Topically Delivered Treg-Attracting Microparticle-Hydrogel Composites Recruit Regulatory T Cells and Limit Cardiac Injury After Severe Ischemia.

Ischemic heart disease is a highly prevalent and debilitating condition throughout the world. Following an ischemic injury, the immune respo … d can suppress disadvantageous inflammation. Here we combine our Treg-attracting MP technology with Hydrogels to Enhance or Attract Regulatory T cells (HEART). Topically applied HEART significantly reduced infarct size and improved ejection fraction after permanent-ligation MI, and in a cardiac transplant ischemia model reduced graft fibrosis while signific …

Multifunctional chito-oligosaccharide-and deferoxamine-modified GelMA-OHA hydrogel dressing with antibacterial, antioxidant, and pro-angiogenic activities for infected wound healing.

Infected wounds resist healing due to multidrug-resistant bacterial colonization and persistent inflammation, which render conventional antibiotic therapies largely ineffective. Consequently, non-antibiotic strategies that coordinate infection control with tissue regeneration are urgently required. We design a dual-crosslinked hydrogel composed of gelatin methacryloyl (GelMA) and oxidized hyaluronic acid (OHA), further functionalized with chitosan oligosaccharide (COS) and deferoxamine (DFO), yielding the multifunctional GOCD hydrogel. Dynamic Schiff base linkages confer controlled release of

PEG-grafted dopamine methacrylate-functionalized hydroxyl-terminated PDMS-based silicone hydrogel contact lenses for sustained ocular drug delivery.

Ocular drug delivery systems characterized by extended retention time and high bioavailability are urgently required to overcome the limitations of conventional eye drops. In this study, a novel drug-loaded silicone hydrogel contact lens (SHCL) was developed utilizing hydroxyl-terminated polydimethylsiloxane (PDMS-OH) as the silicone matrix, poly(ethylene glycol)-grafted methacryloyldopamine (PEG-MAD) as a functional monomer, and sodium acetyl hyaluronate (SAH) as a surface coating. PEG-MAD was synthesized via a lipase-catalyzed reaction between carboxyl-terminated PEG (PEG-COOH) and methacryl

A high-performance, self-adaptive sprayable hydrogel dressing based on zinc alginate, quaternized chitosan, and gallic acid for wound healing applications.

Conventional wound dressings often fail to conform adequately to irregular wound beds, thereby limiting their therapeutic efficacy. Therefore, the development of multifunctional dressings with self-adaptive capabilities is crucial for improving the quality of wound healing. Herein, we developed a spray-crosslinking strategy to construct a multifunctional hydrogel dressing by compounding natural polymers‑sodium alginate (Alg), quaternized chitosan (QAC), and gallic acid (GA). Through a dual-nozzle spray system, the dressing was formed via in situ and rapid crosslinking with zinc ions (Zn2+). Th

Fabrication of NIR and ROS-responsive β-cyclodextrin/hyaluronic acid-based hybrid hydrogel for the treatment of HER2-positive gastric cancer cells.

The combined therapeutic approaches by unique biomaterial have great attention for gastric cancer therapy with peritoneal metastasis. In this research report, we designed NIR-responsive Fe3O4@MIL-100(Fe)-Upconversion nanoparticles (FMUNPs) functionalized injectable β-cyclodextrin/hyaluronic acid macromolecular hydrogel-encapsulated with HER-2 targeting trastuzumab (TZB) molecules to induce ROS against gastric cancer cells. The developed FMUNPs could provide effective ROS generation ability by photo-Fenton rection with NIR irradiation for toxicity and cell death against gastric cancer cells (NC

Injectable and Self-Healing Multifunctional Nanocomposite Hydrogel with an All-in-One Strategy for Efficient Synergistic Photothermal-Sonodynamic-Chemodynamic-Chemotherapy of Pancreatic Cancer.

Traditional pancreatic cancer treatment has long been severely hindered by the dense stromal matrix barrier, which drastically impedes the deep penetration of chemotherapy drugs and thus compromises therapeutic efficacy, leading to unsatisfactory clinical outcomes. An injectable and rapid self-healing hydrogel based on β-cyclodextrin (β-CD) and hydroxypropyl chitosan (HPCS) loading multifunctional nanoparticles was designed for efficient synergistic therapy. Specifically, aldehyde-modified β-CD (ACD) and HPCS were cross-linked via a dynamic Schiff base reaction between aldehyde and amino group

An intelligent nano-engineered PLGA@IGF-1 hydrogel for programmed immune modulation and tendon regeneration via IGF-1R/AKT signaling.

Tendon repair remains a major clinical challenge, largely because current strategies often fail to simultaneously achieve early inflammatory control and subsequent matrix remodeling. To address this problem, we developed a dynamically responsive nano-hydrogel, CAPP@IGF-1, composed of 3-carboxyphenylboronic acid-modified chitosan, polyvinyl alcohol, and IGF-1-loaded PLGA microspheres, which were integrated through phenylborate ester crosslinking. The hydrogel exhibited favorable self-healing behavior together with enhanced mechanical properties, with an elastic modulus of 4.5 MPa and a toughnes

In Situ Forming ROS-Scavenging Hydrogel with a STING Inhibitor Improves Recovery After Acute Spinal Cord Injury by Modulating Inflammatory Microenvironment.

