several different isoforms, including αSynΔ3, αSynΔ5, and αSynΔ3Δ5. Here, we have used experimental and computational approaches to characterize these splice variants, in addition to the full-length αSyn, in terms of their monomer conformation and amyloid propensity as a function of changes in ionic streng

Primary Pulmonary Leiomyosarcoma: A Case of Prolonged Survival With Radiotherapy and Pazopanib in a 90-Year-Old Female.

Primary pulmonary leiomyosarcoma is an exceedingly rare malignancy, comprising less than 0.5% of all lung cancers. Due to its rarity, no standardised treatment guidelines exist. Prognosis is especially poor in cases which are deemed unresectable. We describe the case of a 90-year-old woman who presented with progressive dyspnea and cough over 1 month. Chest computed tomography revealed a large right hilar mass extending into the right upper and middle lobes, abutting the mediastinum and pericardium. Histopathological analysis confirmed primary pulmonary leiomyosarcoma. Given her advanced age a

Delayed Diagnosis of Imported Cystic Echinococcosis and Successful Treatment With Percutaneous Drainage and Albendazole in Korea: A Case Report.

Echinococcosis, caused by the tapeworm Echinococcus, is rare in Korea and is primarily imported from endemic areas. We report a case of a 37-year-old Korean man with multiple large hepatic cysts, initially diagnosed as simple cysts at a local clinic in 2018. The patient had lived in Oman, an endemic area, for several months in 2016. Upon referral to a tertiary hospital in 2023, due to progressive cyst enlargement, liver magnetic resonance imaging revealed three large cysts with a water lily sign. Serum IgG against Echinococcus was positive by enzyme-linked immunosorbent assay. After diagnosis

Curcumin ameliorates benzalkonium chloride-induced dry eye disease in mice.

Dry eye disease (DED) is exacerbated by benzalkonium chloride (BAC), a preservative in ophthalmic formulations, which induces ocular damage via inflammation, apoptosis, and oxidative stress. Using network toxicology, molecular docking, dynamics simulations, and murine models, we elucidated BAC's mechanisms and evaluated curcumin (CUR) as a therapeutic agent. Network analysis identified 492 shared BAC-DED targets, functionally enriched in inflammation, leukocyte migration, and cytokine production. TNF/IL-17 signaling and core mediators (IL6, TNFα, CASP3) were prioritized. CUR exhibited strong b

Molecular mechanism of Arp2/3 complex activation by nucleation-promoting factors and an actin monomer.

Arp (actin-related protein) 2/3 complex nucleates actin filament branches on the sides of preexisting actin filaments during cell and organelle movements. We used computer simulations of mammalian Arp2/3 complex to address fundamental questions about the mechanism. Metadynamics and umbrella free energy sampling simulations of the pathway revealed that a clash between the D-loop of Arp2 and Arp3 produces an energy barrier of 20 ± 6 kcal/mol between the inactive splayed and active short-pitch conformations of Arp2/3 complex. Atomistic molecular dynamics simulations showed that binding the CA mot

Identification of novel genes regulating the development of the palate.

BACKGROUND: The International Mouse Phenotyping Consortium (IMPC) has generated thousands of knockout mouse lines, many of which exhibit embryonic or perinatal lethality. Using micro-computed tomography (micro-CT), the IMPC has created and publicly released three-dimensional image data sets of embryos from these lethal and subviable lines. In this study, we leveraged this data set to screen homozygous null mutants for anomalies in secondary palate development. We analyzed optical sections from 2987 embryos at embryonic days E15.5 and E18.5, representing 484 homozygous mutant lines. RESULTS AN

Mechanistic insights into enterocin C targeting the undecaprenyl phosphate recycling protein BacA.

Enterococcus faecalis is an important opportunistic pathogen responsible for healthcare-associated infections. It is intrinsically resistant to various antibiotics, particularly to cephalosporins and vancomycin, creating an urgent need for alternative therapeutics. In this context, bacteriocins warrant investigations as a potential source of medically useful antibiotics. Herein, we demonstrate that Enterocin C, a class IIb two-peptide bacteriocin, specifically targets the membrane-embedded undecaprenyl phosphate recycling protein BacA from enterococci as a cell surface receptor. Using biochemi

Nanopore Identification of Polyglutamine Length via Cross-Slit Sensing.

Nanopore sensing is now reshaping analytical proteomics with its simplicity, convenience, and high sensitivity. Determining the length of polyglutamine (polyQ) is crucial for the rapid screening of Huntington's disease. In this computational study, we present a cross-nanoslit detection approach to determine the polyQ length, where the nanoslit is carved within a two-dimensional (2D) in-plane heterostructure of graphene (GRA) and hexagonal boron nitride (hBN). We designed a heterostructure with an hBN strip embedded in the graphene sheet. With such a design, polyQ peptides can spontaneously and

The Role of Multimodality Imaging in Cardiomyopathy.

PURPOSE OF REVIEW: There has been increasing use of multimodality imaging in the evaluation of cardiomyopathies. RECENT FINDINGS: Echocardiography, cardiac magnetic resonance (CMR), cardiac nuclear imaging, and cardiac computed tomography (CCT) play an important role in the diagnosis, risk stratification, and management of patients with cardiomyopathies. Echocardiography is essential in the initial assessment of suspected cardiomyopathy, but a multimodality approach can improve diagnostics and management. CMR allows for accurate measurement of volumes and function, and can easily detect uniqu

Ab initio treatment of molecular Coster-Kronig decay using complex-scaled equation-of-motion coupled-cluster theory.

Vacancies in the L1 shell of atoms and molecules can decay non-radiatively via Coster-Kronig decay whereby the vacancy is filled by an electron from the L2,3 shell while a second electron is emitted into the ionization continuum. This process is akin to Auger decay, but in contrast to Auger electrons, Coster-Kronig electrons have rather low kinetic energies of less than 50 eV. In the present work, we extend recently introduced methods for the construction of molecular Auger spectra that are based on complex-scaled equation-of-motion coupled-cluster theory to Coster-Kronig decay. We compute ion
