Circulating adipokines level and the risk of neurodegenerative diseases: a two‑sample mendelian randomization study and proteomic analysis.

BACKGROUND: Observational studies have suggested associations between circulating adipokines and neurodegenerative diseases, but the causal nature of these relationships remains unclear. This study evaluated the causal effects of adipokines on neurodegenerative diseases using Mendelian randomization (MR) and validated key findings through proteomic analysis. METHODS: Two-sample MR was performed using genome-wide association study (GWAS) summary statistics for adiponectin (N = 39,883), leptin (N = 57,232), resistin (N = 21,758), and monocyte chemoattractant protein-1 (MCP-1; N = 21,758). Detecting hidden threats: proteomic changes in urinary extracellular vesicles are associated with pediatric kidney allograft rejection.

BACKGROUND: Early detection of kidney allograft injury is essential to preserve long-term graft function. Current diagnostics, particularly biopsy, are invasive and may be inconclusive. Extracellular-vesicles (EVs) carry molecular cargo that reflects renal cellular states. Although EV proteomics has shown promise for detecting acute rejection (AR) in adults, its value in pediatrics(PKT) is not well defined. METHODS: In this pilot study, urine samples from 29 PKT recipients and 3 healthy controls were analyzed, including 7 with biopsy-confirmed AR, 9 with chronic rejection (CR), and 13 with st

Proteome-level evidence that tebuconazole, both alone and in interaction with thiacloprid, affects epigenetic events in bumblebee heads.

Tebuconazole, a widely used ergosterol biosynthesis-inhibiting fungicide, can affect nontargets, especially when combined with insecticides. We employed label-free quantitative proteomics to investigate the effects of long-term exposure to sublethal concentrations (100 μg/L) of tebuconazole, either by itself or alongside the neonicotinoid thiacloprid (100 μg/L), on the heads of Bombus terrestris workers. A Bayesian factor power analysis revealed that the experiment produced conclusive proteomic results. Tebuconazole treatment revealed eleven differentially abundant proteins, which increased el

Extracellular vesicles with unique proteomic signatures from self-organized cardiac spheroids under hydrodynamic stimulation.

Cardiac extracellular vesicles (EVs) hold promise as cell-free therapeutics for heart repair. However, robust methods to produce scalable, functional and cardiac-specific EVs at high yield remain a limiting factor in their exploitation. Here, we report an engineered platform that combines 3D cardiac microtissues with hydrodynamic stimulation to address these hurdles. We exploited differential cell mechanics, quantified via surface tension measurements, to establish a unique 3D spheroid architecture showing cardiomyocytes preferentially localizing at the spheroid periphery. A controlled-flow bi

Proteomic profiling of colorectal liver metastases reveals histopathological response-specific molecular signatures of chemotherapy efficacy.

BACKGROUND: Chemoresistance in treatment of colorectal liver metastases (CRLM) poses a major challenge in preventing disease relapse, with up to 80% of patients developing drug resistance over the course of treatment. Proteomic signatures of responsive vs. non-responsive metastases can provide insights into functional expression patterns, potentially identifying biomarkers for therapy efficacy. METHODS: A total of 33 CRLM tissue samples from 31 patients were subjected to histopathological and proteomic analysis. The patients were included in the study after undergoing preoperative treatment (n

Distinct proteomic signatures of urinary extracellular vesicles link to albuminuria and treatment responses in diabetic kidney disease.

INTRODUCTION: Urinary extracellular vesicles (uEVs) offer a noninvasive approach to gain pathophysiological insight into kidney disease and treatment effects. Our study aimed to investigate the impact of albuminuria and antialbuminuric therapies on the uEV proteome in diabetic kidney disease. METHODS: uEVs were isolated from urine samples of 20 patients with diabetes and albuminuria (age 64 years, estimated glomerular filtration rate 85 ml/min per 1.73 m2, urinary albumin creatinine ratio 130 mg/g) who participated in a crossover trial. Participants received telmisartan, empagliflozin, linagl

Clinical Deployment of Interpretable AI: Bridging Routine Clinical Tests and Proteomic Signatures for Preeclampsia Risk Stratification.

INTRODUCTION: Preeclampsia (PE) is the second-leading global cause of maternal mortality, affecting 5% of primigravidas. Owing to the substantial heterogeneity of clinical manifestations in PE, an urgent need arises to quantitatively evaluate the efficacy of existing diagnostic methods based on positive proteinuria (PRO) and to develop novel biomarkers to enhance diagnostic accuracy. METHODS: We based 1,215 pregnant women obtained from who delivery at the hospital in January 2018 and April 2022 and involved predictors of 66 routine clinical laboratory tests (RCLTs). In addition, from 362 peri

Midlife plasma proteomic profiles indicate altered amyloid and tau processing in former elite rugby players.

BACKGROUND: Contact sports, including rugby union, are associated with higher rates of neurodegenerative dementia, due to various underlying pathologies such as Alzheimer's disease (AD) and chronic traumatic encephalopathy (CTE). New ultrasensitive multiplexed immunoassays may clarify disease mechanisms after repetitive head impacts (RHI) and traumatic brain injury, potentially aiding risk-stratification, early diagnosis and dementia treatment. METHODS: Midlife participants in the ABHC cohort underwent plasma biomarker quantification (NULISA - NUcleic acid Linked Immuno-Sandwich Assay; n=124

Proteomic Mediators of Chronic Obstructive Pulmonary Disease Phenotypes and Coronary Artery Calcification Burden in Ever Smokers.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) increases cardiovascular disease risk. Coronary artery calcification (CAC) predicts cardiovascular events and mortality in COPD. We hypothesized that plasma proteins linked to pulmonary phenotypes mediate CAC burden. METHODS: Pulmonary function, emphysema, airway wall thickening, Agatston CAC scores (inverse normal transformed), and relative abundance of 1305 plasma proteins (log-transformed) were assessed in 989 Phase 1 COPDGene (Genetic Epidemiology of COPD) participants. Proteins associated with both pulmonary phenotypes (FEV1[forced

Proteomic Analysis of Human Chronic Traumatic Encephalopathy Brain Implicates Proteasome and Ribosome Dysfunction in Disease Progression.

