FoxO1, together with Notch1, promotes microglial activation to induce pathological changes in the retinal vasculature under hypoxia.

Microglia, the resident immune cells in the retina, play important roles in the retinopathies. Although the role of the Notch signaling pathway in microglial activation and inflammation in neuroinflammatory diseases has been extensively studied, less is known about the effects of Notch signaling on retinal microglia in ischemic retinopathies. Here, we demonstrated that hypoxia triggers Notch1-FoxO1 nuclear translocation in retinal microglia, driving proinflammatory and proangiogenic phenotypes that disrupt vascular homeostasis. We first showed that hypoxia induced microglial activation and upr

Preeclampsia as predisposing factor for hypertensive retinopathy: Participation by the RAAS and angiogenic factors.

Preeclampsia (PE) is a hypertensive complication of pregnancy. Its cause is still unknown and it could be a risk factor for future ophthalmic problems. Retinal vascular bed alterations have been described as a consequence of PE, suggesting a retinopathy. Factors related to angiogenesis and vascular permeability, such as vascular endothelial growth factor (VEGF) and pigment epithelium derived factor (PEDF) or components of the renin angiotensin aldosterone system (RAAS), prorrenin/renin receptor ((P)RR) and angiotensin II type I receptor (AT1R) have been located in the retina, participating in

Monogenic causes of stroke: now and the future.

Most stroke is multifactorial with multiple polygenic risk factors each conferring small increases in risk interacting with environmental risk factors, but it can also arise from mutations in a single gene. This review covers single-gene disorders which lead to stroke as a major phenotype, with a focus on those which cause cerebral small vessel disease (SVD), an area where there has been significant recent progress with findings that may inform us about the pathogenesis of SVD more broadly. We also discuss the impact that next generation sequencing technology (NGST) is likely to have on clinic