The inflammatory cascade initiated by acute spinal cord injury (SCI) is a crucial element contributing to subsequent pathological damage. Uncontrolled or excessive inflammatory responses aggravate neural tissue destruction and hinder the regenerative process. Stimulator of interferons genes (STING) is a key regulator in the innate immune signaling pathway that promotes the generation of inflammatory mediators by activating the TBK1 and IRF3 signaling pathways, thereby shaping the post-injury microenvironment. In this study, we developed a hyaluronic acid-phenylboronic acid-polyvinyl alcohol-ba

Dual-functional regulation of macrophage polarization by ROS-scavenging and chemokine-adsorbing heparin-based hydrogel microspheres for the treatment of chronic inflammatory diseases.

For chronic and progressive diseases such as osteoarthritis (OA) and diabetic wounds (DW), there are currently no effective drugs that can prevent or reverse their progression. Pathological overproduction of reactive oxygen species (ROS) and sustained inflammation are identified as key drivers of OA or DW progression. Herein, we developed a hydrogel microsphere system via cross-linking of star-shaped polyethylene glycol (star-PEG) with heparin, followed by surface functionalization with the radical scavenger Heparin @ PEG @ TEMPO (HPT) microspheres. The TEMPO-functionalized groups on the micro

Rechargeable N-Halamine Hydrogel Paint Coating Based on Upcycled Sesame Meal Protein for Preventing Cross-Contamination on Food-Contact Surfaces.

Sesame meal protein isolate (SPI) derived from sesame meal was utilized as an upcycled plant protein source to develop N-halamine structure-based antimicrobial coating system to prevent cross-contamination for fresh food produce. The coating matrix was formulated using SPI, tannic acid (TA), cellulose nanocrystal (CNC), and sodium hypochlorite (NaOCl). The N-halamine structure maintained a stable active chlorine content of approximately 5 µmol/cm2 and enabled repeated recharging to the initial level on the coated model food-contact surface, stainless steel (SS). The stability of the coated sur

CaO2 nanoparticle-loaded injectable hydrogel with sustained oxygen release and ROS-scavenging functions for accelerating frostbite wound healing at high altitude.

Frostbite is common in high-altitude regions, where low oxygen levels and the cold climate aggravate wound hypoxia and reactive oxygen species (ROS) accumulation, making frostbite lesions prone to becoming hard-to-heal wounds. Traditional hydrogel dressings remain unsatisfactory under such extreme conditions, as they lack freezing resistance, low-temperature adhesion, sustained oxygen release, and ROS-scavenging activity. Herein, we report a multifunctional hydrogel for the management of high-altitude frostbite wounds, formed from polyvinylpyrrolidone-coated CaO2 nanoparticles (CPO NPs), polyv

Biomass-Derived Conductive Hydrogel-Based Electronic-Skin Patch for Integrated Wearable Bioelectronics and Real-Time Wound-Status Monitoring and Treatment.

Conductive hydrogel-based electronic-skin (e-skin) patches represent an emerging category of smart wound dressings, facilitating innovative approaches to chronic wound repair and real-time monitoring. Despite their advantages in biocompatibility and exudate management, current hydrogel patches frequently suffer from limitations─including single functionality, inadequate real-time monitoring capabilities, and poor adaptability to diverse wound types. Herein, we present a multifunctional biomass-derived conductive hydrogel e-skin patch. It is fabricated via supramolecular assembly of carboxymeth

Synergistic effects of berberine and adipose stem cells in a silk fibroin-PEG hydrogel for diabetic wound healing via macrophage polarization modulation.

Diabetic wound repair remains a considerable clinical challenge, largely due to the limited efficacy of current therapies. Herein, we engineered a novel silk fibroin (SF)-polyethylene glycol (PEG) hydrogel (SPB) incorporated with berberine (BBR) and adipose-derived stem cells (ASCs) to synergistically promote diabetic wound healing. By optimizing the physicochemical properties of SPB, we found that a 7:3 SF:PEG ratio provided an ideal balance of pore size (25.83 ± 3.94 μm), mechanical strength (32.33 ± 4.07 kPa) and sustained BBR release. In vitro, SPB significantly enhanced ASC paracrine func

Composite Hydrogel With Ultra-High Mechanical Anisotropy and Biotissue-Like Softness by Confining Electrostatic and Entropic Repulsion Between Cofacially Aligned Nanosheets.

Owing to their biotissue-like nature, mechanically anisotropic hydrogels have attracted extensive attention. Recent studies have achieved high mechanical anisotropy by densely assembling oriented polymers and/or nanofillers into stiff networks. However, this strategy involves soft-direction hardening to afford intrinsically hard hydrogels, limiting their applications, particularly in biomedical fields. Here, we developed the first hydrogel that simultaneously achieves excellent mechanical anisotropy and a low elastic modulus in the soft direction, using magnetically oriented, negatively charge…

A mechanically tunable photo-cured hydrogel system based on methacrylated collagen-keratin inter-penetrating networks for rapid and enhanced wound healing.

Conventional hydrogel wound dressings have non-tunable mechanical properties and serve only as passive barriers, resulting in slow wound healing. Advanced hydrogel dressings that can stimulate wound contraction and tissue deformation can potentially accelerate the healing of chronic wounds characterized by long healing times and high infection risk. In this study, we developed injectable photo-cured methacrylated collagen-keratin interpenetrating network (ColMA-K@IPN) hydrogels for rapid enhanced wound healing. Fourier transform infrared spectroscopy (FTIR) and 1H nuclear magnetic resonance (N…