Chronic traumatic encephalopathy (CTE) is a progressive neurodegenerative disease associated with repeated head injuries (RHI) commonly experienced by contact sport athletes, military personnel, and domestic abuse victims. Despite growing recognition of CTE, the molecular mechanisms underlying disease progression remain poorly understood. This study aims to identify proteomic alterations associated with CTE pathology and clinical features to elucidate key biological pathways involved in disease pathogenesis. Proteome-Wide Dissection of Cachexia-Inducing Evolution in a Paired Human Neuroendocrine Carcinoma Cell Line Model.

Cancer cachexia is a devastating systemic syndrome, yet tumor-intrinsic programs that enable cachexia induction remain poorly defined. Here we dissect cachexia-inducing tumor evolution using a uniquely paired model derived from a rare human duodenal neuroendocrine carcinoma, in which repeated in vivo passaging converts a non-cachexia-inducing cell line (TCC-NECT-2) into a cachexia-inducing derivative (AkuNEC). AkuNEC xenografts reproducibly induced progressive body weight loss, whereas TCC-NECT-2 xenografts did not. Using data-independent acquisition (DIA) proteomics, we quantified the proteom

Proteomic remodeling of outer membrane vesicles in antibiotic-resistant Helicobacter pylori reveals adaptive stress responses.

BACKGROUND: Antibiotic resistance in Helicobacter pylori has become a major obstacle to successful eradication therapy worldwide. Although outer membrane vesicles (OMVs) are increasingly recognized as key mediators of bacterial adaptation and host interaction, their contribution to antibiotic resistance in H. pylori remains poorly understood. METHODS: Metronidazole and levofloxacin-resistant derivatives of the H. pylori G27 strain were generated by stepwise antibiotic exposure. OMVs isolated from susceptible and resistant strains were characterized by transmission electron microscopy and dyna

Magnolia officinalis (L.) Bark Extract Counteracts Oxidative Brain Injury: A Proteomic Investigation into Neuroprotective Mechanisms.

Neurodegenerative diseases involve progressive neuronal loss associated with oxidative stress (OS) and inflammation. Given the limited efficacy of current therapies, natural compounds with multitarget neuroprotective potential are of growing interest. In this study, we investigated the neuroprotective effects of a standardized Magnolia officinalis (L.) bark extract (MOE) in rat brain cortical slices exposed to hydrogen peroxide-induced OS. MOE significantly recovered tissue viability and reduced ROS and malondialdehyde levels caused by OS while attenuating caspase-3, -8, and -9 activation, sug

Global Proteomic Analysis Reveals Inflammatory Pathway Modulation Associated with miR-146a in LPS-Stimulated Macrophages.

Inflammation is essential for host defense, but, when dysregulated, it contributes to tissue damage and chronic disease. MicroRNA-146a (miR-146a) is a well-recognized negative regulator of inflammatory signaling, primarily through suppression of the NF-κB pathway; however, its broader proteomic impact under inflammatory conditions remains incompletely defined. In this study, we overexpressed an miR-146a mimic in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages and applied quantitative mass spectrometry to characterize global protein abundance changes. Functional overexpression was sup

Inflammatory-neurodegenerative crosstalk in pediatric severe traumatic brain injury: a multi-domain plasma proteomic study.

BACKGROUND: Pediatric severe traumatic brain injury (sTBI) remains a leading cause of death and long-term disability, yet its molecular characterization is incomplete. Single-biomarker approaches fail to capture the biological heterogeneity underlying variable clinical outcomes. Comprehensive proteomic profiling across neuropathologic domains to identify clinical endotypes has not been performed in pediatric sTBI. This study addresses this gap using high-throughput plasma neuro-proteomic profiling during the acute injury phase. METHODS: We conducted an exploratory case-control study enrolling

Proteome-wide Mendelian randomization identifies APOM and TNXB as actionable mediators of steroid-sensitive nephrotic syndrome.

BACKGROUND: Discovery of autoantibodies in steroid-sensitive nephrotic syndrome (SSNS) has transformed our understanding of SSNS as an immune-mediated disease; however, mechanisms underlying autoantibody production are unknown. Current treatments for SSNS are non-targeted and cause serious adverse effects. We sought to identify circulating proteins with a causal relationship to SSNS, which likely reflect underlying immune derangements, using an unbiased two-sample Mendelian randomization (MR) and colocalization approach to inform novel drug targets for disease. METHODS: Summary-level data fro

Functional and proteomic characterization of Phytophthora nicotianae extracellular vesicles and putative cargo-sorting signals in filamentous pathogens.

Extracellular vesicles (EVs) are lipid-bound structures that transport bioactive molecules and are increasingly recognized as key mediators of plant-pathogen interactions. Although EV secretion has been reported in filamentous pathogens, their roles in plant immunity, microbial interactions and cargo-sorting mechanisms remain poorly understood. This study aimed to characterize the functional roles and composition of EVs produced by P. nicotianae, and to investigate shared EV-associated protein categories and putative cargo-sorting signals, including KFERQ-like (Lys-Phe-Glu-Arg-Gln) motifs, acr
